Estimation of aneuploidy for chromosomes 3, 7, 16, X and Y in spermatozoa from 10 normospermic men using fluorescence in-situ hybridization 总被引:3,自引:0,他引:3
Fluorescence in-situ hybridization (FISH) is a fast and efficient method of
estimating aneuploidy in human spermatozoa. In this study, we have
estimated baseline disomy frequencies in spermatozoa from a group of 10
normospermic men, using stringent scoring criteria. A triple- probe FISH
procedure was used for chromosomes 3, X and Y, while a double-probe FISH
method was used for chromosomes 7 and 16. A total of 101273 spermatozoa
were scored for chromosomes 3, X and Y, resulting in 97.83% haploidy (3X or
3Y), 0.39% disomy (33X, 33Y, 3XX, 3YY or 3XY) and 0.35% diploidy (33XX,
33YY or 33XY). A total of 100760 spermatozoa were scored for chromosomes 7
and 16, giving 98.9% haploidy (716), 0.11% disomy (7716 or 71616) and 0.27%
diploidy (771616). Disomy frequencies for individual chromosomes differed
(chromosome 3, 0.20%; chromosome 7, 0.05%, chromosome 16, 0.06%; X + Y,
0.19%). The frequency of disomy 3 was significantly higher than disomy 7 (P
= 0.019) and disomy 16 (P = 0.022), while the frequency of sex chromosome
disomy was significantly higher than disomy 7 (P = 0.0058) and disomy 16 (P
= 0.0067), but not disomy 3 (P = 0.73). The disomy and diploidy (0.27-
0.35%) estimates obtained for this normospermic population were generally
low and were similar to other recent reports.

In experimental autoimmune glomerulonephritis (EAG), a model of Goodpasture's disease, Wistar Kyoto (WKY) rats immunized with collagenase-solubilized glomerular basement membrane (GBM) or the recombinant NC1 domain of the α3 chain of type IV collagen [α3(IV)NC1] develop anti-GBM antibodies and focal necrotizing glomerulonephritis with crescent formation. However, Lewis (LEW) rats, which share the same major histocompatibility complex (MHC) haplotype, are resistant to EAG development. A genome-wide linkage analysis of backcrossed animals with EAG revealed a major quantitative trait locus (QTL) on rat chromosome 13 (LOD = 3.9) linked to the percentage of glomerular crescents. To investigate the role of this QTL in EAG induction, reciprocal congenic rats were generated (LEW.WCrgn1 congenic and WKY.LCrgn1 congenic), immunized with recombinant rat α3(IV)NC1, and assessed for EAG development. WKY.LCrgn1 rats showed a marked reduction in albuminuria, severity of crescentic nephritis, and number of glomerular macrophages compared with WKY controls. No reduction in antibody levels was observed. However, LEW.WCrgn1 rats were resistant to EAG development, as were LEW controls. Macrophage activation in vitro was assessed in parental and congenic rat bone marrow-derived macrophages (BMDMs). WKY.LCrgn1 BMDMs showed a significant reduction in Fc receptor-mediated oxidative burst, phagocytosis of opsonised polystyrene beads, and LPS-induced levels of MCP-1 secretion and iNOS mRNA expression compared with WKY rats. These results confirm the importance of Crgn1 on chromosome 13 in EAG susceptibility, mediated partly through differences in Fc receptor-mediated macrophage activation. 相似文献

A new flow injection spectrophotometric method for the determination of analgin has been developed. The main factors which affect the determination were investigated. The absorption maximum is 650 nm. Under optimum conditions the calibration graph is linear in the range of 0. 2~3 mg/ml, the average recovery is 99. 8% with a relative standard deviation of 0. 5%. The proposed system permits the analysis of about 240 samples per hour and has the advantages of analytical efficiency, sensitivity and accuracy for the determination of analgin. 相似文献

Extracorporeal shock wave lithotripsy (ESWL) was performed on 925 patients (1,222 treatments) for 446 calyceal, 345 pelvic, 172 ureteral, and 108 staghorn calculi. ESWL necessitated 6.3 KUB and 1.2 renal ultrasound studies per treatment session. Intravenous urography was required in 6% of patients after ESWL. Percutaneous nephrostomy tube placement was performed by the radiologists in 10% of patients. Procedures related to nephrostomy tube placement included percutaneous nephrolithotripsy, tube changes, nephrostography, and stone fragment irrigations and retrievals. Staghorn calculi treatment with ESWL required the most procedures by radiologists (34% for partial staghorn and 56% for complete staghorn) compared with 3%, 8%, and 11% for calyceal, pelvic, and ureteral stones, respectively. In all, 8,478 radiologic examinations and procedures were performed pertaining to ESWL. This is approximately 35 studies per day. While this number may be high because it represents the early experience of the authors with ESWL, the impact on the radiology department can be substantial. 相似文献
