Mammalian cardiomyocytes (CMs) maintain a low capacity for self-renewal in adulthood, therefore the induction of CMs cycle re-entry is an important approach to promote myocardial repair after injury. Recently, photobiomodulation (PBM) has been used to manipulate physiological activities of various tissues and organs by non-invasive means. Here, we demonstrate that conditioned PBM using light-emitting diodes with a wavelength of 630 nm (LED-Red) was capable of promoting the proliferation of neonatal CMs. Further studies showed that low-power LED-Red affected the expression of miR-877-3p and promoted the proliferation of CMs. In contrast, silencing of miR-877-3p partially abolished the pro-proliferative actions of LED-Red irradiation on CMs. Mechanistically, GADD45g was identified as a downstream target gene of miR-877-3p. Conditioned LED-Red irradiation also inhibited the expression of GADD45g in neonatal CMs. Moreover, GADD45g siRNA reversed the positive effect of LED-Red on the proliferation of neonatal CMs. Taken together, conditioned LED-Red irradiation increased miR-877-3p expression and promoted the proliferation of neonatal CMs by targeting GADD45g. This finding provides a new insight into the role of LED-Red irradiation in neonatal CMs biology and suggests its potential application in myocardial injury repair.  相似文献

BACKGROUNDHepatocellular carcinoma (HCC) exhibits high invasiveness and mortality rates, and the molecular mechanisms of HCC have gained increasing research interest. The abnormal DNA damage response has long been recognized as one of the important factors for tumor occurrence and development. Recent studies have shown the potential of the protein RING finger and WD repeat domain 3 (RFWD3) that positively regulates p53 stability in response to DNA damage as a therapeutic target in cancers. AIMTo investigate the relationship between HCC and RFWD3 in vitro and in vivo and explored the underlying molecular signalling transduction pathways. METHODS RFWD3 gene expression was analyzed in HCC tissues and adjacent normal tissues. Lentivirus was used to stably knockdown RFWD3 expression in HCC cell lines. After verifying the silencing efficiency, Celigo/cell cycle/apoptosis and MTT assays were used to evaluate cell proliferation and apoptosis. Subsequently, cell migration and invasion were assessed by wound healing and transwell assays. In addition, transduced cells were implanted subcutaneously and injected into the tail vein of nude mice to observe tumor growth and metastasis. Next, we used lentiviral-mediated rescue of RFWD3 shRNA to verify the phenotype. Finally, the microarray, ingenuity pathway analysis, and western blot analysis were used to analyze the regulatory network underlying HCC. RESULTSCompared with adjacent tissues, RFWD3 expression levels were significantly higher in clinical HCC tissues and correlated with tumor size and TNM stage (P < 0.05), which indicated a poor prognosis state. RFWD3 silencing in BEL-7404 and HCC-LM3 cells increased apoptosis, decreased growth, and inhibited the migration in shRNAi cells compared with those in shCtrl cells (P < 0.05). Furthermore, the in vitro results were supported by the findings of the in vivo experiments with the reduction of tumor cell invasion and migration. Moreover, the rescue of RFWD3 shRNAi resulted in the resumption of invasion and metastasis in HCC cell lines. Finally, gene expression profiling and subsequent experimental verification revealed that RFWD3 might influence the proliferation and metastasis of HCC via the Wnt/β-catenin signalling pathway.CONCLUSIONWe provide evidence for the expression and function of RFWD3 in HCC. RFWD3 affects the prognosis, proliferation, invasion, and metastasis of HCC by regulating the Wnt/β-catenin signalling pathway.  相似文献

目的 分析益肾补骨汤辅助经皮椎体成形术治疗老年骨质疏松性胸腰椎骨折患者的临床价值。方法 回顾性分析2019年2月—2021年2月收治的老年骨质疏松性胸腰椎骨折119例。根据治疗方案分为常规组57例和中药组62例。常规组给予经皮椎体成形术及常规治疗,中药组在常规组基础上给予益肾补骨汤。比较2组临床疗效、骨密度、术后疼痛、伤椎恢复情况、腰椎功能及再骨折与并发症发生率。结果 中药组总有效率高于常规组(P<0.05)。治疗后,中药组腰椎正位(L2~4)、右股骨颈骨密度高于常规组,视觉模拟评分法评分低于常规组(P<0.05)。治疗后,中药组伤椎楔变角及后凸Cobb角低于常规组,伤椎前缘高度高于常规组(P<0.05)。治疗后,中药组日本骨科协会评估治疗分数评分高于常规组,Oswestry功能障碍指数评分低于常规组(P<0.05)。中药组术后椎体再骨折发生率低于常规组(P<0.05)。结论 益肾补骨汤有利于减轻老年骨质疏松性胸腰椎骨折经皮椎体成形术后疼痛,降低并发症发生率,促进伤椎愈合和改善椎体功能。  相似文献

目的研究哮喘患者不同时期血清C反应蛋白(CRP)和肺功能测定的临床意义。方法 85例哮喘患者(哮喘组)和23例健康对照者(对照组)为研究对象,测定哮喘组急性发作期、慢性持续期、缓解期及对照组的血清CRP、血常规、肺功能及动脉血气。结果哮喘组急性发作期血清CRP、外周血白细胞计数(WBC)及动脉血二氧化碳分压(p(CO2))均明显高于慢性持续期、缓解期和对照组(P均<0.01);哮喘组急性发作期FEV1%、FEV1/FVC%及动脉血氧分压(p(O2))均明显低于慢性持续期、缓解期和对照组(P均<0.01);哮喘组慢性持续期、缓解期CRP水平均高于对照组(P均<0.05)。病情3,4级哮喘患者慢性持续期FEV1%、FEV1/FVC%较第1,2级明显下降(P<0.05或0.01)。结论血清CRP可作为判断哮喘患者病情的一个炎性反应指标,有助于哮喘的诊断与疗效观察。  相似文献

目的 :研究补骨脂对黑素细胞黏附和迁移的影响。方法 :用补骨脂处理黑素细胞 ,检测其黏附于牛血清纤维结合素和通过微孔滤膜的细胞数量并与生姜和未经药物处理的黑素细胞比较。结果 :补骨脂可显著促进黑素细胞黏附于牛血清纤维结合素和通过微孔滤膜并呈剂量依赖关系 ,但当补骨脂浓度超过 0.2mg·L-1时这种作用未见增加。补骨脂浓度在 0.01mg·L-1时 ,黑素细胞黏附并无显著增加 ,但对迁移的影响已可见统计学差异 (P <0.01)。生姜则不具有这样的作用。结论 :在体外 ,补骨脂能明显促进黑素细胞的黏附和迁移 ,这至少可以部分解释应用补骨脂治疗白癜风是有效的。  相似文献

Objective To establish a rapid and effective supercritical fluid extraction(SFE) and rapid resolution liquid chromatography method coupled with diode-array detector(RRLC-DAD) to quantify the chromones in a species. Methods The effects of four parameters including ethanol concentration(50%-90%), pressure(25-45 MPa), temperature(40-60 ℃), and time(30-90 min) on the chromones yields, namely prim-O-glucosylcimifugin, cimifugin, 5-O-methylvisammioside, and sec-O-glucosylhamaudol, were investigated using SFE system with orthogonal array design(OAD). Furthermore, the extracts were analyzed using rapid resolution liquid chromatography coupled with diode-array detector(RRLC-DAD) system to confirm the results. Results Under the optimized conditions, i.e., 35 MPa of pressure, 60 ℃ of temperature, 70% ethanol, and 60 min of time, the yields of prim-O-glucosylcimifugin, cimifugin, 5-O-methylvisammioside, sec-O-glucosylhamaudol, and total chromones were 3.514, 0.132, 6.242, 0.342, and 10.231 mg/g, respectively. In comparison with ultrasonic assisted extraction(UAE), SFE was able to yield a 20.7% increase in the total chromones from Saposhnikoviae Radix. Conclusion SFE is an alternative and promising method to extract chromones from this species, and the established RRLC-DAD method could serve as a rapid and effective method for the identification of chromones from Saposhnikoviae Radix.  相似文献
