人类白细胞抗原-G突变体cDNA克隆及在K562细胞上的表达 总被引:2,自引:0,他引:2
目的:克隆人类白细胞抗原-G(Human leukocyte antligen-G,HLA-G)突变体cDNA,并使其在HLA-I类阴性的K562细胞上获得稳定表达,为研究配-受体之间的识别机制奠定基础。方法:用RT-PCR方法从人子宫蜕膜组织扩增出HLA-GcDNA,得到全长HLA-GPCR产物后,用桥式PCR方法进行定点点突变,将突变的目的基因亚克隆于逆转录,将突变的目的基因亚克隆于逆转录mG-pLNCX表达载体,采用感染的方法将重组质粒转入K562细胞,最后经G418筛选及有限稀释,利用单克隆抗体W6/32进行FACS及mRNA检测,观察HLA-G突变体在靶细胞表面的表达。结果:HLA-G突变体分子在经mG-pLNCX转染的靶细胞表面获得稳定高表达。结论:成功构建了mG-pLNCX表达载体,并使HLA-G突变体分子在HLA-I类阴性的靶细胞K562细胞上获得稳定表达。 相似文献

The localization of the A-rafcellular oncogene on the mouse X chromosome has been determined using Xbal-restricted DNAs prepared from progeny of an interspecies backcross between the B6.CBA.R1 and the Spe/Pas mouse strains. This localization to the proximal part of the mouse X chromosome has been confirmed by the use of somatic cell hybrids, carrying partially deleted X chromosomes and suggests that the A-raf oncogene localizes to a region lying some 10–17 centimorgans proximal to the hypoxanthine phosphoribosyltransferase (Hprt) gene between the locus DXPas4and the locus DXPas7defined by the cross-reacting human X chromosome-specific probe DXS32 (M2C). This localization on the mouse X chromosome is compatible with the presence of the A-rafoncogene on the short arm of the human X chromosome between the centromere and Xp21. 相似文献

Summary Kinematic variables of the vertical jump (jumping height, jump phase durations and joint angles) were measured on 39 male
physical education students. In addition, kinetic parameters of the hip and knee extensors, and of the plantar flexors (maxima
voluntary force and its rate of development) were recorded on the same subjects, in isometric conditions. The results demonstrated
significant positive correlations between kinetic parameters of the active muscle groups and jumping height (r=0.217−0.464). The dominant effect on these correlations was due to the knee extensors. Correlations between these parameters
and the duration of the jump phases were much weaker. Correlation coefficients between kinetic parameters and limb angles
in the lowest body position showed that fast force production in one muscle group was related to a significant decrease in
the joint angles of distant body segments. Multiple correlation coefficients between leg extensor parameters and kinematic
variables (ranging between 0.256 for the duration of the counter-movement phase and 0.616 for jump height) suggested that
kinetic parameters could explain more than a quarter of the variability of this complex human movement. Therefore, the conclusion
was drawn that an extended set of measurements of the relevant musculo-skeletal system parameters could predict a considerable
amount of the variability of human movement. However, high correlation coefficients between the same kinetic parameters of

Rett syndrome is a severe neurodevelopmental disorder that arises from mutations in the X-linked MECP2 gene. It is almost exclusively seen in girls due to the predominant occurrence of the mutations on the paternal X-chromosome, and also the early postnatal lethal effect of the disease causing mutations in hemizygous boys. We identified a boy with features of classic Rett syndrome who is mosaic for the truncating MECP2 mutation R270X. Chromosome analysis showed normal karyotype. These results indicate that a MECP2 mutation associated with Rett syndrome in females could lead to a similar phenotype in males as a result of somatic mosaicism. 相似文献

Cadavers remain a principal teaching tool for anatomists and medical educators teaching gross anatomy. Infectious pathogens in cadavers that present particular risks include Mycobacterium tuberculosis, hepatitis B and C, the AIDS virus HIV, and prions that cause transmissible spongiform encephalopathies such as Creutzfeldt-Jakob disease (CJD) and Gerstmann-Straussler-Scheinker syndrome (GSS). It is often claimed that fixatives are effective in inactivation of these agents. Unfortunately cadavers, even though they are fixed, may still pose infection hazards to those who handle them. Specific safety precautions are necessary to avoid accidental disease transmission from cadavers before and during dissection and to decontaminate the local environment afterward. In this brief review, we describe the infectious pathogens that can be detected in cadavers and suggest safety guidelines for the protection of all who handle cadavers against infectious hazards. 相似文献

Adenosquamous carcinomas are rare malignant tumors of the pancreas and periampullary region. We present two cases of pancreatic and periampullary adenosquamous carcinoma with clinicopathologic, histo- and immunohistochemical findings. A 51-year-old and a 48-year-old man presented with right upper quadrant pain of three months duration and jaundice for two weeks. Both cases had an elevation of liver enzymes and CA 19-9. In the first case, computerized tomography showed a 3 cm-mass at the pancreatic head. In the second one, endoscopic retrograde choledocopancreatography (ERCP) revealed a mass at the ampulla Vateri localization. A pancreatoduodenectomy was performed for both cases. Gross pathologic examination displayed a solid, gray-white colored 4 cm-tumor at the pancreatic head and a solid, pink-white colored, 2 cm-tumor at the periampullary region bulging into the duodenal lumen. Microscopically, both tumors were composed of solid nests of squamous cells with pearl formation and mucin-containing glandular cells and diagnoses were adenosquamous carcinoma. Clinicopathologic evaluation of CDw75 antigen expression in patients with gastric carcinoma 总被引:5,自引:0,他引:5
Situated on mature B lymphocytes, CDw75 antigen is a sialylated carbohydrate epitope generated by the enzyme beta-galactosyl alpha-2,6-sialyltransferase. Although CDw75 antigen expression was found to be correlated with aggressive behaviour of tumour cells in gastric adenocarcinomas, its prognostic role still remains unknown. The objective of this study was to determine the value of CDw75 antigen expression as a marker of the metastatic potential and prognosis of gastric adenocarcinomas. CDw75 antigen expression was evaluated immunohistochemically in 64 tumours and their nodal metastases. The correlation was analysed between CDw75 antigen expression and selected clinicopathological variables, including survival. Positive staining was detected in 31 cases. Non-neoplastic gastric mucosa was consistently negative. CDw75 expression was correlated with larger tumour size (p<0.006), infiltrative growth pattern (p<0.044), advanced stage (p<0.0006), and positive lymph nodes (p<0.0003).The overall survival rate of patients with CDw75 expression was 28%, which was significantly worse than that of patients without CDw75 expression (53%) (p<0.0005). Multivariate analysis showed that CDw75 expression was an independent prognostic indicator, together with the growth pattern of the tumour. These results indicate that immunohistochemical detection of CDw75 antigen expression may be a good indicator of metastatic potential and of prognosis in patients with gastric carcinomas. 相似文献

目的:观察TCPVβ7.1基因转染前后正常人外周血淋巴细胞对乳腺癌细胞株杀伤活性的影响。方法:脂质体包裹PdWA3.1vβ7.1后转染健康人PBMC,流式细胞仪检测PdWA3.1Vβ7.1基因表达,改良MIT法检测TCRVβ7.1基因转染前后正常人外周血淋巴细胞对乳腺癌细胞株杀伤活性。结果:TCRVβ7.1基因转染可显著增加正常人PBMC该基因表达,转染前后正常人外周血淋巴细胞对乳腺癌细胞株杀伤活性有显著性差异。结论:用TCR基因修饰可明显提高正常人PBMC对乳腺癌细胞杀伤作用。 相似文献
