Terminal deletions of the chromosome 6q27 region are rare genomic abnormalities, linked to specific brain malformations and other neurological phenotypes. Reported cases have variable sized genomic deletions that harbor several genes including the DLL1 and TBP. We report on an inherited 0.38 Mb terminal deletion of chromosome 6q27 in a 22‐week fetus with isolated bilateral ventriculomegaly and her affected mother using microarray‐based comparative genomic hybridization and fluorescent in situ hybridization (FISH). The deleted region harbors at least seven genes including DLL1 and TBP. The affected mother had a history of hydrocephalus, developmental delay, and seizures commonly associated with DLL1 and TBP 6q27 deletions. This deletion is one of the smallest reported isolated 6q27 terminal deletions. Our data provides additional evidence that haploinsufficiency of the DLL1 and TBP genes may be sufficient to cause the ventriculomegaly, seizures, and developmental delays associated with terminal 6q27 deletions, indicating a plausible role in the abnormal development of the central nervous system. 相似文献

Background: Natural Killer (NK) cell plays an important role in the innate immune system and is known to produce IFN-γ at an early stage of infection that is essential to eliminate intracellular infection like Leishmania spp. It is already established that Leishmania parasite inhibits the activity of NK cells, avoiding the encounter with the early innate immune response. This, in turn, favors establishment and further dissemination of the infection. Methods: In the present study, we have tried to measure the frequency of different phenotypic subsets of NK cells among visceral leishmaniasis (VL) patients. Results: We have phenotyped three distinct three distinct subsets (CD56–CD161+, CD56+CD161–, and CD56+CD161+) of NK (CD3–) cell using their specific markers CD161 and CD56. Conclusion: Interestingly, we observed selective loss of CD56+CD161+ subset of circulating NK (CD3–) cells. Importantly, the other subsets (i.e., CD56?CD161+ and CD56+CD161–) of circulating NK cells remain unaffected as compared with healthy subjects. 相似文献

Objective : To conduct a clinical study of holoprosencephaly (HPE).Method : Thirteen cases of HPE were studied regarding their clinical features, family history, and prenatal and imaging studies.
Chromosomal analysis was done whenever fresh sample was available.Results : Six cases were antenatally detected by ultrasound; four cases were stillborn. Three cases were identified by neuroimaging

Two of these had subtle facial features and microcephaly. Karyotype was abnormal in 2 of 7 cases studied.Conclusion : Most of the cases of HPE present antenatally or at birth. Milder forms like lobar and semilobar can present as developmental
delay during infancy. Facial anomalies are usually associated with HPE. Chromosomal study of the case and clinical examination

To find some prognostic factors for the outcome of frozen-thawed cycles, we
have retrospectively analysed all frozen pre-embryos that were thawed
during 1993 and 1994 at two in-vitro fertilization (IVF) units in Sweden.
Supernumerary pre-embryos were frozen from 551 oocyte retrievals and these
resulted in 660 frozen-thawed cycles which lead to 623 thawed embryo
transfers. The outcome of these transfers was 137 clinical pregnancies with
a pregnancy rate of 22% per frozen-thawed embryo transfers. Women <40
years of age had a higher birth rate than those > or =40 years, 19 and
5% respectively (P < 0.01). Transfers with two and three pre-embryos
resulted in pregnancy rates of 23 and 27%, respectively, compared with 14%
for transfer of one embryo. A pregnancy resulting from the initial embryo
transfers had a predictive value for results of the subsequent
frozen-thawed cycle. Embryo grade and cleavage stage at the time of
freezing was important for the survival of the frozen-thawed pre-embryos.
The pregnancy rate was not influenced by the cleavage stage, but a tendency
toward a lower pregnancy rate was seen for the embryos with lower grading.
To conclude, cryopreservation seems to be beneficial in women <40 years
of age, who have supernumerary pre-embryos of good quality for freezing and
of which at least two can be transferred.

Campylobacter coli in swine production: antimicrobial resistance mechanisms and molecular epidemiology 下载免费PDF全文
The aim of this study was to determine antimicrobial resistance, to evaluate and compare the use of two genotyping methods for molecular epidemiology purposes, and to determine the genotypic diversity of Campylobacter coli of porcine origin. A total of 100 C. coli isolates from swine were tested for susceptibility to six antimicrobials using the agar dilution method and genotyped using two high-resolution fingerprinting approaches: multilocus sequence typing (MLST) and pulsed-field gel electrophoresis (PFGE). Evaluation of the methods was based on their resistance patterns, discriminatory indexes (DI), high test throughputs, costs, and turnaround times. Resistance to erythromycin and tetracycline was the most common. Both genotypic methods were found to have high discriminatory power, although MLST had a higher DI (0.936) than PFGE (DI = 0.889). It also had a higher throughput than PFGE. Isolates were clustered into 27 groups by MLST compared to 11 by PFGE. MLST was able to further discriminate the isolates grouped under the same cluster by PFGE. Out of the 65 MLST sequence types (STs) identified among the total isolates, 50 were reported for the first time. Most STs were found to be specific to the farm (n = 38) and to slaughter (n = 22). Resistance against tetracycline and erythromycin was encoded by the tet(O) gene and a A2075G point mutation in the 23S rRNA gene, respectively. A high ciprofloxacin MIC (>64 microg/liter) was conferred by a point mutation in the gyrA gene. The weak clonal structure of the C. coli population among swine was further highlighted by the index of association value of 0.293. The findings of this study indicate that multidrug-resistant diverse C. coli strains exhibiting resistance to ciprofloxacin and erythromycin are concerning, since these are the drugs of choice for treating invasive campylobacteriosis cases in humans. 相似文献

Candida lipolytica is weakly pathogenic yeast, which is rarely isolated from the blood. We recovered this species from repeated blood samples and in the central venous catheter in a debilitated pediatric patient of tubercular meningitis. Identity was established on the basis of colony morphology and sugar assimilation tests (ID 32C assimilation profile). The fungemia and associated fever subsided after the removal of catheter and amphotericin B therapy. The data suggest that though of low virulence and usually a contaminant, C. lipolytica is emerging yeast pathogen in cases of catheter-related candidemia. Pathogenicity is indicated by isolation from repeated samples as in our case. Intensive therapy is recommended in cases not resolving spontaneously or responding to removal of catheter alone. 相似文献

Reciprocal effect of Waardenburg syndrome mutations on DNA binding by the Pax-3 paired domain and homeodomain 总被引:1,自引:1,他引:1
The Pax-3 protein contains two DNA-binding domains, a paired domain and a
homeodomain. Mutations in Pax-3 cause Waardenburg syndrome (WS) in humans
and the mouse Splotch (Sp) phenotype. In the Sp-delayed mouse, a mutation
in the Pax-3 paired domain (G9R) abrogates the DNA-binding activity of both
the paired domain and the homeodomain, suggesting that they may
functionally interact. To investigate this possibility further, we have
analyzed the DNA-binding properties of additional point mutants in the
Pax-3 paired domain and homeodomain that occur in WS patients (F12L, N14H,
G15S, P17L, R23L, G48A, S51F and G66D in the paired domain, V47F and R53G
in the homeodomain), the Pax-1 un mutation (G15A) and a substitution
associated with Peters' anomaly in the PAX-6 gene (R23G). Within the paired
domain, seven of 10 mutations were found to abrogate DNA-binding by the
paired domain. Remarkably, these seven mutations also affected DNA binding
by the homeodomain, causing either a complete loss (P17L and G66D), a
reduction (R23G, R23L, G15S and G15A) or an increase in DNA-binding
activity (N14H). In addition, the effect of paired domain mutations
occurred at the level of monomer formation by the homeodomain, while the
dimerization potential of this domain seemed unaffected in mutants where it
could be analyzed. Furthermore, while both homeodomain mutations were found
to abolish DNA binding by this domain, the R53G mutation also abrogated DNA
binding by the paired domain. The important observation that independent
mutations in either domain can affect DNA binding by the other in the
intact Pax- 3 protein strongly suggests that the two domains are not
functionally independent but bind DNA through cooperative interactions.
Modeling the deleterlous mutations on the three-dimensional structure of
the paired domain of Drosophila Prd shows that these mutations cluster at
the DNA interface, thus suggesting that a series of DNA contacts are
essential for DNA binding by both the paired domain and the homeodomain of
Pax-3.

Congenital nephrogenic diabetes insipidus (NDI) is a rare disease caused
most often by mutations in the vasopressin V2 receptor (AVPR2). We studied
a family which included a female patient with NDI with symptoms dating from
infancy. The patient responded to large doses of desmopressin (dDAVP) which
decreased urine volume from 10 to 4 I/day. Neither the parents nor the
three sisters were polyuric. The patient was found to be a compound
heterozygote for two novel recessive point mutations in the aquaporin-2
(AQP2) gene: L22V in exon 1 and C181W in exon 3. Residue Cys181 in AQP2 is
the site for inhibition of water permeation by mercurial compounds and is
located near to the NPA motif conserved in all aquaporins. Osmotic water
permeability (Pf) in Xenopus oocytes injected with cRNA encoding C181W-AQP2
was not increased over water control, while expression of L22V cRNA
increased the Pf to approximately 60% of that for wild-type AQP2.
Co-injection of the mutant cRNAs with the wild-type cRNA did not affect the
function of the wild-type AQP2. Immunolocalization of AQP2-transfected CHO
cells showed that the C181W mutant had an endoplasmic reticulum-like
intracellular distribution, whereas L22V and wild-type AQP2 showed endosome
and plasma membrane staining. Water permeability assays showed a high Pf in
cells expressing wild-type and L22V AQP2. This study indicates that AQP2
mutations can confer partially responsive NDI.

Competitive control of the self-renewing T cell repertoire 总被引:1,自引:0,他引:1
We develop a mathematical model for the self-renewing part of the T cell
repertoire. Assuming that self-renewing T cells have to be stimulated by
immunogenic MHC-peptide complexes presented on the surfaces of
antigen-presenting cells, we derive a model of T cell growth in which
competition for MHC-peptide complexes limits T cell clone sizes and
regulates the total number of self-renewing T cells in the animal. We show
that for a sufficient diversity and/or degree of cross-reactivity, the
total T cell number hardly depends upon the diversity of the T cell
repertoire or the diversity of the set of presented peptides. Conversely,
for repertoires of lower diversity and/or cross-reactivity, steady-state
total T cell numbers may be limited by the diversity of the T cells. This
provides a possible explanation for the limited repertoire expansion in
some, but not all, mouse T cell re-constitution experiments. We suggest
that the competitive interactions described by our model underlie the
normal T cells numbers observed in transgenic mice, germ-free mice and
various knockout mice.
