To elucidate the early events of blood-borne metastasis under actual blood flow, real-time trafficking of RAW117 large cell lymphoma cells, namely parental RAW117-P and liver-metastatic RAW117-H10 cells, was investigated using positron emission tomography (PET). Both types of cells accumulated in the liver immediately after injection via the portal vein, and were eliminated from the liver time-dependently. The elimination rate of RAW117-H10 cells, however, was slower than that of RAW117-P cells, suggesting that RAW117-H10 cells interact more strongly with hepatic sinusoidal endothelium than the parental cells. This result correlated with the metastatic potential of these cells: RAW117-H10 cells metastasized in the liver to a greater extent than RAW117-P cells after injection via this route. To investigate the role of sialylglycoconjugates in the interaction of RAW117-H10 cells with the hepatic endothelium after injection via the portal vein, the trafficking of RAW117-H10 cells was examined after the cells had been treated with sialidase. The elimination rate of RAW117-H10 cells from liver was observed to be greatly accelerated by sialidase treatment. To elucidate what kind of sialylglycoconjugates is related to this phenomenon, we analyzed the distribution of sialyl Lewis A and sialyl Lewis X antigens of both sublines of RAW117 by using flow cytometry. RAW117-H10 cells were found to express a much higher level of sialyl Lewis A than RAW117-P cells, whereas the amount of sialyl Lewis X did not differ significantly. These findings suggest that some sialylglycoconjugates, perhaps sialyl Lewis A in particular, play an important role in the initial interaction of RAW117-H10 cells with the hepatic endothelium, leading to metastasis. 相似文献

The purpose of this study was to apply an electronic nose system for evaluation of unpleasant odor in tablets containing L-cysteine, an unpleasant odor drug, and demonstrate the odor masking ability of thin-layer sugarless coated tablets, which we have newly developed, by both electronic nose system and sensory evaluations. We demonstrated the qualitative evaluation of the unpleasant odor using air as a reference indicator and the quantitative evaluation of the unpleasant odor using the distances between air and samples in the electronic nose system evaluation. The electronic nose system evaluation was positively and well-correlated with the sensory evaluation by volunteers. We suggest that the electronic nose system evaluation is appropriate as an alternative or a support method for sensory evaluation by volunteers. As the results of both electronic nose system and sensory evaluations, we demonstrated that the thin-layer sugarless coated tablets have excellent masking ability of the unpleasant odor, equivalent to that of sugar-coated tablets due to the dense coating layers. 相似文献

Objective: To report four cases of rice bodies (RBs) showing remarkable size variations and discuss their pathogenesis.
Design and patients: Based on analysis of the clinical data, we speculate on the pathogenesis of RBs using immunohistochemical and ultrastructural
methods. The patients comprised three men and one woman, three with RBs in the subacromial bursae and one in the wrist synovial
sheath, aged 28 (woman), 44, 50 and 81 (wrist) years, respectively.
Results: There were no particular differences in clinical data among the patients. T2-weighted MR imaging was very useful for diagnosis
of the RBs, allowing their clear delineation from the bursal fluid. The RBs consisted of a layered protein- aceous substance
with vague targetoid cut surfaces. Much fibrin and a lesser amount of collagen fibers were recognized together with various
mononuclear cells, which were few in number and predominantly T cells. The bursae and synovial sheath had multiple fibrinoid
spheroids at the luminal surface. Conclusion: Fibrinoid nodular deposits probably became detached, forming the nuclei of RBs and growing to a giant RB 65 mm in diameter.

Cerebrospinal fluid (CSF) tau protein phosphorylated at both Thr231 and Ser235 sites (CSF/phospho-tau(231-235)) and total tau (CSF/total-tau) were quantified by sandwich ELISA in 20 patients with mild cognitive impairment (MCI) who eventually developed AD on follow-up as well as seven memory complainers with no objective memory loss. 13/20 (65%) of the MCI patients had high CSF/total-tau and detectable levels of CSF/phospho-tau(231-235), whereas these markers were low and under a detectable level in all of the memory complainers. Although either a total-tau, phospho-tau measurement or a combination of these can help in predicting if MCI will develop AD, our results suggest that the pathogenic steps of AD may be at the stage that finally leads to an accumulation of abnormally phosphorylated tau and neuron death, at least in some brain areas, when MCI patients present with the earliest detectable clinical symptoms of dementia. 相似文献

Social skill deficits in schizophrenia profoundly affect patients' life-long outcome, although the profile of the underlying cognitive dysfunction still remains a matter of debate. In the present study, we investigated the relationship between social skills and event-related potentials (ERPs) in an auditory selective attention task, in addition to the neurocognitive indices obtained from the degraded-stimulus continuous performance test (CPT) and clinical indices, such as Brief Psychiatric Rating Scale (BRPS) and global assessment of function (GAF) scores. Social skills were assessed using a Japanese version of the structured role play test. Fourteen persons with schizophrenia participated in the study. Non-verbal skills showed a positive correlation with GAF, the performance level, N1 and N2b amplitude in the ERP task, and hit rate in the CPT, and a negative correlation with reaction time in the CPT. Verbal communication skills showed a positive correlation with GAF, the performance level and N2b amplitude in the ERP task, and hit rate in the CPT, and a negative correlation with reaction time in the CPT. Processing skills showed a positive correlation with the performance level and N1 amplitude in the ERP task and a negative correlation with reaction time in the CPT. These findings suggested that the social skill deficits of persons with schizophrenia were related to the vigilance level and controlled stimulus detection processing. 相似文献

It is still in doubt whether the standard-dose growth hormone (GH) used in Japan (0.5
IU/kg/week, 0.167 mg/kg/week) for growth hormone deficiency is effective for achieving
significant adult height improvement in non-growth hormone deficient (non-GHD) short
children. We compared the growth of GH-treated non-GHD short children with that of
untreated short children to examine the effect of standard-dose GH treatment on non-GHD
short children. GH treatment with recombinant human growth hormone (rhGH) was started
before the age of 11 yr in 64 boys and 76 girls with non-GHD short stature registered at
the Foundation for Growth Science who have now reached their adult height. In 119
untreated boys and 127 untreated girls whose height standard deviation score (SDS) was
below –2 SD at the age of 6 yr, height growth was followed until 17 yr. Height SDS was
significantly lower before GH treatment in the GH-treated group than at the age of 6 yr in
the untreated group, in both sexes. Adult height and adult height SDS were significantly
greater in the untreated group than in the GH-treated group, in both sexes, although the
change in height SDS did not differ significantly. Height SDS was significantly lower
before GH treatment in the GH-treated group than at the age of 6 yr in the untreated
group, so 57 boys and 57 girls whose height SDS at the age of 6 yr in the untreated group
closely matched the height SDS before GH treatment in the GH-treated group were chosen for
comparison. Height SDS did not differ significantly between the GH-treated group before GH
treatment and the untreated group at the age of 6 yr, nor were there differences between
these subgroups in adult height, adult height SDS, or height SDS change, in either sex.
The effect of GH treatment is reported to be dose-dependent and doses over 0.23 mg/kg/week
are reported to be necessary to improve adult height in non-GHD short children. Currently,
the GH dose is fixed at 0.175 mg/kg/week in Japan, and we expected to find, and indeed
concluded, that ordinary GH treatment in Japanese, non-GHD short children does not improve

Cancer immunotherapy by fusion of antigen-presenting cells and tumor cells has been shown to induce potent antitumor immunity. In this study, we characterized syngeneic and allogeneic, murine macrophage/dendritic cell (DC)-cancer fusion cells for the antitumor effects. The results showed the superiority of allogeneic cells as fusion partners in both types of antigen-presenting cells in an in vivo immunotherapy model. A potent induction of tumor-specific CTLs was observed in these immunized conditions. In addition, the immunization with DC-cancer fusion cells was better than that with macrophage-cancer fusion cells. Both syngeneic and allogeneic DC-cancer fusion cells induced higher levels of IFN-gamma production than macrophage-cancer fusion cells. Interestingly, allogeneic DC-cancer fusion cells were superior in that they efficiently induced Th1-type cytokines but not the Th2-type cytokines interleukin (IL)-10 and IL-4, whereas syngeneic DC-cancer fusion cells were powerful inducers of both Th1 and Th2 cytokines. These results suggest that allogeneic DCs are suitable as fusion cells in cancer immunotherapy. To further enhance the antitumor immunity in the clinical setting, we prepared DCs fused with IL-12 gene-transferred cancer cells and thus generated IL-12-secreting DC-cancer fusion cells. Immunization with these gene-modified DC-cancer fusion cells was able to elicit a markedly enhanced antitumor effect in the in vivo therapeutic model.
