Cervical cancer remains a significant cause of morbidity and mortality in women worldwide and is the leading cause of cancer-related death in Botswana. It is well established that women with HIV have a higher risk of persistent HPV infection leading to cervical cancer. We assessed HPV prevalence and genotype distribution in 126 tissue specimens from confirmed invasive cervical cancer cases using Abbott real-time PCR assay. Overall, 88 (69.8%) women were HIV-infected. Fifty-seven (64.8%) of the HIV-infected women had a baseline CD4+ count ≥350 cells/μl, and 82 (93.2%) were on antiretroviral therapy at the time of cervical cancer diagnosis. The median age of HIV-infected patients was significantly younger than that of HIV-uninfected patients (p < 0.001). HPV DNA was detected in all of 126 (100%) of tissues analyzed in our study. The HPV genotypes identified included the HPV-16 (75.4%), HPV-18 (28.6%) and other high-risk (hr) HPV genotypes (16.7%). HIV infection was positively associated with the presence of the HPV-16 genotype (p = 0.036), but not with HPV-18 or with other high-risk (hr)-HPV genotypes. Thirty-three percent of the patients had multiple hr-HPV genotypes, with higher rates in HIV-infected women. These results highlight the importance and potential impact of large-scale HPV vaccination programs covering HPV-16 and HPV-18 genotypes in countries like Botswana with high burden of HIV infection. 相似文献

The present study demonstrates the applicability of a strategy involving use of liquid chromatography (LC) and liquid chromatography-mass spectrometry (LC-MS) techniques for the identification and characterization of minute quantities of degradation products, without their isolation from the reaction matrix in pure form. Valsartan was used as a model drug. It was subjected to forced degradation studies under the International Conference on Harmonisation (ICH) prescribed conditions of hydrolysis (acid, base and neutral), photolysis, oxidation and thermal stress. The drug showed lability under acid/neutral hydrolytic and photolytic conditions, while it was stable to base hydrolytic, oxidative and thermal stress. Three small degradation products were formed, which were separated on a C-18 column using a gradient method. The same were characterized with the help of their fragmentation pattern and accurate masses obtained upon LC-MS/TOF analyses and online H/D exchange studies. The structures were supported by appropriate mechanistic explanation. The strategy involving use of LC and LC-MS for the identification and characterization of minute quantities of degradation products was applied on a model drug, valsartan. Malakoplakia is a rare chronic granulomatous inflammatory disease, related to immune deficiency and impaired bactericidal macrophage activity. Common sites of involvement include urinary bladder and kidney followed by gastrointestinal tract. We present an unusual case of abdominal malakoplakia diagnosed preoperatively on ultrasound‐guided fine‐needle aspiration cytology (FNAC). It presented as a large locally aggressive mass with extensive involvement of multiple bowel loops in an 18‐year‐old boy following blunt trauma to the abdomen. The demonstration of calcified laminated intracytoplasmic Michaelis–Gutman bodies in histiocytes is a reliable diagnostic feature of malakoplakia on FNAC smears, which can guide an appropriate medical management. Purpose Ether-a-go-go (EAG) or EAG-related (ERG) voltage-gated potassium ion channels are involved in tumor generation and progression.
Their over- and/or misexpression has been demonstrated in various tumors, and inhibition of these channels has suppressed
proliferation of various cancer cells. We investigate and compare the pattern of expression of EAG and human ERG (HERG) channels
in renal cell carcinoma and “normal” renal tissue.
Method Tissue samples, obtained at the time of radical nephrectomy from the tumor-bearing areas, and uninvolved renal tissue were
preserved in 4% paraformaldehyde and cryosectioned at 20 μm. Immunohistochemical and Western blot analysis was performed on
the tumor and uninvolved kidney parenchyma by incubating with polyclonal anti-HERG 1b (Alomone Lab, Israel), anti-EAG1, and
anti-EAG2. Pattern of expression of EAG/HERG channels in normal renal tissue and carcinoma were noted and compared.
Results The study was performed on 16 radical and four partial nephrectomy specimens (n = 20). All tumors in the cohort were clear cell renal carcinoma. Normal renal tissue was found to exhibit heterogeneous cytoplasmic
positivity for EAG1 and focal HERG immunoreactivity (IR) in the proximal (PCT) and distal convoluted tubules (DCT). EAG2 IR
was absent in the normal renal tissue. Clear cell RCC demonstrated a loss of HERG expression while diffuse overexpression
of EAG1 and EAG2 was noted. Western blot analysis corroborated the immunohistochemical observations.
Conclusions In our study both EAG1 and EAG2 potassium channels were overexpressed in clear cell renal cancer. In contrast to other adenocarcinomas,
there is loss of HERG expression in clear cell RCC, which may possibly explain its chemoresistance. These ion channels may
