Glycerophosphodiester phosphodiesterase (GDPD) is a glycerol metabolizing enzyme that has been previously identified in Escherichia coli, Haemophilus influenzae, Bacillus subtilis and Borrelia hermsii. It has been also reported that there is putative protein containing GDPD domain in Mus musculus, but not in Homo sapiens. SMART analysis showed that our predicted protein and three others of the same gene also comprise this domain. Because the gene has not been named, we termed it GDPD1 and accordingly termed our splice variant GDPD1_v1 after acquiring the agreement of HUGO. RT-PCR experiment revealed that our splice variant of the gene is mainly expressed in human ovary and small intestine, basically conforming to the result of serial analysis of gene expression.  相似文献

目的 检测子痫前期患者和正常孕妇胎盘组织中血管细胞黏附分子-1(VCAM-1)、细胞间黏附分子-1(ICAM-1)及血小板内皮细胞黏附分子-1(PECAM-1)的表达水平,以探讨细胞黏附分子(CAM)在子痫前期发病机制中的作用.方法 选取2005年5月-2006年5月在郑州大学第三附属医院住院分娩病例80例,分为3组:①轻度子痫前期组20例;②重度子痫前期组30例;③正常晚孕组(NT组)30例;胎盘组织中MCAM-1、ICAM-1及PECAM-1的表达水平采用免疫组化PV-9000法测定.结果 ①重度子痫前期组胎盘组织中VCAM-1和PECAM-1的表达下降(P<0.05),ICAM-1的表达增强(P<0.05).②轻度子痫前期组胎盘组织中VCAM-1的表达下降(P<0.05).ICAM-1和PECAM-1的表达无明显差异(P<0.05).结论 CAM与子痫前期发病有密切联系,VCAM-1、ICAM-1和PECAM-1参入了子痫前期病理生理过程且与子痫前期的病情程度密切相关.  相似文献

Perfusion MRI has the potential to provide pathophysiological biomarkers for the evaluating, staging and therapy monitoring of prostate cancer. The objective of this study was to explore the feasibility of noninvasive arterial spin labeling (ASL) to detect prostate cancer in the peripheral zone and to investigate the correlation between the blood flow (BF) measured by ASL and the pharmacokinetic parameters Ktrans (forward volume transfer constant), kep (reverse reflux rate constant between extracellular space and plasma) and ve (the fractional volume of extracellular space per unit volume of tissue) measured by dynamic contrast‐enhanced (DCE) MRI in patients with prostate cancer. Forty‐three consecutive patients (ages ranging from 49 to 86 years, with a median age of 74 years) with pathologically confirmed prostate cancer were recruited. An ASL scan with four different inversion times (TI = 1000, 1200, 1400 and 1600 ms) and a DCE‐MRI scan were performed on a clinical 3.0 T GE scanner. BF, Ktrans, kep and ve maps were calculated. In order to determine whether the BF values in the cancerous area were statistically different from those in the noncancerous area, an independent t‐test was performed. Spearman's bivariate correlation was used to assess the relationship between BF and the pharmacokinetic parameters Ktrans, kep and ve. The mean BF values in the cancerous areas (97.1 ± 30.7, 114.7 ± 28.7, 102.3 ± 22.5, 91.2 ± 24.2 ml/100 g/min, respectively, for TI = 1000, 1200, 1400, 1600 ms) were significantly higher (p < 0.01 for all cases) than those in the noncancerous regions (35.8 ± 12.5, 42.2 ± 13.7, 53.5 ± 19.1, 48.5 ± 13.5 ml/100 g/min, respectively). Significant positive correlations (p < 0.01 for all cases) between BF and the pharmacokinetic parameters Ktrans, kep and ve were also observed for all four TI values (r = 0.671, 0.407, 0.666 for TI = 1000 ms; 0.713, 0.424, 0.698 for TI = 1200 ms; 0.604, 0.402, 0.595 for TI = 1400 ms; 0.605, 0.422, 0.548 for TI = 1600 ms). It can be seen that the quantitative ASL measurements show significant differences between cancerous and benign tissues, and exhibit strong to moderate correlations with the parameters obtained using DCE‐MRI. Although chemotherapeutic drugs are widely applied for clinic tumor treatment, severe toxicity restricts their therapeutic efficacy. In this study, we reported a new form of selenium, selenium nanoparticles (Nano Se) which have significant lower toxicity and acceptable bioavailability. We investigated Nano Se as chemotherapy preventive agent to protect against toxicities of anticancer drug irinotecan and synergistically enhance the anti-tumor treatment effect in vitro and in vivo. The underlying mechanisms were also investigated. The combination of Nano Se and irinotecan showed increased cytotoxic effect with HCT-8 tumor cells likely by p53 mediated apoptosis. Nano Se inhibited growth of HCT-8 tumor cells partially through caspases mediated apoptosis. In vivo experiment showed Nano Se at a dose of 4 mg/kg/day significantly alleviated adverse effects induced by irinotecan (60 mg/kg) treatment. Nano Se alone treatment did not induce any toxic manifestations. The combination of Nano Se and irinotecan dramatically inhibited tumor growth and significantly induced apoptosis of tumor cells in HCT-8 cells xenografted tumor. Tumor inhibition rate was about 17.2%, 48.6% and 62.1% for Nano Se, irinotecan and the combination of Nano Se and irinotecan, respectively. The beneficial effects of Nano Se for tumor therapy were mainly ascribed to selectively regulating Nrf2-ARE (antioxidant responsive elements) pathway in tumor tissues and normal tissues. Our results suggest Nano Se is a promising selenium species with potential application in cancer treatment.  相似文献

Background Quantitative dynamic contrast enhancement MR imaging (DCE-MRI),used to measure properties of tissue microvasculature and tumor angiogenesis,is a promising method for distinguishing benign and malignant tumors and characterizing tumor response to antiangiogenic treatment.The aim of this study was to assess the feasibility of quantitative parameters derived from clinically used DCE-MRI for distinguishing benign from malignant tumors in the sinonasal area,which may be potentially useful for prediction and monitoring of treatment response to chemoradiotherapy of sinonasal tumors.Methods One hundred and forty-three patients with sinonasal tumors,including 78 malignant tumors and 65 benign tumors and tumor-like lesions,underwent clinically used DCE-MRI.Parametric maps were obtained for quantitative parameters including Ktrans,kep and ve.Two radiologists reviewed these maps and measured Ktrans,kep and ve in the tumor tissue.Data were analyzed using independent T-test or Mann-Whitney U test analysis and receiver operating characteristic curves.Results Ktrans,kep and ve showed significant differences between benign and malignant tumors in the sinonasal area (P=-0.000 1).The accuracy of Ktrans,kep and ve in differentiation between benign and malignant sinonasal tumors were 72.0%,76.2% and 67.1%,respectively.There were significant differences in kep and ve between malignant epithelial sinonasal tumors and lymphomas (P <0.05).Using a ve value of 0.213 as the threshold value differentiated malignant epithelial tumors from lymphomas with an accuracy of 78.3%,sensitivity of 88.2%,specificity of 68.0%,positive predictive value of 66.7%,and negative predictive value of 90.9%.However,no significant difference in Ktrans and kep was found between malignant epithelial and non-epithelial tumors in the sinonasal area (P >0.05).Conclusions It is feasible that quantitative parameters of tumors can be derived from clinically used DCE-MRI in the sinonasal region.Preliminary findings suggest an  相似文献

目的:对新疆假龙胆全草进行化学成分初步研究,并对分离得到酮类化类物进行定量分析。方法:采用75%乙醇回流法对新疆假龙胆全草进行提取,粗提物经石油醚、乙酸乙酯、正丁醇依次梯度萃取制备不同极性部分,将石油醚部分和乙酸乙酯部分浸膏分别用硅胶柱层析和葡聚糖凝胶LH-20柱层析分离纯化,所得单体化合物经EI-MS、~1H-NMR、~(13)C-NMR等光谱法进行结构鉴定;建立高效液相测定方法测定新疆假龙胆中3个酮类化合物的含量。结果:从新疆假龙胆中分离得到6个化合物,分别为1,8-二羟基-3,5-二甲氧基酮(1)、1-羟基-3,5,8-三甲氧基酮(2)、1-羟基-3,5-二甲氧基酮(3)、木犀草素(4)、胡萝卜素(5)和β-谷甾醇(6);含量测定方法学结果表明1,8-二羟基-3,5-二甲氧基酮、1-羟基-3,5,8-三甲氧基酮和1-羟基-3,5-二甲氧基酮3个酮化合物分离度较好,分别在35.7~713.4、32~1280、34.3~686 ng时与峰面积呈良好的线性关系,精密度试验RSD分别为0.31%、0.27%、0.25%,稳定性实验的RSD分别为1.05%、1.33%、1.58%,重复性实验的RSD分别为2.48%、0.82%、2.63%,加样回收率分别为103.1%(RSD=1.25%)、100.02%(RSD=0.93%),95.10%(RSD=2.62%);1,8-二羟基-3,5-二甲氧基酮等3个化合物的含量范围分别为0.21%~0.28%、0.65%~0.77%、0.19%~0.15%。结论:新疆假龙胆含有黄酮类、酮类和甾醇类化合物;经方法学考察,本研究建立的高效液相测定方法适合新疆假龙胆中1,8-二羟基-3,5-二甲氧基酮等3个化合物的测定;新疆假龙胆中1,8-二羟基-3,5-二甲氧基酮等3个化合物的含量较高。  相似文献

探讨关节镜治疗膝骨关节炎(KOA)的临床疗效。对82例Kellgren-Lawrence分级Ⅲ级及以下的KOA患者采用回顾性分析。观察组行镜检冲洗清理术并辅以理疗及药物治疗,对照组仅行理疗及药物治疗。采用膝关节Lysholm评分进行疗效评价。43例患者行镜检冲洗清理术并辅以理疗及药物治疗;39例患者只行理疗及药物治疗。全部患者均获随访,随访时间24个月。至术后24个月时,观察组患者膝关节Lysholm评分由术前的40.7±8.7分增加到术后的74.9±4.3分;对照组由术前的41.5±9.2分增加到术后的71.1±4.4分。2组疗效比较有极显著性差异(P<0.01)。对于Kellgren-Lawrence分级Ⅲ级及以下的KOA患者,关节镜手术结合理疗及药物治疗在术后近期疗效优于单纯保守治疗。  相似文献
