A new antisense oligonucleotide delivery system based on self-assembled ODN-PEG hybrid conjugate micelles.

The conjugate of antisense c-raf oligonucleotide (ODN) and poly(ethylene glycol) (PEG) was synthesized for intracellular ODN delivery. When combined with polyethylenimine (PEI), the ODN-PEG conjugate self-associated to form polyelectrolyte complex micelles in aqueous solution. The effective hydrodynamic diameter of the micelles was ca. 70 nm with a narrow size distribution. Flow cytometry analysis indicated that the cellular uptake of the micelles by A2780 cells was much higher than that of ODN alone. The micelles also showed a superior antiproliferative activity against ovarian cancer cells in vitro and in vivo.     

The characteristics of Hep-2 cell with multiple drug resistance induced by Taxol.

OBJECTIVE: To investigate the characteristics of Hep-2 cell with multidrug resistance (MDR) induced by Taxol. STUDY DESIGN: Hep-2 cells were exposed in stepwise escalating concentration of Taxol to develop the resistant cell line-Hep-2T. Cell cycle distribution, apoptosis, and rhodamine accumulation were studied through flow cytometry. The MDR1 and MRP1 genes were detected through real-time quantitative RT-PCR, and the corresponding proteins were detected through Western blotting. RESULTS: The drug resistance of Hep-2T cells to Taxol, doxorubicin, gemcitabine, 5-FU, and cisplatin all increased. The percentage of G0/G1 phase and the antiapoptosis ability increased significantly compared with Hep-2 cells. Both MDR1 and MRP1 also increased at gene and protein level, though MDR1 was more prominent. CONCLUSION: More emphasis should be laid on MDR1/Pgp, the non-Pgp substrate chemotherapeutic agents, and the changes of cell cycle distribution to prevent MDR induced by Taxol. SIGNIFICANCE: These findings may provide theoretical support for the reverse of MDR.     

Myoepitheliomas of the soft palate: helical CT findings in two patients.

We describe the enhancement patterns of myoepithelioma in two patients with a soft palate mass. In the first case, helical CT revealed a faintly enhancing mass. Histologically, the tumor was composed of plasmacytoid cells in a background of rich myxoid stroma. Immunostaining for CD34 showed scanty blood vessels. In the second case, helical CT revealed an intensely enhancing mass. Histologically, the mass was a cellular tumor with fibrous stroma. Immunostaining for CD34 also showed frequent blood vessels.     

Recapitulation of fast skeletal muscle development in zebrafish by transgenic expression of GFP under the mylz2 promoter.

A 1,934-bp muscle-specific promoter from the zebrafish mylz2 gene was isolated and characterized by transgenic analysis. By using a series of 5' promoter deletions linked to the green fluorescent protein (gfp) reporter gene, transient transgenic analysis indicated that the strength of promoter activity appeared to correlate to the number of muscle cis-elements in the promoter and that a minimal -77-bp region was sufficient for a relatively strong promoter activity in muscle cells. Stable transgenic lines were obtained from several mylz2-gfp constructs. GFP expression in the 1,934-bp promoter transgenic lines mimicked well the expression pattern of endogenous mylz2 mRNA in both somitic muscle and nonsomitic muscles, including fin, eye, jaw, and gill muscles. An identical pattern of GFP expression, although at a much lower level, was observed from a transgenic line with a shorter 871-bp promoter. Our observation indicates that there is no distinct cis-element for activation of mylz2 in different skeletal muscles. Furthermore, RNA encoding a dominant negative form of cAMP-dependent protein kinase A was injected into mylz2-gfp transgenic embryos and GFP expression was significantly reduced due to an expanded slow muscle development at the expense of GFP-expressing fast muscle. The mylz2-gfp transgene was also transferred into two zebrafish mutants, spadetail and chordino, and several novel phenotypes in muscle development in these mutants were discovered.     

脾脏肿块的超声诊断

目的 :探讨脾脏肿块的超声表现及鉴别要点。材料和方法 :回顾性分析 76例脾脏肿块患者的超声检查资料 ,分析脾脏肿块的大小、数目、边界、内部回声等多种超声表现。结果 :76例脾脏肿块中有 3例错构瘤 ,10例血管瘤 ,16例淋巴瘤 ,2例淋巴管瘤 ,4例血管肉瘤 ,11例脾转移瘤 ,16例脾囊肿 ,4例脾梗死 ,3例脾血肿 ,5例脾脓肿 ,2例付脾 ,其超声表现各具特征。结论 :超声检查可早期发现并区别脾脏肿块的囊、实性 ,借助彩色血流显像可进一步鉴别良、恶性病灶。     

短暂性脑缺血发作患者血浆中TXB2和PGF1α含量检测

本文用放射免疫法测定47例短暂性脑缺血发作(TIA)患者血浆中血栓素B_2(TXB_2)和6酮-前列腺F_(1α)(PGF_(1α))的含量。结果发现TIA患者血浆中TXB_2含量增高,PGF_(1α)降低;头颅CT或MRI示有小灶性梗塞者及TIA发作持续时间长于30min者TXB_2升高和PGF_(1α)降低更显著。治疗3个月后,血浆PGF_(1α)显著增高。TXB_2和PGF_(1α)在体内的失平衡是急性脑血管疾病发病的重要机理之一。     

扶正健脾汤配合环磷酰胺对荷瘤小鼠的实验研究

用环磷酰胺对荷瘤小鼠进行化疗同时配合扶正健脾汤进行治疗的疗效观察,证明扶正健脾汤具有减轻环磷酰胺的副作用,其表现为实验组小鼠的胸腺系数增大;脾内 IgG、IgM 阳性抗体形成细胞增多;肝内巨噬细胞的吞噬功能加强。提示扶正健脾汤具有拮抗环磷酰胺对骨髓、胸腺和脾脏的免疫抑制作用,为该方剂治疗肿瘤的临床应用提供实验依据。     

着色真菌蟾蜍分离株小鼠致病试验研究

目的：观察着色真菌蟾蜍分离株的致病性。方法：抽样取着色真菌蟾蜍分离株与标准人分离株对照，制成菌悬液，做小鼠腹腔接种，观察接种后小鼠的外观表现，做菌种逆培养和观察内脏病理变化。结果：蟾蜍分离株卡氏枝孢霉、疣状瓶霉、裴氏着色霉和甄氏外瓶霉与病人分离株一样，在接种28天后，均可从小鼠脏器逆培养出接种的菌种，并引起内脏病变，形成结核样肉芽肿，肉芽肿内均可查见棕黄色硬壳小体。而瓶霉和外瓶霉属菌种逆培养结果均阴性。结论：从野生蟾蜍分离出的卡氏枝孢霉、疣状瓶霉，裴氏着色霉和甄氏外瓶霉与病人分离株一样，对小鼠有致病性，故可能是人着色芽生菌病的致病菌。     

水稻亚种间杂种优势利用研究

本试验探讨了华南双季稻区利用亚种同杂种优势的特点和问题,结果表明:当前,两系亚种问杂种一代主要存在的问题是结实率低和谷草比低;籼粳交杂种易受环境条件的影响;每穗实粒数和单株有效穗数是决定单株产量的重要因子;每穗实粒数增加的关键在于结实率的提高,本文还就提高结实率和谷草比等问题提出了意见。
