Silk fibroin has acquired increasing interest for biomedical applications, and namely for the fabrication of scaffolds for tissue engineering, because of its highly positive biological interaction and the possibility to adapt the material to several application requirements by adopting different fabrication methods, in order to make films, sponges, fibers, nets or gels with predictable degradation times. For tissue engineering, in most cases porous scaffolds are required, in some cases possibly in situ forming and therefore fabricated in mild body-compatible conditions. In this work, we present a novel one-step method for the preparation of silk fibroin foams starting from water solutions and using low-pressure nitrous oxide gas as foaming agent. This foaming technique allows preparing fibroin porous scaffolds with easily tunable porosity, in mild processing conditions with the use of a relatively inert foaming agent saturating a fibroin water solution, that could be occasionally injected through a thin needle in the implantation site where expansion and foaming would occur. Optimal foaming processing conditions have been investigated, and the prepared foams have been characterized with Fourier Transform Infrared Spectroscopy (FTIR) compressive mechanical and rheological properties measurements, and by scanning electron microscopy and microCT.  相似文献

Surface Enhanced Raman Scattering (SERS) active gold nanostars represent an opportunity in the field of bioimaging and drug delivery. The combination of gold surface chemical versatility with the possibility to tune the optical properties changing the nanoparticles shape constitutes a multimodal approach for the investigation of the behavior of these carriers inside living cells. In this work, SERS active star-shaped nanoparticles were functionalized with doxorubicin molecules and covered with immuno-mimetic thiolated polyethylene glycol (PEG). Doxorubicin-conjugate gold nanoparticles show an intense Raman enhancement, a good stability in physiological conditions, and a low cytotoxicity. The strong adsorption of the anticancer drug doxorubicin in close contact with the gold nanostars surface enables their use as SERS tag imaging probes in vivo. Upon laser irradiation of the nanoparticles, a strong SERS signal is generated by the doxorubicin molecules close to the nanostars surface, enabling the localization of the nanoparticles inside the cells. After long time irradiation, the SERS signal drops, indicating the thermally driven delivery of the drug inside the cell. Therefore, the combination of SERS and laser scanning confocal microscopy is a powerful technique for the real-time analysis of drug release in living cells.  相似文献

Rabies is a lethal infectious disease that causes 55,000 human deaths per year and is transmitted by various mammalian species, such as dogs and bats. The host immune response is essential for avoiding viral progression and promoting viral clearance. Cytokines and chemokines are crucial in the development of an immediate antiviral response; the rabies virus (RABV) attempts to evade this immune response. The virus''s capacity for evasion is correlated with its pathogenicity and the host''s inflammatory response, with highly pathogenic strains being the most efficient at hijacking the host''s defense mechanisms and thereby decreasing inflammation. The purpose of this study was to evaluate the expression of a set of cytokine and chemokine genes that are related to the immune response in the brains of mice inoculated intramuscularly or intracerebrally with two wild-type strains of RABV, one from dog and the other from vampire bat. The results demonstrated that the gene expression profile is intrinsic to the specific rabies variant. The prompt production of cytokines and chemokines seems to be more important than their levels of expression for surviving a rabies infection.  相似文献
