Glutathione ester protects against hydroxynonenal-induced loss of auditory hair cells.

OBJECTIVE: Test the ability of glutathione monoethyl ester (GSH(e)) to protect auditory hair cells against the ototoxic effects of 4-hydroxy-2,3-nonenal (HNE). STUDY DESIGN AND SETTING: Organ of Corti explants were either untreated or treated with one of a series of four concentrations of GSH(e) for one day, then exposed to HNE. Counts of FITC-phalloidin-labeled hair cells determined both HNE ototoxicity and GSH(e) otoprotection. RESULTS: HNE was toxic to hair cells at physiologically relevant levels, eg, 400 muM, and GSH(e) provided a significant level of protection against HNE ototoxicity (P < 0.05) at all levels tested, ie, 1.16 to 9.3 mM. CONCLUSION: GSH(e) protects auditory hair cells from damage and loss initiated by a naturally occurring ototoxic molecule, ie, HNE (a by-product of oxidative stress). SIGNIFICANCE: Treatment with GSH(e) may be an effective therapy to protect the cochlea against the adverse effects of traumas (eg, electrode insertion) that generate oxidative stress.     

New digital method for quantitative assessment of nasal morphology.

Our aim was to develop and validate a new method to assess objectively and quantitatively the morphology of the nostrils after nasal or nasolabial surgery. We used digital analysis using specific mathematical algorithms to assess several geometric measurements, particularly of facial asymmetry, expressed in adimensional units. Forty-five patients with no facial anomalies (control group) were used initially to evaluate the method and to obtain variables for statistical reference. Thirty-five patients operated on for unilateral cleft lip and palate (cleft group) were then analysed and compared with the control group. Individual scores were obtained for each patient, computed, and correlated with those established by a lay panel. Statistical analysis showed good sensitivity and reliability (R>0.8).     

宫腔镜电切术治疗子宫纵隔23例临床分析

目的:探讨子宫纵隔患者接受宫腔镜电切术治疗后的妊娠结局。方法:回顾性分析23例子宫纵隔患者接受宫腔镜下子宫纵隔电切术后的妊娠结局。结果:23例子宫纵隔患者中,子宫完全纵隔5例,不完全纵隔18例,23例均在超声监视下完成手术,平均手术时间25min,平均出血30ml。与术前相比,术后足月活产率从4.35%增至48.83%,流产率从85.55%降至30.43%,具有显著性差异(P<0.01)。结论:宫腔镜电切术治疗子宫纵隔是安全、有效地治疗方法,能显著提高足月活产率,明显降低流产率。     

去甲长春花碱加顺铂联合治疗晚期非小细胞肺癌近期疗效分析

目的:观察国产去甲长春花碱(NVB)加顺铂(DDP)联合治疗晚期非小细胞肺癌的疗效.方法:符合入选条件的ⅢB-Ⅳ期NSCLC患者48例,治疗方案NVB 25mg/m2,iv dl,8,DDP 20mg/m2,iv dl-5,21天为一周期,每3周重复一次,每例至少用2个周期.结果:PR 20例,SD 20例,PD 8例,有效率44%,中位生存期10个月.主要毒副反应为骨髓抑制,轻度的胃肠道反应及少数局部静脉炎.结论:国产去甲长春碱加顺铂联合化疗治疗晚期非小细胞肺癌疗效明显,毒副反应可耐受.     

红三叶总异黄酮对人乳腺癌MCF-7细胞增殖和凋亡的影响

目的研究红三叶总异黄酮对人乳腺癌MCF-7细胞的生长抑制和凋亡的诱导作用。方法在体外细胞培养下,采用不同质量浓度红三叶异黄酮和5-Fu处理MCF-7细胞,MTT法测定红三叶总异黄酮抑制MCF-7细胞增殖的量-效关系;TUNEL法、流式细胞术分析其细胞凋亡率和周期分布。结果红三叶总异黄酮对MCF-7细胞增殖的抑制作用随药物质量浓度提高和作用时间延长而更加明显,而在低质量浓度时却出现轻微促细胞增殖现象。TUNEL法、流式细胞术分析结果表明,红三叶总异黄酮能诱导MCF-7细胞凋亡,阻滞细胞于G2/M期,降低细胞增殖指数,且这种作用也存在剂量-效应关系。结论红三叶总异黄酮可抑制MCF-7细胞增殖并诱导其凋亡,具有抗肿瘤作用。     

Modulation of beta-adrenergic receptors in the pituitary gland following adrenalectomy in rats

The effects of adrenalectomy on beta-adrenergic receptors in the rat pituitary were examined using quantitative in vitro autoradiography with 125I-iodocyanopindolol (125ICYP). 125ICYP binding in the anterior, intermediate and posterior lobes of the pituitary gland was significantly increased in chronically adrenalectomized rats. The increase in 125ICYP binding sites in the rat pituitary following adrenalectomy was not reversed by glucocorticoid replacement with dexamethasone. These data indicate that catecholamines of adrenomedullary origin are capable of modulating beta-adrenergic receptors in the pituitary gland and suggest that peripheral epinephrine may be important in regulating pituitary hormone secretion.     

实时荧光定量PCR检测PML-RARα融合基因的临床应用研究

目的 利用PML-RARα融合基因作为标志物,应用实时荧光定量PCR技术,监测急性早幼粒细胞白血病(APL)微小残留病(minimal residual disease,MRD)患者体内病毒的状态,探讨预测APL复发风险的方法.方法 利用本实验室建立的定量检测APL患者PML-RARα融合基因的方法,对25例处于初诊、完全缓解(complete remission,CR)、复发等不同阶段的APL患者进行了PML-RARα融合基因的定量检测,并对其中6例患者的MRD状态进行了动态监测.结果 应用荧光定量PCR技术分析患者初诊、CR和复发状态的PML-RARα基因对数值(lg)水平,结果 显示有统计学意义(x2=6.847,P＜0.05);随访期患者的PML-RARα对数值(lg)水平变化较大:初诊时较高(-0.61±0.79),CR时下降(-1.31±0.62),复发时又出现上升(-0.57±0.44),提示若该患者在CR时的对数值呈现上升趋势,患者复发的机率大.结论 应用实时荧光定量(RQ)-PCR技术定量检测PML-RARα融合基因,对监测APL患者MRD状态具有临床适用性,随访期的MRD动态追踪监测具有预后价值.

An unexpected Q fever endocarditis. Report of a case

A man with blood culture negative endocarditis since 1983 received in October 1984 a mitral valve bioprosthesis. Reintervention in April 1985 was performed because of a paravalvular leak. In September 1985, mitral insufficiency reappeared and high-titer phase II Q fever antibody was detected, which has since then persisted with concomitant high-titers of phase I antibody. In serum from 1983, phase II antibody was detected on reexamination in September 1985. For unexplained reasons this antibody had not been detected in 1983. The patient has since September 1985 been successfully treated with doxycycline. The current literature is reviewed. Q fever endocarditis should be considered also in Belgium in culture negative endocarditis even in persons with no previous history of valvular disease and no known exposure to animals or unpasteurized dairy products. Quality of viral reagents and diagnosis present sometimes a challenge.
