Fifteen plant species were collected from the Nelspruit Botanical Garden based on a list of plants provided by Phytomedicine Programme at the University of Pretoria and their ethnopharmacological information. Hexane, dichloromethane (DCM), acetone and methanolic extracts were screened for antimycobacterial activity against Mycobacterium smegmatis. The acetone extract of Milletia stulhimannii was the most active, showing activity against Mycobacterium smegmatis with minimum inhibitory concentration (MIC) value of 0.13 mg/ml. Acetone extracts for all plants had lower MIC values ranging between 0.11–1.25 mg/ml against M. smegmatis. Milletia stulhimannii, Albizia gummifera, Xanthocercis zambesiaca and Barringtonia racemosa have shown great potential as anti-tuberculosis agents. Covalent interactions between antibody combining site residues and substrates have been implicated in the catalytic activity of abzymes elicited by design or occurring naturally in autoimmune disease. In this study, the potential for covalent binding by antibodies (Abs) was investigated by the induction of immune responses against molecules presenting chemically reactive haptenic groups. Immunogenic conjugates containing a phosphonate diester or a pyruvate carbonyl group were used to elicit antibodies that could specifically react with the electrophilic moieties. Products formed by covalent binding were detected by a western blot technique or by differential ELISA on reduced or unreduced carbonyl haptens. Antisera to the diphenylphosphonate contained antibodies with covalent reactivity, which increased with immunization. The reactivity was specific to the anti-phosphonate response and not to control immune sera induced against the unmodified carrier protein. Reactivity was focused on the antibody light (L) chain. Antisera to the phenylpyruvate hapten appeared to bind strongly to proteins modified by the carbonyl group hapten. However, anti-carrier antisera and non-immune sera had similar reactivity, indicating that the pyruvate moiety reacts nonspecifically with immunoglobulins. This suggested that affinity maturation of antibodies for reversible binding through hemiacetal or Schiff base adducts with antigens requires a less reactive carbonyl in the antigen. On the other hand, the induction of antibodies with enhanced nucleophilic reactivity toward phosphonate esters implies that irreversible binding to the B cell receptor can drive clonal expansion and antibody selection. These results support a designer strategy for generating nucleophilic abzymes and could also account for the occurrence of chemically reactive or catalytic antibodies in natural immunity or autoimmunity. 相似文献

Cerebral and spinal location of glioneuronal tumors have been recently described as a novel type of primary CNS neoplasia. A distinctive rare form of glioneuronal tumors with neuropil‐like islands (GTNI) have been reported to occur in the adult cerebrum, whereas spinal GTNI localization is extremely rare. In the present report we describe a case of a 15‐month‐old child with a spinal GTNI of the cervical region and meningeal dissemination. Histologically the tumor was composed of round, small neurocytic‐like cells arranged around eosinophilic neuropil cores and embedded in a diffuse fibrillar glial component forming prominent “rosetted” neuropil islands displaying strong immunoreactivity for neuronal markers. Cerebral GTNI shows abundant glial components not rarely exhibiting anaplastic features that justify their inclusion within the group of diffuse astrocytomas. In contrast, including our case, spinal GTNI do not show histological evidence of anaplastic features and exhibits a significant neuronal component that may imply considering these lesions in a separate group. Nevertheless, due to their exceptional rarity, the natural history of these lesions is not yet fully understood, but spinal GTNI seems to have an unfavorable clinical course despite their benign histopathological features, which must be taken into account for appropriate treatment and follow‐up of the patient. 相似文献

Damping is the conversion of mechanical energy of a structure into thermal energy, and it is related to the material viscous behavior. To evaluate the role of damping in the common carotid artery (CCA) wall in human hypertension and the possible improvement of angiotensin-converting enzyme (ACE) inhibition, we used noninvasive CCA pressure (tonometry) and diameter (B-mode echography) waveforms in normotensive subjects (NT group; n=12) and in hypertensive patients (HT group; n=22) single-blind randomized into HT-placebo (n=10) or HT-treated (ramipril, 5 to 10 mg/d during 3 months; n=12). Vascular smooth muscle (VSM) null tonus condition was achieved from in vitro pressure and diameter waveforms (Konigsberg microtransducer and sonomicrometry) measured in explanted human CCA (n=14). Arterial wall dynamics was described by viscous (eta), inertial (M), and compliance (C) parameters, mean circumferential wall stress, viscous energy dissipation (WD), peak strain energy (WSt), damping ratio (xi=WD/WSt), and modeling isobaric indexes CIso and WSt(Iso). The lack of VSM tonus isobarically increased wall stress and reduced eta, CIso, and damping (P<0.01). Wall stress, eta, and WD were greater in HT than in NT (P<0.015) and arrived near normal in HT-treated (P<0.032 respect to HT), with no changes in HT-placebo. Whereas CIso increased in HT-treated (P<0.01) approaching the NT level, xi did not vary among groups. During hypertension, because of the WSt increase, the arterial wall reacts increasing WD to maintain xi. ACE inhibition modulates VSM activation and vessel wall remodeling, significantly improving wall energetics and wall stress. This protective vascular action reduces extra load to the heart and maintains enhanced arterial wall damping. 相似文献
