The purpose of this study was to provide information concerning the possible early effects of smoking on measures of respiratory capacity and control in young adult female smokers vs. nonsmokers. In particular, maximum performance test results (vital capacity and maximum phonation time) and measures of air pressures and airflows during voiceless, stop‐plosive productions were analysed. Subjects were 45 female nonsmokers and 30 female smokers (total n = 75) between the ages of 18–30 years. For the purposes of this study, a smoker was defined as any subject who, at the time of this study, had smoked at least two cigarettes per day for at least 1 year. All of the subjects in the nonsmoker category were those who, at the time of this study, did not smoke and who had not smoked for at least 5 years prior to the study. Vital capacity measures were conducted using a hand‐held digital spirometer, while maximum phonation productions and voiceless, stop‐plosive pressures and flows were recorded and measured using the Aerophone II Model 6800 (Kay Elemetrics Corp., Lincoln Park, NJ). Results showed significantly lower vital capacities and maximum phonation times in smokers vs. nonsmokers. The maximum phonation time task was also produced with significantly higher mean airflow rates in smokers than nonsmokers. In addition, the smokers were observed to produce significantly greater peak and mean pressures during the voiceless stop‐plosive task than the nonsmokers. A weak, but significant correlation was observed between the number of days the subject had smoked and the mean pressure produced during the voiceless, stop‐plosive task. The findings of reduced respiratory capacity and control in smokers may be associated with factors such as increased bronchial reactivity secondary to exposure to cigarette smoke and/or mild airway obstruction, increased airflow secondary to increased glottal gap size during phonation, and increased vocal fold mass and/or inefficiency in vocal fold closure. The aim of the present study was to determine the role of chronic stress in the pathogenesis of ligature-induced periodontal disease in rats. Fifty-three Wistar rats were randomly assigned to 4 experimental groups: 1--ligature; 2--ligature + stress; 3--stress only; 4--control. After 30 days the animals were sacrificed, blood samples were collected and histological sections were made for histometric analysis. The stress parameters evaluated were weight of thymus, spleen, adrenal glands and plasma glucocorticoid levels. Analysis of adrenal glands showed statistically significant differences between stressed and non-stressed groups (one-way ANOVA, p < 0.05). Plasma glucocorticoid levels were higher in Group 3 and lower in Group 2 (81.1 nmol/I versus 62.5 nmol/l, p < 0.05). Histometric measurements from the bone crest and from the first attached fiber were taken for all groups and for Groups 1 and 2 for the sites with and without ligatures. The ligature sites always displayed higher mean values than non-ligated sites (paired sample t test, p < 0.05). No statistically significant differences were observed between Groups 1 and 2 with regard to the ligated sites. However, differences were observed between Groups 1 and 2 in histometric bone levels in the non-ligated sites (mean values of 0.81 and 0.55 mm, respectively, p < 0.05). It may be concluded that stress can have a possible role in the activation of the hypothalamus-pituitary-adrenal (HPA) axis, with different levels of glucocorticoid release.  相似文献

Hydriding/dehydriding properties of a series of LaNi5 based alloys were compared by applying both hydrogen gas phase and electrochemical hydrogen charge/discharge methods. The highest hydrogen absorption capacity of 1.4 wt.% H2 was found for LaNi4.3Co0.4Al0.3, although LaNi4.8Sn0.2 also reveals comparable hydrogen capacity (>1.3%). A significant difference in the hydriding kinetics was observed for all studied alloys before and after activation. The activated alloys (5 cycles at 65 °C, 40 atm. H2) reach their maximum capacities after less than a minute, whereas the pure LaNi5 alloy needs several minutes for complete hydriding. The electrochemical hydriding/dehydriding behavior of the alloys reveals superior performance of LaNi4.3Co0.4Al0.3 and LaNi4.8Sn0.2 compared to the other compositions studied, as the capacity of LaNi4.8Sn0.2 decreases by only 10% for 60 charge/discharge cycles at a current density of 100 mA/g. Good agreement between the hydrogen sorption kinetics of the alloys obtained electrochemically and from hydrogen gas phase has also been observed.  相似文献

Phospholipase D (PLD) regulates the polymorphonuclear leukocyte (PMN) functions of phagocytosis, degranulation, and oxidant production. Ceramide inhibition of PLD suppresses PMN function. In streptolysin O-permeabilized PMNs, PLD was directly activated by guanosine 5'-[gamma-thio]triphosphate (GTP gamma S) stimulation of adenosine diphosphate (ADP)-ribosylation factor (ARF) and Rho, stimulating release of lactoferrin from specific granules of permeabilized PMNs; PLD activation and degranulation were inhibited by C2-ceramide but not dihydro-C2-ceramide. To investigate the mechanism of ceramide's inhibitory effect on PLD, we used a cell-free system to examine PLD activity and translocation from cytosol to plasma membrane of ARF, protein kinase C (PKC)alpha and beta, and RhoA, all of which can activate PLD. GTP gamma S-activated cytosol stimulated PLD activity and translocation of ARF, PKC alpha and beta, and RhoA when recombined with cell membranes. Prior incubation of PMNs with 10 microM C2-ceramide inhibited PLD activity and RhoA translocation, but not ARF1, ARF6, PKC alpha, or PKC beta translocation. However, in intact PMNs stimulated with N-formyl-1-methionyl-1-leucyl-1-phenylalamine (FMLP) or permeabilized PMNs stimulated with GTP gamma S, C2-ceramide did not inhibit RhoA translocation. Exogenous RhoA did not restore ceramide-inhibited PLD activity but bound to membranes despite ceramide treatment. These observations suggest that, although ceramide may affect RhoA in some systems, ceramide inhibits PLD through another mechanism, perhaps related to the ability of ceramide to inhibit phosphatidylinositol-bisphosphate (PIP2) interaction with PLD.  相似文献

Obesity is a world problem that requires a better understanding of its physiological and genetic basis, as well as the mechanisms by which the hypothalamus controls feeding behavior. The volcano mouse Neotomodon alstoni develops obesity in captivity when fed with regular chow diet, providing a novel model for the study of obesity. Females develop obesity more often than males; therefore, in this study, we analysed in females, in proestrous lean and obese, the differences in hypothalamus expression of receptors for leptin, ghrelin (growth hormone secretagogue receptor GHS-R), and VPAC, and correlates for plasma levels of total ghrelin. The main comparisons are between mice fed ad libitum and mice after 24 hours of fasting. Mice above 65 g body weight were considered obese, based on behavioral and physiological parameters such as food intake, plasma free fatty acids, and glucose tolerance. Hypothalamic tissue from obese and lean mice was analysed by western blot. Our results indicate that after ad libitum food access, obese mice show no significant differences in hypothalamic leptin receptors, but a significant increase of 60% in the GHS-R, and a nearly 62% decrease in VPAC2 was noted. After a 24-hour fast, plasma ghrelin increased nearly two fold in both lean and obese mice; increases of hypothalamic leptin receptors and GHS-R were also noted, while VPAC2 did not change significantly; levels of plasma free fatty acids were 50% less after fasting in obese than in lean animals. Our results indicate that in obese N. alstoni mice, the levels of orexigenic receptors in the hypothalamus correlate with overfeeding, and the fact that lean and obese females respond in different ways to a metabolic demand such as a 24-hour fast.  相似文献

Background

As low bone mineral density is a risk factor for fracture in childhood, optimizing age appropriate bone mass is recommended and might lower the impact of bone loss related to age. Consumption of omega-3 polyunsaturated fatty acids (PUFAs), including eicosapentaenoic and docosahexaenoic (DHA) acids have been shown to beneficially modulate bone metabolism. The objective of this study was to determine the incidence of fracture in neonates receiving a fish compared with soybean oil–based intravenous lipid emulsion and evaluate the effect of varying dietary omega-3 PUFA consumption on growing bone in young mice.

Materials and methods

Eligibility criteria for the clinical study included gestational age ≤37 wk and parenteral nutrition–dependence for ≥4 wk. Radiographs were reviewed after lipid initiation to identify radiologic bone fracture. The animal study evaluated female C57/Bl6 mice randomized into one of five groups from age 3–12 wk, at which time femurs were harvested for micro–computed tomography and light microscopy analysis.

Results

A lower incidence of bone fracture was found in neonates maintained on fish compared with soybean oil. In the animal study, findings suggest the DHA diet provides the best protection against trabecular bone loss as evidenced by increased bone volume fraction, increased trabecular number, and decreased trabecular separation on micro–computed tomography. These protective effects appeared to affect the bone microstructure alone.

Conclusions

The lower fracture risk observed in fish oil fed neonates in combination with the protective effects of DHA observed in the femurs of young C57/BL6 mice suggest an important role for omega-3 PUFAs on bone growth.  相似文献
