The effector functions of natural killer cells are regulated by activating receptors, which recognize stress-inducible ligands expressed on target cells and signal through association with signaling adaptors. Here we developed a mouse model in which a fusion of the signaling adaptor DAP10 and ubiquitin efficiently downregulated expression of the activating receptor NKG2D on the surfaces of natural killer cells. With this system, we identified coupling of the signaling pathways triggered by NKG2D and DAP10 to those initiated by the interleukin 15 receptor. We suggest that this coupling of activating receptors to other receptor systems could function more generally to regulate cell type-specific signaling events in distinct physiological contexts.  相似文献

Nitric oxide (NO(.)) is a short-lived physiological messenger. Its various biological activities can be preserved in a more stable form of S-nitrosothiols (RS-NO). Here we demonstrate that at physiological NO(.) concentrations, plasma albumin becomes saturated with NO(.) and accelerates formation of low-molecular-weight (LMW) RS-NO in vitro and in vivo. The mechanism involves micellar catalysis of NO(.) oxidation in the albumin hydrophobic core and specific transfer of NO(+) to LMW thiols. Albumin-mediated S-nitrosylation and its vasodilatory effect directly depend on the concentration of circulating LMW thiols. Results suggest that the hydrophobic phase formed by albumin serves as a major reservoir of NO(.) and its reactive oxides and controls the dynamics of NO(.)-dependant processes in the vasculature.  相似文献

In this study, an integrated approach was used for the preparation of a nanotitanium-based bioactive material. The integrated approach included three methods: severe plastic deformation (SPD), chemical etching and atomic layer deposition (ALD). For the first time, it was experimentally shown that the nature of the etching medium (acidic or basic Piranha solutions) and the etching time have a significant qualitative impact on the nanotitanium surface structure both at the nano- and microscale. The etched samples were coated with crystalline biocompatible TiO2 films with a thickness of 20 nm by Atomic Layer Deposition (ALD). Comparative study of the adhesive and spreading properties of human osteoblasts MG-63 has demonstrated that presence of nano- and microscale structures and crystalline titanium oxide on the surface of nanotitanium improve bioactive properties of the material.  相似文献

Objective: To compare pregnancy outcomes of two consecutive pregnancies in a cohort of women with recurrent pregnancy loss (RPL), in order to determine the long-term prognosis of women with RPL managed in a dedicated RPL clinic.

Methods: A retrospective cohort study including 262 patients with two or more consecutive pregnancy losses followed by two subsequent pregnancies – index pregnancy (IP) and post-index pregnancy (PIP). All patients were evaluated and treated in the RPL clinic in the Soroka University Medical Center.

Results: Comparing IP with PIP, no significant difference in perinatal outcome was observed. The perinatal outcome remained encouraging with approximately 73% birth rate (73.7% versus 72.5%; p?=?0.83). Only 11% of the women with RPL continued to experience pregnancy losses for two subsequent pregnancies. In a multivariate logistic regression analysis, number of miscarriages pre-Index was the only factor independently associated with birth in the PIP.

Conclusion: There is no significant difference between IP and PIP regarding perinatal outcome. Appropriate management in the RPL clinic conferred a significant beneficial effect on long-term pregnancy outcome of a cohort of women with RPL.  相似文献

We have recently shown that the development of endothelial dysfunction in lambs with increased pulmonary blood flow (PBF) correlates with a decrease in peroxisome proliferator activated receptor-γ (PPAR-γ) signaling. Thus, in this study we determined if the loss of PPAR-γ signaling is necessary and sufficient to induce endothelial dysfunction by exposing lambs with normal PBF to the PPAR-γ antagonist, GW9662. Two-weeks of exposure to GW9662 significantly decreased both PPAR-γ protein and activity. In addition, although eNOS protein and nitric oxide metabolites (NOx) were significantly increased, endothelial dependent pulmonary vasodilation in response to acetylcholine was attenuated, indicative of endothelial dysfunction. To elucidate whether downstream mediators of vasodilation were impaired we examined soluble guanylate cyclase (sGC)-α and β subunit protein, cGMP levels, and phosphodiesterase 5 (PDE5) protein and activity, but we found no significant changes. However, we found that peroxynitrite levels were significantly increased in GW9662-treated lambs and this correlated with a significant increase in protein kinase G-1α (PKG-1α) nitration and a reduction in PKG activity. Peroxynitrite is formed by the interaction of NO with superoxide and we found that there was a significant increase in superoxide generation in GW9662-treated lambs. Further, we identified dysfunctional mitochondria as the primary source of the increased superoxide. Finally, we found that the mitochondrial dysfunction was due to a disruption in carnitine metabolism. We conclude that loss of PPAR-γ signaling is sufficient to induce endothelial dysfunction confirming its important role in maintaining a healthy vasculature.  相似文献

ObjectiveTo evaluate the effect of a 12-month course of weekly lipid apheresis on vein graft patency after coronary artery bypass grafting (CABG) in patients with hyperlipidemia refractory to statins.MethodsIn a 12-month prospective controlled clinical trial we enrolled 34 male patients (mean age 57 ± 8 years) who passed through successful CABG and low-density lipoprotein cholesterol (LDL-C) level >2.6 mmol/L prior to the operation despite statin treatment. Patients were allocated into 2 groups: active (n = 17, weekly apheresis by cascade plasma filtration (CPF) plus atorvastatin), and control (n = 17, atorvastatin alone). Graft patency was evaluated by multislice computed tomography at 3 months and by angiography at 12 months after an operation.ResultsBoth groups were comparable in clinical and biochemical characteristics. During each CPF procedure, LDL-C level decreased by 64 ± 9%, apoB – by 65 ± 8%, Lp(a) – by 52 ± 15%,; these changes were significant compared to baseline and the control group. Mean net difference in LDL-C level between apheresis and control groups was 1.1 ± 0.3 mmol/L. Vein graft patency at study end was 88.2% (45 of 51) in the apheresis group versus 72.7% (40 of 55) in the control group (p = 0.05). Use of apheresis was associated with decreased vein graft occlusions by 46%: relative risk 0.54; 95% confidence interval 0.27 to 1.02; p = 0.05.ConclusionOur data suggest that the use of lipoprotein apheresis with CPF results in a better vein graft patency during the first year after CABG in patients with hyperlipidemia refractory to statins.  相似文献

Due to renovation and fighting in the world, a huge accumulation of construction and demolition waste is formed. These materials are effectively used as aggregates, but there is very little information about the use of scrap concrete to create cementless binders. The purpose of the work is to be a comprehensive study of the composition and properties of concrete wastes of various fractions with the aim of their rational use as cementless binders. The scientific novelty lies in the fact that the nature of the processes of structure formation of a cementless binder based on sandy fractions of the screening of fragments of destroyed buildings and structures, as a complex polyfunctional system, has been theoretically substantiated and experimentally confirmed. Different percentages of non-hydrated clinker minerals in concrete scrap were determined. In the smallest fraction (less than 0.16 mm), more than 20% of alite and belite are present. Waste of the old cement paste is more susceptible to crushing compared to the large aggregate embedded in it, therefore, particles of the old cement paste and fine aggregate predominate in the finer fractions of the waste. Comprehensive microstructural studies have been carried out on the possibility of using concrete scrap as a completely cementless binder using scanning electron microscopy, X-ray diffraction analysis, and differential thermal analysis. It has been established that for cementless samples prepared from the smallest fractions (less than 0.315 mm), the compressive strength is 1.5–2 times higher than for samples from larger fractions. This is due to the increased content of clinker minerals in their composition. The compressive strength of the cementless binder after 28 days (7.8 MPa), as well as the early compressive strength at the age of 1 day after steaming (5.9 MPa), make it possible to effectively use these materials for enclosing building structures.  相似文献

The aim of this study was to improve the properties of lightweight particleboards by their veneering. The industrially produced wood particles, rotary-cut birch veneer, expanded polystyrene (EPS) granules and urea-formaldehyde (UF) resin were used to manufacture non-veneered and veneered boards in laboratory conditions. The boards were manufactured with different densities of 350, 450 and 550 kg/m3 and with various levels of EPS content 4, 7 and 10%. Boards without EPS granules as the reference were also manufactured. Bending strength (MOR), modulus of elasticity in bending (MOE), internal bond (IB) strength, thickness swelling (TS) and water absorption (WA) of lightweight particleboards were determined. This study confirmed that veneering of lightweight particleboards by birch veneer improved mechanical properties significantly. The MOR and MOE of veneered boards throughout the whole density range of 350–550 kg/m3 meet the requirements of the CEN/TS 16368 for lightweight particleboards types LP1 and LP2. The IB strength of veneered boards only with density of 550 kg/m3 meets the requirements of CEN/TS 16368 (type LP1). The MOR, MOE and IB of non-veneered boards also meet the requirements of CEN/TS 16368 (type LP1) except boards with density of 350 kg/m3 for MOR and MOE, and except densities of 350 and 450 kg/m3 for IB.  相似文献
