Cell transplantation in wounded mixed connective tissues   总被引:1,自引:0,他引:1  
Direct transplantation of multipotent precursor cells into the periodontium could provide a therapeutic approach for restoring periodontal tissues destroyed by periodontitis or trauma. To improve the understanding of cell migration, proliferation, and differentiation, we used a rodent model combining orthodontic tooth movement and transplantation of Lac-Z-positive murine-cultured periodontal ligament (PL) or femur-derived bone marrow precursor cells into a defined mandibular wound site, thus promoting tissue regeneration in wounded periodontium. Our results show that in orthodontically traumatized tissues, transplanted PL and bone marrow cells migrated systemically, contributing to the repopulation of sites with reduced cell/matrix density. The transplanted PL cells proliferated in adjacent alveolar bone marrow spaces, thus migrating to vascular tissues in the PL. The capillary walls in the PL serve as delivery sites for these cells and other marrow-derived hematopoietic cells, including monocytes. The transplanted marrow cells, extracted from femur of transgenic (TgR) mice exhibited similar behavior to those of transplanted PL cells, showing high proliferative activity in alveolar marrow as well as intensive repopulating capacity in wounded periodontium. On the other hand, the buccal skin fibroblasts failed to migrate and home effectively and thus the transplantation of these cells had no effect on periodontium regeneration. Based on these results, we conclude that the transplanted PL and bone marrow cells migrate systemically and following a cyclical process of growth and development and differentiate into PL fibroblasts, osteoblasts, and cementoblasts, thereby contributing to periodontal regeneration.  相似文献

To investigate the role of SOX9 gene in amphibian gonadogenesis, we analyzed its expression during male and female gonadogenesis in Xenopus tropicalis. The results showed that in both sexes SOX9 mRNA and protein were first detectable after metamorphosis when the gonads were well differentiated and remained present until the adult stage. In the testis, SOX9 expression was restricted to the nucleus of Sertoli-like cells, similarly to what has been observed in other vertebrates suggesting a conserved role in vertebrate testicular differentiation. In the ovary, in sharp contrast with what has been observed in all vertebrates examined so far, the SOX9 protein was localized in the cytoplasm of previtellogenic oocytes before being translocated into the nucleus of vitellogenic oocytes suggesting an unexpected role during oogenesis. These results suggest that the SOX9 gene may not be a sex-determining gene in X. tropicalis and may play different functions in testicular and ovarian differentiation.  相似文献

DVL proteins are central mediators of the Wnt pathway and relay complex input signals into different branches of the Wnt signaling network. However, molecular mechanism(s) that regulate DVL-mediated relay of Wnt signals still remains unclear. Here, for the first time, we elucidate the functional significance of three DVL-1 lysines (K/Lys) which are subject to post-translational acetylation. We demonstrate that K34 Lys residue in the DIX domain regulates subcellular localization of β-catenin, thereby influencing downstream Wnt target gene expression. Additionally, we show that K69 (DIX domain) and K285 (PDZ domain) regulate binding of DVL-1 to Wnt target gene promoters and modulate expression of Wnt target genes including CMYC, OCT4, NANOG, and CCND1, in cell line models and xenograft tumors. Finally, we report that conserved DVL-1 lysines modulate various oncogenic functions such as cell migration, proliferation, cell-cycle progression, 3D-spheroid formation and in-vivo tumor growth in breast cancer models. Collectively, these findings highlight the importance of DVL-1 domain-specific lysines which were recently shown to be acetylated and characterize their influence on Wnt signaling. These site-specific modifications may be subject to regulation by therapeutics already in clinical use (lysine deacetylase inhibitors such as Panobinostat and Vorinostat) or may possibly have prognostic utility in translational efforts that seek to modulate dysfunctional Wnt signaling.  相似文献

This work aimed at the gamma irradiation-assisted synthesis of silver (Ag)-decorated ZnFe2O4 (ZFO) ferrite nanoparticles (NPs), which were tested for their antibacterial and antibiofilm activities against some pathogenic bacteria from medical operating room surfaces. The prepared Ag-decorated ZFO NPs were characterized via XRD, SEM, EDX, elemental mapping, and FTIR analysis. The antibacterial potential was tested as ZOI and MIC, while antibiofilm activity was estimated by the tube method. The growth curve assay, the effect of UV on the antimicrobial activity, and cell membrane leakage were evaluated, and the antibacterial reaction mechanism was investigated by SEM/EDX analysis. The XRD and FTIR results confirmed the successful preparation of Ag-decorated ZFO NPs. Antibacterial results revealed that the most potent decorated sample was Ag0.75@ZFO NPs, recording the most significant inhibition zone against Staphylococcus vitulinus (24.67 ± 0.577 mm) and low MIC (0.097 μg mL−1) against S. vitulinus. The antibiofilm activity of Ag0.75@ZFO NPs was the highest, recorded as 97.3% for S. aureus and 95.25% for Enterococcus columbae. In the case of UV exposure, bacterial growth reached the lowest grade. Finally, it was seen that the amount of cellular protein released from bacterial cells is directly proportional to the concentration of Ag0.75@ZFO NPs, which clearly explains the formation of pits in the cell membrane. The synthesized nanocomposites may find an application after mixing with operating room paints to reduce the harmful effect of pathogenic microbes and, therefore, eliminate bacterial contamination.

Despite significant improvements in the way breast cancer is managed and treated, it continues to persist as a leading cause of death worldwide. If detected and diagnosed early, when tumours are small and localised, there is a considerably higher chance of survival. However, current methods for detection and diagnosis lack the required sensitivity and specificity for identifying breast cancer at the asymptomatic or very early stages. Thus, there is a need to develop more rapid and reliable methods, capable of detecting disease earlier, for improved disease management and patient outcome. Raman spectroscopy is a non-destructive analytical technique that can rapidly provide highly specific information on the biochemical composition and molecular structure of samples. In cancer, it has the capacity to probe very early biochemical changes that accompany malignant transformation, even prior to the onset of morphological changes, to produce a fingerprint of disease. This review explores the application of Raman spectroscopy in breast cancer, including discussion on its capabilities in analysing both ex-vivo tissue and liquid biopsy samples, and its potential in vivo applications. In this article a new approach to the distinction between serial/contingent and parallel/independent processing in the human cognitive system is applied to semantic knowledge retrieval and phonological encoding of the word form in picture naming. In two-choice go/nogo tasks pictures of objects were manually classified on the basis of semantic and phonological information. An additional manipulation of the duration of the faster and presumably mediating process (semantic retrieval) allowed to derive differential predictions from the two alternative models. These predictions were tested with two event-related brain potentials (ERPs), the lateralized readiness potential (LRP) and the N200. The findings indicate that phonological encoding can proceed in parallel to the retrieval of semantic features. A suggestion is made how to accommodate these findings with models of speech production.  相似文献
