BACKGROUND: Small heat shock proteins are expressed in many tissues and are proposed to regulate actin filament dynamics when dissociated into small aggregates and phosphorylated in a p38 mitogen-activated protein kinase (p38MAPK)-dependent manner. METHODS: p38MAPK activity and small heat shock protein-25 (Hsp25) were determined in glomeruli from rats with experimental diabetes induced by streptozotocin administration and in isolated glomeruli exposed to a free radical stress. Contractile responsiveness of mesangial cells was determined by the serum-induced contraction of cell-embedded type I collagen gels. RESULTS: In experimental diabetes, there is an activation of p38MAPK, a decrease in the size of Hsp25 molecular aggregates, from large to small homo-oligomers, and an increase in the phosphorylation of Hsp25. In control glomeruli, a free radical stress, H2O2, activated p38MAPK and increased Hsp25 in a concentration-dependent manner. Additionally, H2O2 decreased the contractility of cultured mesangial cells concomitant with an increase in Hsp25 phosphorylation and a reduction in Hsp25 aggregate size. These effects were significantly reduced by SB202190, an imidazole-derivative cell-permeable inhibitor of p38MAPK. CONCLUSIONS: It has been proposed that the generation of oxygen-derived free radicals in diabetes may be linked causally to a loss of glomerular contractile reactivity and thus hyperfiltration in the early stages of diabetes mellitus. This study provides a mechanism for alteration of mesangial cell contractile responsiveness through phosphorylation of Hsp25 and may be a mechanism underlying abnormalities in glomerular hemodynamics in diabetes and in the presence of free radical stress.  相似文献

Our objective was to identify the determinants of plasma levels of anti-oxidant vitamins which have been linked with decreased risk of cancer and other chronic diseases. Correlation analyses were performed between baseline plasma levels of ascorbic acid, α- and β-carotenes, cryptoxanthin, lycopene and α- and γ-tocopherols and baseline information on dietary and other demographic and life-style factors among 1,364 subjects 35–69 years of age, who are participants in a chemoprevention trial on pre-cancerous lesions of the stomach in Venezuela. Males had lower levels of ascorbic acid, α- and β-carotene and cryptoxanthin and higher levels of α-tocopherol than females. This finding was confirmed in non-smokers and non-drinkers. In females, but not in males, age was positively associated with levels of ascorbic acid, cryptoxanthin, α- and β-carotene and γ-tocopherol. Male tobacco users had lower plasma levels of ascorbic acid, α- and β-carotene and cryptoxanthin than non-users, and regular alcohol drinkers had a decreased plasma levels of β-carotene compared with non-drinkers. Female tobacco users had lower levels of ascorbic acid and cryptoxanthin than non-users, and regular alcohol drinkers had lower levels of ascorbic acid and lycopene than non-drinkers. Frequencies of consumption of fresh fruits, fruit juice, raw vegetables and plantains showed weak positive associations with plasma levels of several vitamins studied in both sexes. Sex, age in females, tobacco and alcohol use and dietary consumption affected plasma anti-oxidant vitamin levels in this population significantly. These factors may influence the effect of anti-oxidant treatment in intervention trials. © 1996 Wiley-Liss, Inc.  相似文献

Objective

To examine the influence of superoxide on the severity of collagen‐induced arthritis (CIA) in mice.

Methods

CIA was induced in DBA/1J mice lacking the extracellular superoxide dismutase (EC‐SOD) gene (knockout [KO]) and in normal DBA/1J mice (wild‐type [WT]).

Results

The clinical disease activity score was significantly higher in EC‐SOD–KO mice than in WT mice between days 36 and 53, and the histologic scores for joint damage on day 53 increased 2‐fold or more in the EC‐SOD–KO mice. There were no significant differences between the 2 groups of mice in proliferation indices of spleen or lymph node cells in vitro after stimulation with type II collagen. Although both IgG1 and IgG2a anticollagen antibody levels increased in both groups of mice between days 21 and 53, there were no significant differences between the 2 groups. Lipopolysaccharide‐stimulated spleen cells from EC‐SOD–KO mice produced greater levels of tumor necrosis factor α (TNFα) over 48 hours in culture compared with cells from WT mice. Increased steady‐state levels of messenger RNA (mRNA) for interferon‐γ (IFNγ), TNFα, and interleukin‐1β (IL‐1β), and lower levels of IL‐1 receptor antagonist (IL‐1Ra) mRNA were present in the joints of the EC‐SOD–KO mice compared with the WT mice.

Conclusion

The absence of EC‐SOD leads to more severe CIA, which may be accompanied by enhanced production of the proinflammatory cytokines IFNγ, TNFα, and IL‐1β, and decreased production of the antiinflammatory cytokine IL‐1Ra in the joints.

The aim of this study was to assess whether nebulized budesonide may substitute for oral prednisolone in the management of children whose asthma is severe enough to warrant hospital admission, but who have no life threatening features. In a prospective, double-blind, randomized study nebulized budesonide (2 mg 8 hourly) was compared with oral prednisolone (2 mg/kg at entry and again at 24 h) in 46 children admitted to hospital with severe asthma exacerbations. Efficacy variables (including lung function measurements such as the primary outcome variable, Forced Expiratory Volume in 1 second (FEV1) and symptoms) were measured 24 h after treatment initiation. FEV1 improved significantly compared to baseline in patients who received nebulized budesonide compared to the prednislone group. The data show nebulized budesonide to be at least as effective as oral steroid in improving lung function and symptom severity in severe exacerbations of childhood asthma.  相似文献

Forty-three patients who had undergone direct-contrast ventriculography were submitted to intravenous digital subtraction ventriculography and first-pass radionuclide ventriculography to compare the left ventricular ejection fractions obtained by each method. Ejection fractions were calculated by the area-length method from the direct contrast ventriculograms, by both area-length and videodensitometric methods from the digital subtraction ventriculograms, and by count densitometry from the radionuclide ventriculograms. Satisfactory correlations were found between values obtained by the late mask resubtracted videodensitometric method and the radionuclide method (r = 0.85) and by the digital ventriculographic area length method and direct-contrast method (r = 0.88). Videodensitometric methods may be an alternative way to estimate left ventricular ejection fractions accurately without reliance on geometric assumptions about the shape of the left ventricular cavity.  相似文献

Ninety-eight newborn infants, less than 34 weeks at birth, were studied to examine the relationship between newborn hypotension and hypoxemia and brain damage. Heart rate, blood pressure and oxygen tension were recorded continuously during the 96 h following delivery. Outcome measures included neuropathology in children who died, and motor and cognitive development at one year corrected age in children who survived. There were 22 children with a minor and 27 with a major abnormal outcome. There was a relationship between newborn hypotension, newborn hypoxemia and low birth weight, and a major abnormal outcome. The probability of a major abnormal outcome increased from 8% in newborns with no hypotension or hypoxemia, to 53% in children with both hypotension and hypoxemia. These findings support the contention that combinations of sustained newborn hypotension and hypoxemia are important factors in the development of brain damage, accounting for a major abnormal outcome.  相似文献

BACKGROUND: Dietary alpha-linolenic acid (ALA) can be converted to long-chain n-3 polyunsaturated fatty acids (PUFAs) in humans and may reproduce some of the beneficial effects of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on cardiovascular disease risk factors. OBJECTIVE: This study aimed to compare the effects of increased dietary intakes of ALA and EPA+DHA on a range of atherogenic risk factors. DESIGN: This was a placebo-controlled, parallel study involving 150 moderately hyperlipidemic subjects randomly assigned to 1 of 5 interventions: 0.8 or 1.7 g EPA+DHA/d, 4.5 or 9.5 g ALA/d, or an n-6 PUFA control for 6 mo. Fatty acids were incorporated into 25 g of fat spread and 3 capsules to be consumed daily. RESULTS: The change in fasting or postprandial lipid, glucose, or insulin concentrations or in blood pressure was not significantly different after any of the n-3 PUFA interventions compared with the n-6 PUFA control. The mean (+/- SEM) change in fasting triacylglycerols after the 1.7-g/d EPA+DHA intervention (-7.7 +/- 4.99%) was significantly (P < 0.05) different from the change after the 9.5-g/d ALA intervention (10.9 +/- 4.5%). The ex vivo susceptibility of LDL to oxidation was higher after the 1.7-g/d EPA+DHA intervention than after the control and ALA interventions (P < 0.05). There was no significant change in plasma alpha-tocopherol concentrations or in whole plasma antioxidant status in any of the groups. CONCLUSION: At estimated biologically equivalent intakes, dietary ALA and EPA+DHA have different physiologic effects.  相似文献
