The free radical trapping nitrone compounds alpha-phenyl-N-tert-butylnitrone (PBN), 2-sulfophenyl-N-tert-butylnitrone (S-PBN) and disodium 2,4-disulfophenyl-N-tert-butyl nitrone (NXY-059) are effective neuroprotective agents in experimental models of both transient and permanent focal ischemia. A recent in vivo study suggested that NXY-059 had poor brain uptake in a transient ischemia model. We have now examined its blood-brain barrier permeability and cerebral endothelial uptake during hypoxic and ischemic conditions using an in vitro model of the blood-brain barrier. The in vitro blood-brain barrier permeability and cerebral endothelial uptake of NXY-059 and S-PBN were low during normoxic conditions. In contrast, PBN had very high blood-brain barrier penetration in vitro which confirmed earlier in vivo results. The permeability of [14C]NXY-059 increased 3.5 times after 9 h of hypoxia or 3 h of ischemia. There was, respectively, a 5-fold and more than 10-fold increase, after 6 and 9 h of ischemia. The control molecule [3H]inulin (M(r) approximately 5000) showed a similar increase in permeability under the same experimental conditions indicating a major change in the transport properties of the endothelium. There was a 60% reduction in the ATP levels of astrocytes after 3 h of ischemia and a 90% reduction after 9 h. The reduction in ATP levels in endothelial cells was somewhat lower. The uptake of NXY-059 in cerebral endothelial cells under normoxic, hypoxic or 9 h of ischemic conditions was negligible. NXY-059, S-PBN and PBN showed no effects on vesicular transport or the integrity of the blood-brain barrier in normoxic or ischemic conditions, nor did the compounds induce any change in the ATP levels of the cells. In conclusion, it is possible that the increase in blood-brain barrier permeability of [14C]NXY-059 which occurs during prolonged ischemia in vitro reflects a change which may be of importance to the neuroprotective effects of this nitrone free radical trapping agent.  相似文献

BACKGROUND: Adverse life events occurring early in development may alter the correct program of brain maturation and render the organism more vulnerable to psychiatric disorders. Identification of persistent changes associated with these events is crucial for the development of novel therapeutic strategies. METHODS: We used postnatal repeated maternal deprivation (MD) from postnatal day (PND) 2-14 to investigate changes in brain-derived neurotrophic factor (BDNF) levels. RNase protection assay and enzyme linked immunosorbent assay were employed to determine the anatomic profile of neurotrophin expression at different ages following MD. RESULTS: We found that MD produces a short-term up-regulation of neurotrophin expression in hippocampus and prefrontal cortex, as measured on PND 17, whereas at adulthood, a selective reduction of BDNF expression was observed in prefrontal cortex. When adult animals were challenged with a chronic swim stress paradigm, both a reduced expression of BDNF in prefrontal cortex and a significant reduction in striatal protein levels were found only in control subjects, whereas levels in the MD group were not further decreased. CONCLUSIONS: Our data suggest that MD produces a significant reduction of BDNF expression within prefrontal cortex and striatum, which may render these structures less plastic and more vulnerable under challenging conditions.  相似文献

Auditory response profiles for a group of ten healthy young and ten healthy elderly subjects, evoked by implicit memory and delayed verbal recognition tasks, were evaluated to determine if effects of stimulus repetition could be identified in the superior temporal gyrus (STG) and prefrontal cortical regions. We hypothesized that effects of stimulus repetition should occur both early in time and at early levels of the nervous system (STG) followed by later effects in prefrontal regions. Magnetoencephalographic (MEG) responses were recorded using a whole-head MEG system and automated, multi-start analysis methods were applied to the data in order to characterize the temporal response profiles from distributed but focal, cortical regions engaged in memory-related tasks. The findings revealed a main effect of age for early activity ( approximately 50 ms) in STG which appeared to be nonspecific for Old/New words and an Age x Task interaction for late activity ( approximately 100-800 ms) in STG which was specific to Old/New words. Although the behavioral performance measures did not reveal traditional effects of response priming, the MEG measures did reveal a reduction in amplitude with stimulus repetition in young subjects. The elderly did not reveal a reduction in amplitude concomitant with stimulus repetition for either the global attributes of words or for specific Old/New words. Long duration effects of stimulus repetition noted in the present study raise the possibility that results from sensory gating, mismatch negativity and P300 paradigms may represent a continuum of stimulus repetition effects. Two of these paradigms evoke greater enhancement to novel or infrequent stimuli, or rather, greater reduction of amplitude with repetition.  相似文献

The mammalian lignans enterolactone (ENL) and enterodiol, commonly found in human plasma and urine, are phytoestrogens that may contribute to the prevention of breast cancer and coronary heart disease. They are formed by the conversion of dietary precursors such as secoisolariciresinol and matairesinol lignans by the colonic microflora. The identification of lignins, cell-wall polymers structurally related to lignans, as precursors of mammalian lignans is reported here for the first time. In study 1, rats were fed rye or wheat bran (15% diet) for 5 d. Untreated brans and brans extracted with solvents to remove lignans were compared. ENL was estimated in urine samples collected for 24 h by time-resolved fluoroimmunoassay. ENL urinary excretion was reduced from 18.6 to 5.3 nmol/d (n=8; P<0.001) when lignans were removed from rye bran and from 30.5 to 6.2 nmol/d (P<0.001) when they were removed from wheat bran. These results suggest that lignins, embedded in the cell wall and retained in the bran during solvent extraction, account for 26-32% of the ENL formed from cereal brans. In study 2, rats were fed a deuterated synthetic lignin (0.2% diet) together with wheat bran (15%) for 3 d. The detection of deuterated ENL by LC-tandem MS in urine (20 nmol/d) clearly confirms the conversion of lignin into mammalian lignans. More research is warranted to determine the bioavailability of lignins in the human diet.  相似文献

This article describes the author's personal and professional dilemmas, encountered as a psychiatrist in the process of migration from Central Europe to New Zealand. The dilemmas include: (i) personal experiences in the stages of migration, (ii) struggles within the author's own multi-ethnic community, (iii) biculturalism, (iv) the unexpected seriousness of psychopathology, and (v) the tendency for overidentification. Professionals in a cultural transition should be aware that personal and professional challenges are interconnected. The concepts of loyalty, relational ethics and existential humanism were valuable in the resolution of these dilemmas.  相似文献
