Cholinergic neurons in the basal forebrain of aged female mice   总被引:3,自引:0,他引:3  
Aging is associated with at least down-regulation of several cellular functions and diminished responsiveness to internal and external signals, and possibly with direct cell death. Consequently, pharmacological manipulations may be less effective in aged than in young organisms. In the present study, we investigated whether the basal forebrain cholinergic neurons and the estrogen receptor alpha (ERalpha) which they contain respond to changes in estrogen availability in aged female mice. The mice were sham-operated, ovariectomized, or ovariectomized and treated with 17beta-estradiol at the age of 18 months. Three months later, the mice were perfused and brain sections were double immunostained for choline acetyltransferase (ChAT) and ERalpha. Cell counting with a stereological method revealed that changes in the estrogen level have no effect on the total number of ChAT-immunoreactive (ir) neurons in the basal forebrain. However, the percentage of ChAT-ir neurons containing ERalpha-ir was higher in the ovariectomized mice than in the sham-operated or estrogen-treated mice. This was specific for the medial septum and vertical diagonal band of Broca. The findings indicate that even at old age the ERalphas in cholinergic cells are able to respond to changes in estrogen levels, though in a region-specific manner. This is naturally important for studies aiming to develop therapies for the elderly.  相似文献

Abnormalities in regional left ventricular (LV) function in aortic stenosis (AS) have yet to be appropriately characterized. One-dimensional strain (epsilon) and strain rate imaging (SRI), new ultrasound (US) indices for quantifying regional wall deformation, might allow this. The aims of this study were 1. to define regional radial and longitudinal epsilon /SR in AS; 2. to establish if they are related to the severity of the disease; and 3. to determine if regional deformation is further altered by coexistent coronary artery disease (CAD). A total of 40 patients were studied: Group I with isolated AS (10 women, 10 men; mean age 66 years) and group II with AS and concomitant CAD (CAD/AS) (13 women, 7 men, mean age 68 years). Data were compared to 20 age-matched healthy people (N). Regional systolic maximal velocity/SR and end-systolic and maximal epsilon were measured. The maximal systolic velocity/SR in AS and CAD/AS patients were significantly reduced compared to N. The two patient groups could be further differentiated by end-systolic and maximal epsilon, which demonstrated a further reduction in both epsilon indices in CAD/AS (i.e., maximal radial epsilon 29.3%, AS; 23.7%, CAD/AS; 40.4%, N; AS and CAD/AS vs. N, AS vs. CAD/AS, p < 0.05). Indices of radial and longitudinal deformation correlated both with aortic valve area (AVA) and stroke volume (SV) (i.e., radial maximal epsilon and AVA, r = 0.77, p < 0.05). A significant correlation was also found between epsilon indices and the severity of left anterior descending (LAD) or circumflex artery (CX) coronary artery. Regional myocardial deformation in AS is abnormal. In the absence of CAD, the degree of abnormality correlates with aortic valve area (AVA). The severity of the disease was best expressed by changes in regional epsilon. In CAD/AS patients, there was a significant further reduction in end-systolic and maximal epsilon. These changes correlated with the severity of coronary narrowing in the subtending vessel.  相似文献

The early consequences of Helicobacter pylori infection and the role of bacterial virulence determinants in disease outcome remain to be established. The present study sought to measure the development of host inflammatory and immune responses and their relationship to the putative bacterial virulence factors cag pathogenicity island (cagPAI), vacA allele, and oipA in combination with bacterial colonization density in a feline model of the early stages of H. pylori infection. Gastric tissues obtained from infected and uninfected cats were evaluated for H. pylori ureB, cagPAI, vacA allele, and oipA and colonization density (urease, histology, and real-time PCR). Inflammation was assessed by measuring mRNA upregulation of gamma interferon (IFN-gamma), interleukin (IL)-1 alpha, IL-1 beta, IL-4, IL-6, IL-8, IL-10, and IL-12 p40 and histopathology. The mucosal immune response was characterized by morphometric analysis of lymphoid follicles and by differentiating lymphocyte populations with antibodies against surface markers. Infecting H. pylori strains were positive for vacAs1 but lacked cagPAI and an active oipA gene. Colonization density was uniform throughout the stomach. Upregulation of IFN-gamma, IL-1 alpha, IL-1 beta, and IL-8 and increased severity of inflammatory infiltrates and fibrosis were observed in infected cats. The median number and total area of lymphoid aggregates were 5 and 10 times greater, respectively, in the stomachs of infected than uninfected cats. Secondary lymphoid follicles in uninfected cats were rare and positive for BLA.36 and B220 but negative for CD3 and CD79 alpha, whereas in infected cats they were frequent and positive for BLA.36, CD79 alpha, and CD3 but negative for B220. Upregulation of IFN-gamma, IL-1 alpha, IL-1 beta, and IL-8 and marked hyperplasia of secondary lymphoid follicles are early consequences of H. pylori infection in cats. The response appears to be similar to that of infected people, particularly children, can develop independently of the pathogenicity factors cagPAI and oipA, and is not correlated with the degree of colonization density or urease activity.  相似文献

Finite element model of radiofrequency ablation (RFA) with cooled-tip probe in liver has been developed by employing COMSOL Multiphysics software. It describes coupled electric, thermal and sodium chloride solution infiltration flow phenomena taking place during ablation processes. Features of hydraulic capacity, saturation of the tissue by infiltration, and dependency of electrical conductivity on the damage integral of the tissue have been supplied to the model. RFA experiments have validated the model. Physical parameters describing hydraulic capacity and hydraulic conductivity in the tissue, as well as, the relation of electrical conductivity against the value of damage integral have been determined.  相似文献

Objectives

The use of prophylactic antibiotics in severe acute pancreatitis (SAP) is controversial. The aim of this study was to compare the effects of antibiotics administered as prophylaxis and as treatment on demand, respectively, in two prospective, non-randomized cohorts of patients.

Methods

The study population consisted of 210 patients treated for SAP. In Group 1 (n = 103), patients received prophylactic antibiotics (ciprofloxacin, metronidazole). In Group 2 (n = 107), patients were treated on demand. Ultrasound-guided drainage and/or surgical debridement of infected necrosis were performed when the presence of infected pancreatic necrosis was demonstrated. The primary endpoints were infectious complication rate, need for and timing of surgical interventions, incidence of nosocomial infections and mortality rate.

Results

Ultrasound-guided fine needle aspiration [in 18 (16.8%) vs. 13 (12.6%) patients; P = 0.714], ultrasound-guided drainage [in 15 (14.0%) vs. six (5.8%) patients; P = 0.065] and open surgical necrosectomy [in 10 (9.3%) vs. five (4.9%) patients; P = 0.206] were performed more frequently and earlier [at 16.6 ± 7.8 days vs. 17.2 ± 6.7 days (P = 0.723); at 19.5 ± 9.4 days vs. 24.5 ± 14.2 days (P = 0.498), and at 22.6 ± 13.5 days vs. 26.7 ± 18.1 days (P = 0.826), respectively] in Group 2 compared with Group 1. There were no significant differences between groups in mortality and duration of stay in the surgical ward or intensive care unit.

Conclusions

The results of this study support the suggestion that the use of prophylactic antibiotics does not affect mortality rate, but may decrease the need for interventional and surgical management, and lower the number of reoperations.  相似文献

The study investigates effect of the additive consisting of natural zeolite (clinoptilolite) and soda lime glass powder on the hydration, mechanical properties and porosity of Portland cement concrete. The effect of mineral additive on the technological, physical-mechanical properties and porosity of the mortar was investigated by increasing the content of natural zeolite and glass powder added to the mortar up to 20% by weight of cement in increments of 5% and different particles size of natural zeolite. The mixes with the best technological and mechanical properties were identified and further studies were conducted by replacing 10% and 15% of cement with natural zeolite and soda lime glass with an average grain size of 59.3 μm, 29.0 μm or 3.6 μm of zeolite, and 29.6 μm of glass powder. The hydration process and microstructure of hardened cement paste modified with the aforementioned mineral additives was analysed by microcalorimetry, X-ray diffraction tests and thermogravimetric analysis. The optimal composition of cement paste and particle size distribution of natural zeolite were determined to achieve the higher flexural and compressive strength and lower open porosity. The mixture of mineral additives has the highest effect in terms of flexural and compressive strength and open porosity when added at the proportion 75:15:10 (cement:natural zeolite:soda lime glass) and when zeolite with an average particle size of about 3.6 μm is used  相似文献
