The hypercoagulable state in patients with toxemia of pregnancy was investigated in comparison with normal pregnant women using new coagulation parameters, mainly thrombin-antithrombin III (TAT) complexes, alpha 2-antiplasmin-plasmin complexes (PIP), and D-dimer FDP. When the patients were categorized by the classification of the WHO Study Group (1985), significant increases of TAT complexes and alpha 2-PIP complexes with decreases of the ATIII level were observed in the groups with preeclampsia and severe gestational hypertensive disease as compared to normal pregnant women. A significant increase of D-dimer FDP was observed in a group with severe gestational hypertensive disease. Additionally, the relationship between clinical signs and the hypercoagulable state in the patients was analyzed using canonical correlation analysis as a multivariate analysis. The clinical signs and coagulation parameters had a significantly high correlation of lambda 1 = 0.7219, p less than 0.01. The results showed that clinical signs were associated with simultaneous coagulation abnormalities. The indices obtained from the results of canonical correlation analysis, which were called the clinical index and the coagulation index, should be useful in evaluating the efficacy of anticoagulation therapy.  相似文献

Monoclonal antibodies (moABs) MA54 and MA61, directed toward the O-linked mucin-type glycoprotein, have been established and showed highly specific reactivity with human ovarian cancer. Fetal intestinal and colonic mucosal cells expressed this antigen and meconium staining was also frequently positive. To investigate the characteristic of an epitopic carbohydrate recognized by these moABs, the reactivity of each moAB with meconium extract was monitored by solid-phase enzyme-linked immunosorbent assay with mono-, di-, and oligosaccharides. MA54 and MA61 react with meconium extract and the reactivities of these moABs are neuraminidase sensitive. Ovine submaxillary mucin had a strong inhibitory activity toward the reaction between meconium extract and MA54 as well as MA61, suggesting that these moABs recognize NeuAc 2-6GalNAc epitope in meconium. The second aim of this study is to investigate the possible application of moABs to diagnose ovarian cancer and to compare these levels with those of the CA125 antigen. While serum CA54/61 antigen levels were elevated in 44.4% of ovarian cancer cases and serum CA125 antigen levels were elevated in 86.7% of the same population, the use of both assays indicated a sensitivity of detection of 97.8% (44 of 45 patients) in the population studied.  相似文献

Unknown signals from the fetus are thought to be involved in the onset of parturition. We recently discovered that urinary trypsin inhibitor (UTI) from fetal urine inhibits uterine muscle contraction. Objectives: The aim of this research was to elucidate the mechanism of action of UTI in suppressing cervical maturation. Study design: Non-pregnant and pregnant rabbits pretreated with and without UTI suppositories containing 1000 U (400 μg) for 3 days were treated for 2 days with vaginal suppositories containing 100 ng of interleukin-8 (IL-8). Results: IL-8 induced softening and dilatation of the rabbit cervices. In contrast UTI inhibited IL-8 induced cervical softening and dilatation. Water content, collagen content, neutrophil counts, elastase activity and collagenase activity of the cervix were increased by IL-8, but they did not increase by IL-8 with UTI. Conclusion: These results suggest that UTI inhibits cervical maturation induced by IL-8.  相似文献

Plants can sense the direction of gravity and change the growth orientation of their organs. The molecular mechanisms of gravity sensing and signal transduction during gravitropism are not well known. We have isolated several shoot gravitropism (sgr) mutants of Arabidopsis. The sgr3-1 mutant exhibits a reduced gravitropic response in the inflorescence stems. In the inflorescence stems of Arabidopsis, gravity is sensed in endodermal cells that contain sedimentable amyloplasts. In sgr3-1, some amyloplasts in the endodermis failed to sediment in the direction of gravity. SGR3 encodes a syntaxin, AtVAM3, which had previously been cloned as a homologue of yeast Vam3p. AtVAM3 is localized to the prevacuolar compartment and vacuole and is suggested to function in vesicle transport to the vacuole. We have also cloned another soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE), ZIG/AtVTI11, a mutation that causes abnormal gravitropism. This mutant displayed an abnormal distribution of amyloplasts in the endodermal cells similar to that in sgr3-1. Endodermis-specific expression of SGR3 and ZIG by using the SCR promoter could complement the abnormal shoot gravitropism of each mutant. Protein-protein interaction between AtVAM3 and AtVTI11 in the endodermal cells was detected immunologically. The sgr3-1 mutation appeared to reduce the affinity of AtVAM3 for AtVTI11 or SYP5. These results suggest that vesicle transport to the prevacuolar compartment/vacuole in the endodermal cells, mediated by a specific SNARE complex containing AtVAM3 and AtVTI11, plays an important role in shoot gravitropism.  相似文献

Soy isoflavonoids have attracted much attention because of their estrogenic activity. To study the intestinal absorption of the isoflavonoids, we investigated the cellular uptake and metabolism of genistein and daidzein and their glucosides, genistin and daidzin, by Caco-2 cell monolayers as a model of the human intestinal epithelium. When Caco-2 monolayers were incubated with genistein or daidzein at 10 micromol/L from the apical (mucosal) side, aglycone was lost from the apical solution for 2.0 h (P < 0.05) and the glucuronide/sulfates appeared at the level of 1-2 micromol/L. In the basolateral (serosal) solution, both intact aglycones and their glucuronide/sulfates increased (P < 0.05) with time and reached approximately 20 and 15% of the initial dose, respectively. The transport of their glucosides, genistin and daidzin, through Caco-2 monolayers was less than one tenth that of the aglycones. The cellular metabolism of genistein was compared with quercetin, kaempferol, luteolin and apigenin. Only genistein aglycone was transported intact to the basolateral solution. Transport was greater (P < 0.05) than that of flavonoid aglycones and was without an appreciable decrease of transepithelial resistance. Radical scavenging activity was not related to the susceptibility to conjugation of flavonoids/isoflavonoids. Affinity to the liposomal membrane was increased in the order of genistin = daidzin < daidzein < genistein < flavonoid aglycones. These results strongly suggest that isoflavone aglycones are taken up into enterocytes more efficiently than their glucosides because of their moderate lipophilicity. Furthermore, they are generally transported to the basolateral side in intact form in contrast to flavonoids, probably due to their unique isoflavonoid structure.  相似文献

Right-handed participants performed the categorical and coordinate spatial relation judgments on stimuli presented to either the left visual field - right hemisphere (LVF-RH) or the right visual field - left hemisphere (RVF-LH). The stimulus patterns were formulated either by bright dots or contrast-balanced dots. When the stimuli were bright, an RVF-LH advantage was observed for the categorical task, whereas an LVF-RH advantage was observed for the coordinate task. When the stimuli were contrast balanced, the RVF-LH advantage was observed for the categorical task, but the LVF-RH advantage was eliminated for the coordinate task. Because the contrast-balanced dots are largely devoid by of low spatial frequency content, these results suggest that processing of low spatial frequency is responsible for the right hemisphere advantage for the coordinate spatial processing.  相似文献

A phase I study of gemcitabine (GEM) and docetaxel (TXT) combination chemotherapy was performed for unresectable non-small-cell lung cancer. Chemotherapy consisted of a fixed dose of GEM (1,000 mg/m2) on day 1, 8 and an escalated dose of TXT (50, 60, 70 mg/m2) on day 8 every 21 days, > or = 2 courses. Nine patients were entered (each dose level: 3 patients). Leukopenia, neutropenia, GOT increase, GPT increase, anorexia, fatigue, fever, and alopecia occurred, but no dose-limiting toxicity was found at any dose level and no MTD was reached. The recommended dose for the phase II study is GEM 1,000 mg/m2 and TXT 70 mg/m2 with consideration of application to outpatients and continuing courses.  相似文献

Various neurofibroma cell lines isolated from either dermal, plexiform, or diffuse neurofibromas were found to respond to human gamma interferon by decreasing proliferation rates in vitro. The cell number decreased to around 40-50% of controls (without gamma interferon) six days after the treatment. The cell lines showed dramatic inhibition of tritium-labeled thymidine uptake one day after the treatment with gamma interferon. Either 100 IU/ml or 1,000 IU/ml of gamma interferon resulted in the same range of inhibition. It is calculated that venous infusion of one vial of commercial recombinant gamma interferon (200 x 10(6) IU/ml) reaches more than 100 IU/ml in the peripheral blood, which means that it may be clinically useful. The cell lines also responded to gamma interferon by initiating expression of CD54. Immunological modulation of neurofibroma cell components by gamma interferon in vivo remains to be studied.  相似文献
