A fused gene of the gag and env sequences of human T-cell leukemia virus type I (HTLV-I), the causative agent of adult T-cell leukemia, was constructed in vitro and expressed in Escherichia coli. The gag-env hybrid protein accumulated as insoluble granules with a yield of approximately 12% of the total proteins. In an enzyme-linked immunosorbent assay done with the use of the gag-env hybrid protein, all 57 seropositive sera gave positive signals, and none of the sera from normal persons did. This system can produce large quantities of the gag-env hybrid protein, which can be used for mass screening of human sera for HTLV-I infection.  相似文献

We investigated nitric oxide (NO)-producing neurons in the amygdala which project to the hypothalamic paraventricular nucleus (PVN) of the rat using retrograde tracing and NADPH-diaphorase histochemistry. Numerous NADPH-diaphorase positive neurons with moderate staining were observed mainly in the medial amygdaloid nucleus. We confirmed that these NADPH-diaphorase positive neurons are identical to NO synthase (NOS)-immunoreactive neurons by double staining with NADPH-diaphorase histochemistry and NOS immunocytochemistry. Most neurons containing cholera toxin B subunit (CTb) – which was retrogradely transported from the PVN – were observed in the medial amygdaloid nucleus. In other amygdaloid nuclei, they were observed much less in the central nucleus, basomedial and anterior cortical nucleus. Double labeled neurons by NADPH-diaphorase and CTb were also identified mostly in the medial nucleus. Approximately 40% of the neurons projecting to the PVN were nitrergic neurons and 16% of NADPH-diaphorase positive neurons in the medial nucleus were revealed to project to the PVN. These results suggest that NO-producing neurons in the medial amygdala directly innervate PVN neurons and regulate neuroendocrine systems such as vasopressin and corticotropin releasing factor release.  相似文献

We found anti-nuclear antibodies (ANA) (1 : 20 or higher) in sera from 11 of 16 patients with a diagnosis of the optic-spinal form of multiple sclerosis (OpS-MS) and 13 of 59 patients with other forms of MS (other MS), both rates being significant (P = 0.0004). Six of the OpS-MS patients had a high level of ANA (1 : 80 or higher), while levels were high in only three of the other MS patients, showing a significant difference (P = 0.0022). Titres of ANA were significantly higher in OpS-MS patients than in other MS patients (P < 0.0001). There was no relationship between the presence of ANA and age in OpS-MS patients, while patients with were older than those without high ANA levels among other MS patients. ANA-positive rate and titres were significantly higher in OpS-MS patients than in sex- and age-matched other MS patients. These results support the concept that OpS-MS constitutes a distinct subgroup among patients with a clinical diagnosis of MS and may indicate that systemic dysregulation of the inflammatory or immune response is more common in OpS-MS patients; the possible pathogenetic relevance of ANA-associated vasculopathy to some patients with a clinical diagnosis of OpS-MS should be considered. The sex and age distributions of values of bone metabolic markers were investigated among the inhabitants of a mountain village in Wakayama Prefecture, Japan. The survey was carried out in Miyama village, which has a population of 2372 (1129 men and 1243 women) based on the resident registration of 1990. In this village, all residents aged 40 years or over are eligible for general mass health examinations once a year. In the present study, bone metabolic markers were measured in recruited participants in the health examinations conducted in 1992 in each of four age strata: 40–49, 50–59, 60–69, and 70–79 years. The bone metabolic markers measured were osteocalcin (BGP), carboxyterminal peptide of type I procollagen (PICP), alkaline phosphatase (ALP), crosslinked carboxyterminal telopeptide region of type I collagen (ICTP), pyridinoline (Pyr), and deoxypyridinoline (D-Pyr). In men in their 40s, 50s, 60s, and 70s, the mean serum BGP level was 6.15, 7.04, 7.65, and 7.69 ng/ml, respectively; the level in men in their 40s was significantly lower than that in those in their 60s. The serum PICP level in the 40- to 49-year age group was significantly higher than those in the other age groups, while that of serum ICTP tended to increase with age. The mean values of urinary Pyr and D-Pyr were higher in the older groups. In women, the mean values of all bone metabolic markers were all higher among those in the 50–59, 60–69, and 70–79 age groups compared to those in the 40–49 age group. These findings suggest that active bone metabolic turnover occurs even in the older age groups in both men and women.  相似文献

Two siblings who developed adult T-cell leukemia (ATL) are presented. The patient and 7 of 26 healthy family members examined had the serum antibodies against ATL-associated antigens (ATLA). This family study shows that two main routes of transmission of human T-cell leukemia virus (HTLV) may be involved: one is the route from parents to children and the other is horizontal transmission among spouses, especially from husband to wife; the anti-ATLA-positive family is considered to be a high-risk group for ATL.  相似文献

Postnatal changes in acid phosphatase, β-glucuronidase, arylsulfatase and cathepsin D in ocular tissues and liver were studied biochemically. Lysosomal enzymes, except for cathepsin D, showed postnatal age-dependent variations in activity. We examined recurrent vitreous hemorrhage in two boys with prepapillary arterial loops. Both patients complained of visible floaters in the left eye during gymnastic exercise. Prepapillary arterial loops and vitreous hemorrhage were found in the left eye in both boys. The vitreous hemorrhage decreased gradually, but recurred. Prepapillary arterial loops may be a risk factor for recurrent vitreous hemorrhage in children. The authors have stated that they do not have a significant financial interest or other relationship with any product manufacturer or provider of services discussed in this article.
