HSP90 inhibition might be a promising strategy to overcome the radioresistance of some cancers. In the current study, we further explored the mechanisms of HSP90 in regulating the radiosensitivity of cervical cancer cells. Bioinformatic analysis was performed based on data from TCGA‐CESC. Cellular and molecular studies were conducted using CaSki and SiHa and the derived radioresistant (RR) subclones. Through a proteomics screen, we identified HSP90 chaperones (both HSP90α and HSP90β) as CD147‐binding partners supporting its stabilization. Targeting HSP90 sensitized CaSki‐RR and SiHa‐RR cancer cells to irradiation partially through CD147 destabilization. Mechanistically, HSP90 interacts with FBXO6 and reduces FBXO6‐mediated proteasomal degradation of CD147. Enforced FBXO6 overexpression also sensitized CaSki‐RR and SiHa‐RR cancer cells to irradiation. These effects were enhanced using 17‐AAG treatment but were weakened by CD147 overexpression. Survival analysis further confirmed the association between high FBXO6 expression and favorable progression‐free survival among patients with cervical cancer. In conclusion, this study showed that HSP90 promotes radioresistance of cervical cancer cells partially via reducing FBXO6 mediated CD147 polyubiquitination. These findings help to explain why HSP90 inhibitor exerts radio‐sensitizing effects in cervical cancer. 相似文献

目的研究单排螺旋 CT 肺动脉造影在肺动脉栓塞诊断中的应用价值。方法对临床怀疑肺动脉栓塞的31例患者,采用单层螺旋 CT 行薄层肺动脉造影(CT pulmonary angiography,CTPA)检查;利用原始数据进行层厚1mm 的轴面重建,比较层厚3mm 和1mm 图像的诊断价值。结果 CTPA 可显示肺动脉栓塞的直接征象(充盈缺损、附壁血栓、完全阻塞)以及间接征象(肺动脉高压、肺梗塞等)。1mm 有效层厚图像对段、亚段肺动脉内血栓的显示高于3mm 有效层厚图像(P<0.05)。结论单层螺旋 CT 肺动脉造影是诊断肺动脉栓塞的快速、有效诊断方法。 相似文献

Under the background of climate change, the steel industry is considered one of the least eco-friendly industries. Flax fiber-reinforced polymer (FFRP) is an emerging sustainable alternative to steel reinforcement bar; however, its application is much restricted due to its interior material properties. This paper proposed a novel way to form closed-shape stirrups with FFRP, which is suitable for replacing steel stirrups. A multi-disciplinary investigation was conducted concerning the structural and environmental performance of FFRP stirrups in reinforced concrete (RC) beams. Seven specimens were tested under a three-point bending load. The FFRP stirrups substantially increased the shear capacity and ultimate vertical displacement by 77% and 74%, respectively, and shifted brittle failure to ductile failure. The closed-shape stirrups avoided the stress concentration and increased the utilization of FFRP tensile capacity to over 80%. Decreasing the spacing of FFRP stirrups effectively increased the shear capacity and ductility; increasing the width or layer of FFRP stirrups improved ductility only. A life cycle assessment (LCA) was later performed to evaluate and compare the environmental performance of steel, FFRP, and carbon FRP stirrups. As compared to carbon FRP and steel ones, FFRP stirrups substantially decreased the global warming and fossil depletion potential by over 60%. The main contributors to the environmental impacts of FFRP stirrups were the heavy metal released into the water and terrestrial environment during the cultivation process. 相似文献

Phospholipids are pivotal polar lipids in human milk and essential for infants’ growth and development, especially in the brain and cognitive development. Its content and composition are affected by multiple factors and there exist discrepancies in different studies. In this study, we determined five major phospholipids classes (phosphatidylethanolamine, phosphatidylinositol, phosphatidylserine, phosphatidylcholine, and sphingomyelin) in 2270 human milk samples collected from 0 to 400 days postpartum in six regions of China. The high-performance liquid chromatography coupled with an evaporative light scattering detector (HPLC-ELSD) was performed to quantify the phospholipids. Total phospholipid median (IQR) content was in a range between 170.38 ± 96.52 mg/L to 195.69 ± 81.80 mg/L during lactation and was higher concentrated in colostrum milk and later stage of lactation (after 200 days postpartum) compared with that in the samples collected between 10 to 45 days postpartum. Variations in five major sub-class phospholipids content were also observed across lactation stages (phosphatidylethanolamine: 52.61 ± 29.05 to 59.95 ± 41.74 mg/L; phosphatidylinositol: 17.65 ± 10.68 to 20.38 ± 8.55 mg/L; phosphatidylserine: 15.98 ± 9.02 to 22.77 ± 11.17 mg/L; phosphatidylcholine: 34.13 ± 25.33 to 48.64 ± 19.73 mg/L; sphingomyelin: 41.35 ± 20.31 to 54.79 ± 35.26 mg/L). Phosphatidylethanolamine (29.18–32.52%), phosphatidylcholine (19.90–25.04%) and sphingomyelin (22.39–29.17%) were the dominant sub-class phospholipids in Chinese breast milk during the whole lactation period. These results updated phospholipids data in Chinese human milk and could provide evidence for better development of secure and effective human milk surrogates for infants without access to breast milk. 相似文献
