化学发光免疫法检测粒细胞抗体IgG的研究

作者以ＡＢＥＩ－ＣｏＣｌ＿２－Ｈ＿２Ｏ＿２作为化学发光指示系统，用固相竞争法检测５８例白细胞减少症患者、４１例无白细胞减少症患者及３２例正常人等的粒细胞抗体ＩｇＧ。判断标准采用正常人血清化学发光强度指数法。实验结果表明，白细胞减少症患者血清中粒细胞抗体阳性率明显高于无白细胞减少症患者（Ｐ＜０．００５）。本法敏感性为４３．１％，特异性为９０．２％，准确性为６２．６％，是一种灵敏而准确的方法，能将粒细胞包被试管在室温下较长时间保存，以供临床监测粒细胞抗体，大大缩短检测时间。     

散发性病毒性脑炎免疫组化和原位杂交研究

２０例临床诊断为散发性病毒性脑炎的尸检和活检脑组织，通过单纯疱疹病毒多克隆抗体免疫组化和生物素标记单纯疱疹病毒ＤＮＡ探针原位杂交研究，５例诊断为单纯疱疹病毒脑炎。结果说明：单纯疱疹病毒是国内散发性病毒性脑炎的重要病原；免疫组化和原位杂交是诊断单纯疱疹脑炎的两种敏感方法。     

保肝解毒胶囊对雷公藤多甙片所致小鼠急性肝损伤保护作用的实验研究

目的：运用新建雷公藤多甙致小鼠急性肝损伤模型，探讨保肝解毒胶囊对该模型肝损伤保护作用的效果和机理。方法：连续5d给小鼠灌服保肝解毒胶囊制剂，而后以雷公藤多甙片造小鼠急性肝损伤模型，检测小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、还原型谷胱甘肽(GSH-Px)、超氧化物歧化酶(SOD)，肝匀浆中过氧化物(LPO)含量，并取肝组织作病理检查。结果：保肝解毒胶囊可以对抗肝损伤，其疗效和剂量有关；保肝解毒胶囊保护肝细胞的机理与对抗脂质过氧化反应有关。结论：保肝解毒胶囊具有保护肝脏的作用，     

原发性十二指肠腺癌影像诊断及手术切除相关影像征象的探讨

目的：探讨原发性十二指肠腺癌的影像表现及与手术切除相关征象。方法：对照手术回顾性分析17例经病理证实的十二指肠腺癌低张十二指肠造影(hypotonic duodenography，HD)和CT的表现。结果：HD显示息肉型9例，溃疡型7例，浸润型1例。CT均示腔内不规则肿块或肠壁增厚，3例示溃疡，胆系扩张10例，胰头浸润14例，血管浸润7例，淋巴结转移3例，肝转移2例。7例行根治术，10例行姑息术。结论：HD与CT结合可充分显示腔内病变，腔外浸润及转移。癌肿局限于腔内或仅侵犯胰头时可行根治术；侵犯周围重要血管，发生淋巴结、肠系膜及肝脏等转移时一般只能行姑息术。     

Chronic Kidney Isograft and Allograft Rejection

In this study antigen-independent factor in the pathogenesis of chronic rejection of organ transplants was examined.Kidney isografts and allografts were transplanted orthotopically into bilaterally nephroectomized rat recipients and studied functionaaly,morphologically and immunohistologically,at serial intervals up to 52 weeks after transplantation.Allograft recipients developed progressive proteinuria after 12 weeks,with gradual renal failure ultimately leading to death.At the same time,morphological changes,including progressive arteriosclerosis and glomerulosclerosis ,tubular atrophy and interstitial fibrosis,developed.Immunohistologically,macrophages infiltrated glomeruli during this period and cytokines became unregulated.Our results showed that antigen-independent functional and morphological changes occurred in long-tern kidney isografts and mimicked those appearing much earlier in allografts that reject chronically.Initial injury and extent of functioning renal mass is suggested to be important factor for such late changes.     

Influence of cell sources, stimulating agents, and incubation conditions on release of interleukin-1 from chicken macrophages

Experiments were conducted to determine the cell source, stimulating agents, and incubation conditions that maximize interleukin-1 (IL-1) release by chicken macrophages/monocytes. Thymocyte co-mitogen proliferation was used to assay IL-1 activity of conditioned or partially purified supernatants. Monolayers of a transformed chicken macrophage cell line, HD11, released greater amounts of IL-1 than adherent cells isolated from peripheral blood, peritoneal cavity, or spleen. E. coli endotoxin and heat-killed S. aureus induced greater release of IL-1 by HD11 and splenic macrophages than latex or a super induction protocol with mezerien. Blocking macrophage eicosanoid synthesis with indomethacin did not influence IL-1 release from HD11 macrophages. Removing low molecular weight compounds from conditioned supernatants by dialysis did not influence IL-1 activity. IL-1 release was increased by incubating macrophages at 42 C compared to 39 C. Thymocyte co-mitogenic activity of IL-1 was increased by incubating thymocytes at 42 C compared to 39 C. Species cross reactivity between chicken and mammalian IL-1 was also investigated. Chicken IL-1 had slight co-stimulation activity on murine thymocytes, but murine and human IL-1 were without activity on chicken thymocytes.
