This study was done to evaluate the effect of sperm source on chromatin integrity and ICSI outcomes. One hundred and thirteen samples containing epididymal aspirates of 57 obstructive azoospermic men and 56-ejaculated semen of normozoospermic men were included in this study. Sperm chromatin status was evaluated by Chromomycin A3 (CMA3), Aniline Blue (AB) and Toluidine Blue (TB). Fertilization rate and embryo quality were recorded. In epididymal group the percentage of sperms stained with AB, CMA3 and TB were significantly higher compared to ejaculate group while fertilization rate (60.6% vs. 74.04%) was significantly lower. However, embryo quality was not significantly different between two groups. In addition, abnormal sperm chromatin condensation and DNA fragmentation were not correlated with fertilization rate and embryo quality. Our results highlight the role of epididymis in sperm maturation and confirm that ICSI using ejaculated sperm is the gold standard for treatment of infertile men. 相似文献

Berberine is an isoquinoline alkaloid that has several pharmacological effects such as antiinflammatory, antimicrobial, apoptosis-inducing
and anticancer effects. It has been illustrated that the antiinflammatory effect is mediated by suppressing the nuclear factor-kappa
B (NF-κB) that activates expression of some antiinflammatory and antiapoptotic proteins including cyclooxygenase-2(COX-2),
inducible nitric oxide synthase (iNOS) and survivin; therefore, berberine may induce apoptosis by reducing antiinflammatory
and antiapoptotic agents, which suggest the relationship between antiinflammatory and apoptosis pathways. For further illustration
of the mechanism of berberine action, the human ductal breast epithelial tumor cell line (T47D cell line) was treated with
different concentrations of berberine (25–100 μM/ml). Berberine in 50 μM/ml had the most reducing effect on cell viability
and inducing of apoptosis. The level of COX-2, iNOS and survivin proteins decreased in berberine-treated cells; however, treatment
of the cells with aspirin and aminoguanidine (AG), COX-2 and iNOS inhibitors, respectively, showed that despite the cell growth-reducing
effect of aspirin, AG did not have a significant effect on cell viability. On the other hand, with the attention to reduction
in survivin protein level in berberine-treated cells, the results suggest that the apoptotic effect of berberine may be mediated
by reduction in both of the COX-2 and survivin in T47D cell line, while the iNOS does not play any effective role in berberine-induced

Nicotine has adverse cellular and molecular effects on oral mucosa, bone, and teeth. Vitamin E (α‐tocopherol) and vitamin C (ascorbic acid) are biological antioxidants with positive effects on wound healing and bone formation. This in vitro study sought to assess the cytotoxic effects of different concentrations of nicotine and cotinine (a metabolite of nicotine) on MG‐63 osteoblast‐like cells and human gingival fibroblasts (HGFs) in the presence and absence of antioxidant vitamins E and C (separately and combined). Cell viability and proliferation were assessed using the methyl thiazol tetrazolium (MTT) assay. Cell migration was assessed using the scratch test, and expression of apoptosis‐related genes was quantitatively analyzed using real‐time PCR. Dose‐dependent negative effects of nicotine on the morphology, viability, proliferation, and migration of MG‐63 and HGF cells were statistically significantly greater than those of cotinine. Vitamin E (separately and combined with vitamin C) was statistically significantly more effective than vitamin C (at the concentration used in this study) at improving cell viability, proliferation, and migration, and at reducing apoptosis of cells exposed to nicotine or cotinine. Based on the positive results of this study, vitamin C and especially vitamin E (systemically and/or locally) may be useful in the repair and regeneration of oral hard and soft tissues in smokers. 相似文献

Pain is an important component of disability problems, and plays a key role in mental health of older adults. This study aims to investigate the relationship between mental health and pain in older adults of Tehran, Iran. This was a cross-sectional study using data on 5326 older adults aged?≥60 years old from a large population-based survey (Urban HEART-2). A multistage cluster random sampling method was used to select the participants in Tehran, Iran, in 2011. General Health Questionnaire (GHQ-28), pain questionnaire, and socioeconomic questionnaires were used to collect the data. A total of 5326 older adults, 3811 (71.6%) married and 2797 (52.5%) female, were included into the study. The mean age of the participants was 68.92?±?7.02 years. Mean of GHQ-28 scores in the sample was 51.08?±?10.94, which indicates of a good level of mental health. The majority of the older adults had knee and back pain (more than 50%). Regardless of the chronicity, time, and the mode of reaction to it, knee and back pain were the highly reported pains among the participants. There was a statistically significant difference between two groups of older adults, with and without pain, in terms of GHQ-28 scores. Multiple regression analysis revealed that there was a relationship between mental health and the following factors: pain in head, shoulder, teeth, upper and lower limbs, education, gender, age, and marital status. Whatever the explanation, the relation of lowered health status to pain in all body parts among older people is considerable. This renders this matter as a top priority in health policy making. 相似文献

To determine the zinc status in patients with major β-thalassemia and its effect on their growth, the authors studied 64 thalassemic patients in comparison with 64 healthy matched individuals. Demographic and anthropometric data and history of the therapies were collected. Serum zinc level in both groups and ferritin in the thalassemic group were assigned. Interestingly, mean serum zinc level was significantly higher in the thalassemic group. No significant correlation between serum zinc level and short stature, serum ferritin level, desferrioxamine dose, initiating time of blood transfusion, and chelation therapy was found. The study indicates zinc deficiency in thalassemic patients who are on regular blood transfusion is rare and it seems that routine zinc supplementation is not necessary. 相似文献

During the COVID-19 pandemic, we should not forget about chronic, underlying and important diseases, especially diseases that cause immune system deficiency, of which TB is one and may be missed. Also, we should pay attention to the past medical history of the patients and their drug-drug interactions during the treatment period of COVID-19. Our main clinical message is that diseases such as TB, which weaken the immune system, may predispose a person to COVID-19 infection and COVID-19 may exacerbate TB and it’s mortality. On the other hand, diseases that target the lung tissue such as TB and COVID-19, may have synergistic effects and increase mortality (for a patient whose lung capacity is reduced due to TB, superimposed COVID-19 can worsen the situation).
