BB-10010 is a genetically engineered variant of human macrophage inflammatory protein-1 alpha with improved solution properties. We show here that it mobilizes stem cells into the peripheral blood. We investigated the mobilizing effects of BB-10010 on the numbers of circulating 8-day spleen colony-forming units (CFU-S8), CFU-S12, and progenitors with marrow repopulating ability (MRA). A single subcutaneous dose of BB-10010 caused a twofold increase in circulating numbers of CFU-S8, CFU-S12, and MRA 30 minutes after dosing. We also investigated the effects of granulocyte colony-stimulating factor (G- CSF) and the combination of G-CSF with BB-10010 on progenitor mobilization. Two days of G-CSF treatment increased circulating CFU-S8, CFU-S12, and MRA progenitors by 25.7-, 19.8-, and 27.7-fold. A single administration of BB-10010 after 2 days of G-CSF treatment increased circulating CFU-S8, CFU-S12, and MRA even further to 38-, 33-, and 100- fold. Splenectomy resulted in increased circulating progenitor numbers but did not change the pattern of mobilization. Two days of treatment with G-CSF then increased circulating CFU-S8, CFU-S12, and MRA by 64-, 69-, and 32-fold. A single BB-10010 administration after G-CSF treatment further increased them to 85-, 117-, and 140-fold, respectively, compared with control. We conclude that BB-10010 causes a rapid increase in the number of circulating hematopoietic progenitors and further enhances the numbers induced by pretreatment with G-CSF. BB- 10010 preferentially mobilized the more primitive progenitors with marrow repopulating activity, releasing four times the number achieved with G-CSF alone. Translated into a clinical setting, this improvement in progenitor cell mobilization may enhance the efficiency of harvest and the quality of grafts for peripheral blood stem cell transplantation. 相似文献

Charnley prostheses, retrieved at revision surgery, were studied to assess
the effects of friction on the total hip replacement procedure. Frictional
resistance was measured using the Durham hip function simulator under both
dry and lubricated conditions. The friction factor values (f) for the
explanted prostheses were found to have a non- Gaussian distribution with
medians of 0.13 [inter-quartile range (IQR) 0.10-0.16] and 0.06 (IQR
0.005-0.08) for dry and lubricated (n = 0.01 Pa s) regimes, respectively.
New Charnley prostheses had values of f equal to 0.11 +/- 0.025 and 0.04
+/- 0.01 under the same conditions, and showed no large deviation from a
Gaussian distribution. There was found to be a statistically significant
difference in the medians of the friction factors for new and retrieved
prostheses in the lubricated regime. Ingression of cement into the worn
region of the cup was found to increase the friction factor significantly
under dry conditions. There was no evidence of an increase in the friction
factor or torque for those joints that had a loose socket with respect to
those that were fixed at revision. A decrease in the frictional torque
against number of cycles undergone by the joint in vivo may indicate that a
fatigue-type process may have a role in the loosening of the socket.
However, this relationship was found not to be significant for friction
measured under lubricated conditions and it seems unlikely that the
frictional torque generated in this type of prosthesis will contribute
significantly to the long-term loosening of the socket.

This molecular epidemiologic case-control study of lung cancer incorporated
three complementary biomarkers: the glutathione S- transferase M1 (GSTM1)
null genotype, a potential marker of susceptibility, and polycyclic
aromatic hydrocarbon-DNA adducts (PAH- DNA) and sister chromatid exchanges
(SCE), both indicators of environmentally induced genetic damage.
Associations between biomarkers and lung cancer were investigated, as were
possible gene-environment interactions between the GSTM1 null genotype and
tobacco smoke exposure. Subjects included 136 primary non-small cell lung
cancer surgical patients and 115 controls at the Columbia Presbyterian
Medical Center. Questionnaire and Tumor Registry data, pre-treatment blood
samples and biomarker measurements on blood were obtained. Overall, GSTM1
null genotype was significantly associated with lung cancer [odds ratio
(OR) = 2.04, 95% confidence interval (CI) = 1.13-3.68]. ORs for GSTM1 and
lung cancer were significant in females (2.50, 1.09-5.72) and smokers
(2.25, 1.11-4.54) and not significant in males (1.4, 0.58-3.38) and
non-smokers (0.88, 0.18-4.33). However, ORs for males versus females and
smokers versus non-smokers did not differ significantly. The OR for GSTM1
and lung cancer in female smokers was 3.03 (1.09- 8.40), compared with 1.42
(0.53-4.06) in male smokers. In contrast to PAH-DNA adducts in leukocytes,
SCE did not differ between cases and controls. Neither biomarker differed
significantly between the two GSTM1 genotypes. The combined effect of
elevated PAH-DNA adducts and GSTM1 genotype on case-control status (16.19,
1.2-115) appeared multiplicative. Results suggest that the effect of the
GSTM1 null genotype is greatest in female smokers, which is consistent with
other evidence that indicates that women are at higher risk of lung cancer
than males, given equal smoking. Persons with both the GSTM1 deletion and
elevated PAH-DNA adducts may represent a sensitive subpopulation with
respect to carcinogens in tobacco smoke and other environmental media.

The ageing ovary appears to be characterized by depletion of primordial
follicles. The relationship between infertility and the number of follicles
in the ovarian cortex is not known. Moreover, there are no accurate markers
or clinical methods to predict the decline in ovarian reserve. This study
investigates the correlation between early follicular follicle stimulating
hormone, ovarian size and follicular density in 60 infertile women aged
19-45 years (mean = 34.4 +/- 5.5). An ovarian biopsy was taken from each
patient while performing diagnostic laparoscopy (n = 28) or laparotomy for
tubal surgery or myomectomy (n = 32). The median number of follicles was
similar in tubal and unexplained infertility patients (9.5 versus 5.5).
Increasing age showed a significant negative correlation with follicular
density and ovarian volume (r = -0.46, P = 0.0003;. r = -0.43, P = 0.0016,
respectively). In women > or = 35 years of age the ovarian volume showed
a strong correlation with follicular density (r = 0.71, P < 0.0001). Our
results indicate that infertile women in their late thirties and over have
a decreased ovarian reserve which could possibly be predicted by ovarian
volume measurement. Ovarian biopsy may have a place as part of infertility
evaluation in older women.

The effects of an ionic contrast agent, meglumine iothalamate (Conray-60), and two newer low-osmolality radiographic contrast media, sodium meglumine ioxaglate (Hexabrix) and iopamidol (B-15,000), on platelet aggregation and secretion responses were studied. All three agents inhibited platelet responses during stimulation with adenosine diphosphate (ADP), epinephrine, and collagen. Platelet function was inhibited by iothalamate at concentrations of 11 mg iodine/ml and above, and by the newer agents at concentrations above 30 mg iodine/ml. Addition of exogenous calcium decreased the iothalamate-induced inhibition of aggregation but did not improve dense granule secretion. There was no consistent effect of exogenous calcium on platelet inhibition by iopamidol and ioxaglate. These studies indicate that the newer agents inhibit platelet function less than iothalamate does, and that chelation of Ca2+ may not be the major mechanism of platelet inhibition by contrast agents. 相似文献

We assessed the potential of proton magnetic resonance (MR) imaging for accurately measuring left ventricular volumes using 15 latex casts of excised human left ventricles. The casts were submerged in water to stimulate the endocardial left ventricular cavity interface in in vivo imaging conditions. Tomographic image sections perpendicular to the long axis of the cast were obtained, spanning each cast from apex to base. Simpson's rule was used to calculate the cast volumes. Correlation between the actual cast volumes (as measured by the displacement method) and the calculated volumes using MR imaging for the 15 casts was excellent. Our data demonstrate that MR imaging accurately measures cardiac chamber volumes in this in vitro model. 相似文献
