广西壮族人群白细胞介素-1α与β的基因多态性分布   总被引:2,自引:0,他引:2  
目的:了解白细胞介素-1α(IL-1α)基因-889位点和IL-1β基因-511位点多态性在广西壮族健康人群中的分布。方法:采用PCR-RFLP方法,对上述两位点进行了检测,计算其基因型和等位基因频率。结果:II-1α(-889)和IL-1β(-511)等位基因C、T频率在壮族正常人群中分别为93.9%、6.1%和53.4%、46.6%;与奥地利、非洲黑人、非洲白人健康人群相比,IL-1α(-889)基因型分布及等位基因频率均存在高度显著性差异;与奥地利、非洲白人健康人群相比,IL-1β(-511)基因型分布及等位基因频率存在显著性差异。结论:广西壮族健康人群IL-1α(-889)基因多态性分布与奥地利、非洲黑人、非洲白人不同;IL-1β(-511)基因多态性分布与奥地利、非洲白人不同。  相似文献

目的:探讨贫铀污染对大鼠皮层及海马结构SS阳性神经元的影响。方法:采用不同剂量贫铀颗粒气管灌注大鼠3个月后,观察大鼠皮层及海马结构各区SS阳性神经元形态和数目的变化情况。结果:DU1mg组、DU3mg组和DU5mg组大鼠皮层SS阳性神经元染色较浅,阳性神经元数目与NS组比较均明显减少。海马结构各区DU3mg组和DU5mg组SS阳性神经元染色较浅,阳性神经元数目与NS组比较也明显减少。结论:贫铀污染能引起大鼠皮层和海马结构SS阳性神经元数目减少,并可能通过此途径引起脑功能障碍。  相似文献

目的 :已知电针能减轻大鼠辐射热引起的疼痛和急、慢性炎症痛 ,本文探讨电针能否减轻大鼠神经源性痛。方法 :将大鼠右侧L5/L6 脊神经结扎 ,用引起 5 0 %缩足的机械刺激阈值评价机械性痛觉超敏 ,用大鼠 5min内在 5± 1℃冷板上的缩足次数反映冷诱发的持续性疼痛。用韩氏穴位神经刺激仪给予电刺激 ,波宽 0 .6ms ,频率 2Hz ,电流强度为 0 .5 ,1和 2mA ,各 10min ,共刺激 30min。结果 :2Hz电针能减轻痛觉超敏 (持续 1h)和冷诱发的持续性疼痛 (持续 12小时 )。电针前15min皮下注射纳洛酮 (1mg/kg)能阻断电针对冷诱发的持续性疼痛的作用 ,但不能阻断对痛觉超敏的作用。结论 :低频 (2Hz)电针可能通过阿片机制减轻持续性的神经源性疼痛 ,低频电针也能减轻机械性痛觉超敏 ,该作用可能不是通过阿片机制完成的。  相似文献

本文采用电子顺磁波谱技术(Electron Spin Resonance,ESR),测定了20例胃癌组织中的自由基浓度.结果,癌浸润至浅肌层的病例为16×10~(10)spins/mg,比对照组(5.6×10~(10)spins/mg)明显升高,P<0.01.癌浸润深肌层以远时为 2.5×10~(10)spins/mg,比对照组明显降低,P<0.05.三组间方差分析,P<0.01.无淋巴结转移为14.9×10~(10)spins/mg,比有淋巴结转移(4.85×10~(10)spins/mg)升高,P<0.01.癌灶较小(<40.0mm)时为15.7×10~(10)spins/mg,小癌灶比大癌灶(>40.0mm)(2.9×10~(10)spins/mg)升高,P<0.01.高分化癌为10.50×10~(10)spins/mg,低分化癌为4.4×10~(10)spins/mg,P>0.05.自由基浓度在不同病理类型癌之间的差别,无统计学意义.  相似文献

Although the cholecystokinin (CCK-8) is reported to antagonize the κ-opioid-receptor-mediated analgesic effect in spinal cord, its mechanism and sites of action remain obscure. In the present study, the whole-cell patch-clamp recording technique was employed to examine the effect of κ-opioid agonist U50488H on voltage-gated calcium channels and the interaction between the CCK-8 and U50488H in acutely isolated rat dorsal root ganglion neurons. The results indicate that the calcium currents elicited in dorsal root antiopioid peptide CCK-8. The effect of the CCK-8 can be abolished by the CCK-B receptor antagonist, L365, 260. While CCK-8 showed a potent opioid-reversal effect, it by itself exerted a slight inhibitory effects on calcium current. This novel observation in the dorsal root ganglion neurons indicates that CCK-8 can antagonize the κ-opioid-receptor-mediated depressant effect on voltage-gated calcium current, and this antagonizing effect appears to be mediated via CCK-B receptor.  相似文献

目的:观察携带胶质细胞源性神经营养因子(glialcellinederivedneurotrophicfactor,GDNF)基因的重组腺病毒(AdCMVgdnf)对损伤运动神经元的保护作用。方法:采用培养运动神经元的谷氨酸损伤模型,评价GDNF重组腺病毒对损伤运动神经元的影响,指标是运动神经元计数。结果:(1)建立了运动神经元谷氨酸损伤模型:当培养液中加入0.5mmol·L-1的谷氨酸,作用24~48h后,运动神经元数目较对照组显著减少(P<0.01);(2)携带GDNF基因的重组腺病毒可显著减轻谷氨酸导致的运动神经元损伤。结论:腺病毒介导的GDNF表达对损伤的运动神经元具有保护作用。  相似文献

Thallium is a rare-earth element, but widely distributed in water environments, posing a potential risk to our health. This study was designed to investigate the chronic effects of thallium based on physiological responses, gene expression, and changes in the activity of relevant enzymes in adult zebra fish exposed to thallium at low doses. The endpoints assessed include mRNA expression of metallothionein (MT)2 and heat shock protein HSP70; enzymatic activities of superoxide dismutase (SOD) and Na+/K+-ATPase; and the histopathology of gill, gonad, and liver tissues. The results showed significant increases in HSP70 mRNA expression following exposure to 100 ng/L thallium and in MT2 expression following exposure to 500 ng/L thallium. Significantly higher activities were observed for SOD in liver and Na+/K+-ATPase activity in gill in zebra fish exposed to thallium (20 and 100 ng/L, respectively) in comparison to control fish. Gill, liver, and gonad tissues displayed different degrees of damage. The overall results imply that thallium may cause toxicity to zebra fish at environmentally relevant aqueous concentrations.  相似文献

本文采用视觉模拟评分法,选择大剂量静脉芬太尼复合吸入麻醉、体外循环下心内直视手术后病人,自拟除气管导管开始至拔管后6天,对其疼痛、焦虑等进行调查。结果表明,疼痛以切口主,咳嗽及活动时加重;疼痛强度在拔除气管导管后12 ̄36小时显著高于平均水平(P〈0.0036,t检验);疼痛程度与焦虑相关(r=0.56咳动/0.57安静,P〈0.05)。聚在分析的结果提示,成人心血管外科手术后,约55%的病例疼痛  相似文献

Hematopoietic stem/progenitor cells (HSPCs) possess the potentials of self-renewal, proliferation, and differentiation toward different lineages of blood cells. These cells not only play a primordial role in hematopoietic development but also have important clinical application. Characterization of the gene expression profile in CD34+ HSPCs may lead to a better understanding of the regulation of normal and pathological hematopoiesis. In the present work, genes expressed in human umbilical cord blood CD34+ cells were catalogued by partially sequencing a large amount of cDNA clones [or expressed sequence tags (ESTs)] and analyzing these sequences with the tools of bioinformatics. Among 9,866 ESTs thus obtained, 4,697 (47.6%) showed identity to known genes in the GenBank database, 2,603 (26.4%) matched to the ESTs previously deposited in a public domain database, 1,415 (14.3%) were previously undescribed ESTs, and the remaining 1,151 (11.7%) were mitochondrial DNA, ribosomal RNA, or repetitive (Alu or L1) sequences. Integration of ESTs of known genes generated a profile including 855 genes that could be divided into different categories according to their functions. Some (8.2%) of the genes in this profile were considered related to early hematopoiesis. The possible function of ESTs corresponding to so far unknown genes were approached by means of homology and functional motif searches. Moreover, attempts were made to generate libraries enriched for full-length cDNAs, to better explore the genes in HSPCs. Nearly 60% of the cDNA clones of mRNA under 2 kb in our libraries had 5′ ends upstream of the first ATG codon of the ORF. With this satisfactory result, we have developed an efficient working system that allowed fast sequencing of 32 full-length cDNAs, 16 of them being mapped to the chromosomes with radiation hybrid panels. This work may lay a basis for the further research on the molecular network of hematopoietic regulation.  相似文献
