Benign tumors and tumor-like lesions of the adult kidney. Part I: Benign renal epithelial neoplasms

The spectrum of renal neoplasms has expanded in recent years. Although most of the work taking place in this field has concerned malignant neoplasms of the kidney, there have been significant improvements in our knowledge of benign renal tumors and tumor-like lesions, especially in renal cell adenoma, renal oncocytoma, and renal angiomyolipoma. Awareness and knowledge of these benign lesions is important because they are often included in the differential diagnoses of malignant tumors, with which they may be confused both clinically and pathologically. The authors review the topic of benign renal neoplasms and tumor-like lesions that occur in adults, emphasizing some of the newly described aspects of these lesions.     

Lipofectin介导转移的外源性ADA基因在反复上感儿T淋巴细胞的表达

本文对４１例健康儿童和１７例反复上呼吸道感染患儿外周血淋巴细胞腺苷脱氨酶（ＡＤＡ）活性进行了检测。在此基础上筛选出２例反复上感伴ＡＤＡ活性低下患儿。在体外对这２例患儿的外周血Ｔ淋巴细胞进行培养后，以Ｌｉｐｏｆｅｃｔｉｎ（脂质体）介导的方法对其进行了外源性ＡＤＡ基因的基因转移。结果显示：２例患儿体外培养淋巴细胞ＡＤＡ活性较转基因前升高。同步进行的标志基因ｐＢＬａｃＺ的基因转移的检测结果也直观地证实了Ｌｉｐｏｆｅｃｔｉｎ介导的基因转移是成功的。该研究为ＡＤＡ－ＳＣＩＤ淋巴细胞基因治疗的研究提供了初步的体外实验资料。     

Immunohistochemical analysis of p53 protein expression in benign and malignant skin tumors using a panel of anti-p53 antibodies.

The expression of p53 in a variety of benign and malignant skin lesions has been first assessed in frozen sections and then compared with the results obtained in corresponding paraffin-embedded sections using various immunohistochemical staining methods with a panel of anti-p53 antibodies. Of the 48 benign and malignant skin lesions studied, 46(96%) had corresponding paraffin sections and immunohistochemical results obtained with DO7 on frozen and paraffin sections were concordant in 97%, qualitatively. Using streptavidin-biotin complex method, p53 was identified in 33% of dysplastic squamous lesions, 50% of squamous cell carcinomas (SCCs) and 36% of basal cell carcinomas (BCCs) on frozen section, whereas 25% of dysplastic squamous lesions, 40% of SCCs, and 32% of BCCs showed p53 positivity on paraffin-embedded sections. In frozen sections, the same regions of each specimen exhibited similar topographic patterns of positive immunoreactivity with both monoclonal antibodies, PAb 1801 and DO7. In contrast, immunohistochemical staining with polyclonal antibody, CM-1, gave poor morphologic resolution, although effective in paraffin-embedded sections.

目的 探讨反义血管内皮生长因子（ＶＥＧＦ）基因转染在抑制恶性肿瘤生长和转移的抗肿瘤血管基因治疗中的意义。方法 利用基因重组技术构建正义和反义ＶＥＧＦ１２１ ｃＤＮＡ真核表达载体,用脂质体法转染高转移性人巨细胞肺癌细胞（ＰＧ）,经Ｎｏｒｔｈｅｍ杂交和Ｗｅｓｔｅｒｎ印迹免疫化学检测ＶＥＧＦ ｍＲＮＡ和蛋白质的表达水平,并对转染前后细胞进行体外生长和裸鼠体内生长转移等多项生物学行为实验,结果 转染反义转     

腺病毒载体介导的lacZ基因在NG细胞系及大鼠黑质的表达

本实验用标记基因ｌａｃＺ５型重组腺病毒（Ａｄ５ＣＭＶｌａｃＺ）转染培养的ＮＧ细胞系，Ｘ－ｇａｌ染色检测转染效率．在培养的ＮＧ细胞系，当病毒滴度为２×１０８时，转集率达到５０％，当滴度为２×１０９时，转染率达１００％，有较好的量效关系；固定病毒液度为１０１０，培养２～１６ｈ，细胞的转染率随时间延长而提高，有较好的时效关系。将Ａｄ５ＣＭＶｌａｃＺ注射到大鼠黑质部位后，分别于注射后３～１２０ｄ取脑、切片、Ｘ－ｇａｌ染色，发现黑质局部从第７ｄ开始有部分蓝染，第１０ｄ达高峰，注射局部感染率１００％；９０ｄ时开始下降，持续至１２０ｄ；纹状体等其它部位无蓝染．上述结果提示，腺病毒载体介导的标记基因可在培养的神经细胞系和中脑黑质部位高效表达，为进一步开展中枢神经系统退变性疾病尤其是帕金森氏病的基因治疗奠定基础。     

Ribosome-lamella complexes in a case of transitional cell carcinoma involving the prostate

We report ribosome-lamella complexes (RLC) in cancer cells of transitional cell carcinoma (TCC) of the prostate in a 52-year-old Caucasian man. Histopathologically, cancer cells were proliferated in various-sized nests, mostly associated with central necrosis. Some invaded into the surrounding normal glandular space and the stroma, with occasional lymphatic invasion. Fine structural study of cancer cells revealed that cross-sectioned RLC as well as densely aggregated ribosomes were detected in their cytoplasm, situated close to, but not directly connected with, dilated rough endoplasmic reticulum. These were composed of a concentric alternative arrangement of both lamellae and ribosomes. In the central and surrounding parts of the RLC, ribosomes were observed, revealing a smooth transition to the ribosomal component of RLC in size and shape. The presence of both RLC and dense aggregation of ribosomes close to the rough endoplasmic reticulum suggests that their functions might be related to specific or aberrant protein synthesis under unknown conditions. Although RLC have been often reported in hematopoietic malignancies, their occurrence in the malignant epithelial component has been only reported in a case of pulmonary adenocarcinoma. This is the first report of RLC in TCC in the literature.     

ADA活性紫外吸收测定法的改良及复感儿淋巴细胞该酶活性的检测

检测了42例健康儿童和17例反复上呼吸道感染患儿（复感儿）的血淋巴细胞腺苷脱氨酶（ADA）活性，结果表明：复感儿的血淋巴细胞中ADA活性较健康儿童低下，且大多同样伴有不同程度的免疫功能低下；从复感儿组中筛选了两侧ADA活性和免疫功能明显低下的患儿，拟采用这两例患儿的血淋巴细胞进行ADA－SCID基因治疗的实验研究。     

Effects of repeated subculturing and prolonged storage at room temperature of enterohemorrhagic Escherichia coli O157:H7 on pulsed-field gel electrophoresis profiles

Three clinical strains of enterohemorrhagic Escherichia coli O157:H7 which were subcultured repeatedly or stored at room temperature over a 25-week period showed appreciable variations in their pulsed-field gel electrophoresis fragment patterns. The variations could be explained by a couple of spontaneous genetic events at most and thus did not invalidate the genetic lineage of the strains.
