Equine herpesvirus 4 (EHV-4) is an important pathogen that causes respiratory tract disease in horse populations worldwide.
Glycoprotein G (gG) homologs have been identified in several alphaherpesviruses as minor non-essential membrane-anchored glycoproteins.
In this study, EHV-4 gG deletion mutant has been generated by using bacterial artificial chromosome technology to investigate
the role of gG in viral pathogenesis. Our findings reported here revealed no significant difference between parental EHV-4
and gG-negative strain in their replication cycle in cell culture. Furthermore, virus titers and plaque formation were comparable
in both viruses. It is noteworthy that these findings disagree with the previously published study describing gG deletion

Background and objective
Laparoscopic sleeve gastrectomy become widely employed and accepted operation in between Arab population as a permanent solution for morbid obesity. There are many acute complications associated with LSG such postoperative leaks or bleeding, abscess formation, splenic infarction and portal vein thrombosis. The purpose of the study is to present how multidetector CT is beneficial as a screening investigation to detect these complications.Methods
We retrospectively review CT images done in the first week post operative for280 patients subjected to LSG the center in the period between July 2017-December 2017 (average age 39.5?years old).Results
the leak was suspected in 3 patients documented by a direct leak of contrast in one of them, 2 patients active bleeding was clinically suspected, but the exact site of bleeding could not be identified by CT, 15 patients had serous fluid collection, splenic infarction was noticed in 18 patients, 3 patients developed postoperative hematoma one of them become infected, anterior abdominal wall hematoma seen in 5 patients, one patient developed portal vein thrombosis.Conclusion

Purpose
Boron neutron capture therapy (BNCT) has the potential to become a viable cancer treatment modality, but its clinical translation has been limited by the poor tumor selectivity of agents. To address this unmet need, a boronated 2-nitroimidazole derivative (B-381) was synthesized and evaluated for its capability of targeting hypoxic glioma cells.Methods
B-381 has been synthesized from a 1-step reaction. Using D54 and U87 glioma cell lines, the in vitro cytotoxicity and cellular accumulation of B-381 has been evaluated under normoxic and hypoxic conditions compared to L-boronophenylalanine (BPA). Furthermore, tumor retention of B-381 was evaluated in vivo.Results
B-381 had low cytotoxicity in normal and cancer cells. Unlike BPA, B-381 illustrated preferential retention in hypoxic glioma cells compared to normoxic glioma cells and normal tissues in vitro. In vivo, B-381 illustrated significantly higher long-term tumor retention compared to BPA, with 9.5-fold and 6.5-fold higher boron levels at 24 and 48 h, respectively.Conclusions
B-381 represents a new class of BNCT agents in which their selectivity to tumors is based on hypoxic tumor metabolism. Further studies are warranted to evaluate B-381 and similar compounds as preclinical candidates for future BNCT clinical trials for the treatment of glioma.57.

Background: Synchronous bilateral breast cancer (SBBC) provides a special condition where two independent breast tumors are exposed to cancer pharmacotherapy within a uniform pharmacokinetic milieu. Both senescence and apoptosis are established responses to therapy; however, they have potentially variable contributions to the overall outcome of treatment, which are yet to be determined. Methods: In this report, we describe the clinicopathological picture of two SBBC cases that received standard anticancer treatment and assess their expression profile of several molecular hallmarks of senescence and apoptosis. Results: Our analysis identified that synchronous tumors have variable expression profiles of both senescence- and apoptosis-associated biomarkers, despite comparable pathological responses to neoadjuvant chemotherapy and current survival rates. Conclusions: Our results highlight the variable expression of senescence- and apoptosis-associated markers in breast tumors (despite the shared somatic genetic background) and invites a large-scale assessment of both senescence and apoptosis in breast cancer tissue in vivo and their contribution to the pathological response and overall survival. 相似文献

The nuclear factor-B (NF-B) path-way has been implicated in tumor B-cell survival, growth, and resistance to therapy. Because tumor cells overcome single-agent antitumor activity, we hypothesized that combination of agents that target differentially NF-B pathway will induce significant cytotoxicity. Therapeutic agents that target proteasome and Akt pathways should induce significant activity in B-cell malignancies as both pathways impact NF-B activity. We demonstrated that perifosine and bortezomib both targeted NF-B through its recruitment to the promoter of its target gene IB using chromatin immunoprecipitation assay. This combination led to synergistic cytotoxicity in Waldenstrom macroglobulinemia (WM) cells that was mediated through a combined reduction of the PI3K/Akt and ERK signaling pathways, found to be critical for survival of WM cells. Moreover, a combination of these drugs with the CD20 monoclonal antibody rituximab further increased their cytotoxic activity.
