Significance of X granules in histiocytosis X: an ultrastructural study

Histiocytosis X is characterized by the presence of cytoplasmic rod structures called Langerhans' cell granules or X granules (XG). It has been speculated that histiocytosis X is a Langerhans' cell disorder. This ultrastructural study was performed to quantitate the number of XG containing histiocytes in the histiocytosis X lesions. Twenty-four specimens from 22 patients with histiocytosis X were studied: 4 from skin, 5 from lymph node, 11 from bone, 2 from lung, 1 from gingiva, and 1 from cheek. The majority of the histiocytes in histiocytosis X lesions do not contain X granules. The majority of the histiocytes in histiocytosis X lesions do not contain X granules. The percentage of histiocytes with XG in a lesion has no relation to the age of the patient or the organ from which it was obtained, except for skin, where they were quite numerous. The relative percent-age of histiocytes with granules does not correlate significantly with the prognosis of these patients.     

The distribution of substance P, enkephalin, and serotonin immunoreactivities in the area postrema of the rat and cat

With the use of the peroxidase-antiperoxidase technique the distribution of substance P (SP), enkephalin (ENK), and serotonin (5HT) immunoreactivities were described in the area postrema of the rat and cat. In both species, immunoreactivity in the area postrema was differentially distributed as either fibers only, or cell bodies and fibers. In the rat and cat, ENK had the greatest accumulation of immunoreactive fibers, followed by 5HT and SP. In the area postrema of the rat the majority of SP-, ENK-, and 5HT-immunoreactive fibers were along the ventral and ventrolateral borders, with fewer immunostained fibers at the dorsal surface. The area postrema of the cat had the majority of SP-, ENK-, and 5HT-immunoreactive fibers at the ventral and lateral borders, with fewer immunostained fibers at the dorsal and medial borders. In both species, the area postrema's central region contained the fewest immunostained fibers. In general, for each putative neurotransmitter examined, immunostained fibers in both species progressively decreased in number rostrocaudally. Serotonin- and ENK-immunoreactive cell bodies were found in the rat area postrema; in the cat area postrema only ENK-immunoreactive cell bodies were present. The area postrema of both species lacked SP-immunoreactive cell bodies. The heterogeneous distribution of immunoreactive fibers and cell bodies within the area postrema of the rat and cat may reflect the different functions of the area postrema in both species.     

Experience in the treatment of carcinoma of the cervix using a rotational technique

The radiation technique described is an unconventional method of treatment for carcinoma cervix patients and is essentially external beam therapy alone, using a ⁶⁰Cobalt rotation plan. This is in contrast to the more conventional series of 2 or 3 intracavitary radium insertions, either preceded and/or followed by fixed field external beam therapy. An advantage to the patient from this treatment scheme is the avoidance of the trauma associated with the repeated anaesthetics required for uterine and vaginal radium applications. Dosage levels have also been determined to ensure minimal post-radiation complications, and the 5- and 10-year survival rates for stage II and stage III cases are comparable with the survival results published by other centres. The series was treated during 1957-64 and consisted of all stage II and III cases referred to the Westminster Radiotherapy Department during this period, together with 13 stage I cases which were considered to be poor anaesthetic risks, and 4 stage IV cases. The 5- and 10-year survival rates for 69 stage II cases were 44% and 36% respectively, and for 81 stage III cases were 38% and 23% respectively.
