Expression and significance of stromal cell-derived factor-1alpha and its receptor CXCR4 in human dental pulp cells 总被引:1,自引:0,他引:1
OBJECTIVE: To investigate the expression of CXCR4 in cultured human dental pulp cells (HDPC) in vitro and the corresponding ligand SDF-1alpha level of HDPC supenatants stimulated by lipopolysaccharide (LPS) and tumor necrosis factor-alpha (TNF-alpha), and to explore the role of SDF-1alpha on the proliferation and the migration of HDPC. METHODS: The expression of CXCR4 in HDPC was detected by immunocytochemistry technique and indirect immunofluorescence technique. The culture supernatants of HDPC were collected after HDPC had been simulated by LPS and TNF-alpha of different concentrations for 48h and then the SDF-1alpha level was assayed by quantitative sandwich ELISA. Meanwhile, the effects of recombinant human SDF-1alpha (rhSDF-1alpha) on the proliferation and the migration of HDPC at different concentrations were observed by MTT and Boyden Chamber Assay. RESULTS: CXCR4 was expressed in cytomembrane of HDPC and SDF-1alpha was secreted into their normal cell supernatants with a concentration of (4513.55 +/- 962.92) ng/L. The secretion of SDF-1alpha was both significantly decreased by stimulation with LPS and TNF-alpha (P < 0.05). In addition, rhSDF-1alpha stimulated the HDPC proliferation at the concentrations of 50, 100, 200 microg/L (P < 0.01) and increased the chemotactic migration of HDPC significantly after 9h's incubation with the concentrations of 50, 100 microg/L (P < 0.05). CONCLUSIONS: SDF-1alpha accelerated the proliferation and the migration of HDPC which expressed CXCR4. SDF-1-CXCR4 axis may play a role in repair of pulp injury. 相似文献

OBJECTIVES: Bone repair strategies continue to be developed for alternatives to autografting, allogeneic implants of banked bone, and other bone substitutes. Efforts have included the delivery of potent growth and/or differentiation factors and the use of gene therapy. For bone regeneration, gene therapy is the delivery, uptake and expression of DNA that has been localized to a wound bed. The objective of the current study is to investigate methods to enhance non-viral-mediated means of gene uptake and expression for use in bone regeneration. METHODS: Several types of DNA-polymer complexes, either applied directly to baby hamster kidney (BHK) cells, or released from a porous, resorbable gene-activated matrix (GAM), were evaluated in vitro for their ability to transfect cells with a circular plasmid DNA construct expressing green fluorescent protein. Complexes included conjugates containing a lipophilic reagent, liposomes, poly-ethyl-oxazoline, and poly-ethyleneimine (PEI). Data were subjected to analysis of variance and Fisher's protected least significant difference for multiple comparisons with significance established at p < 0.05. RESULTS: Transfection efficiencies of the liposome and PEI complexes improved in vitro when released from resorbable GAMs. The lipophilic reagent FuGene 6 demonstrated abundant uptake and expression in the initial 1- and 2-day evaluation periods. In contrast, the DNA-liposome and PEI GAM complexes demonstrated a sustained release, uptake and expression by the BHK cells at the 2-, 4-, and 7-day, and 4- and 7-day evaluation intervals, respectively. CONCLUSION: GAM technology appears to improve the functional stability and release duration of incorporated DNA-polymer complexes in the present in vitro studies. 目的:了解蜂胶及自制蜂胶奥硝唑合剂对人牙龈成纤维细胞的毒性作用,为蜂胶及合剂的临床推广应用提供理论依据.方法:人牙龈成纤维细胞在不同浓度的蜂胶、奥硝唑和自制蜂胶奥硝唑合剂中体外培养24 h,用MTT法测定细胞相对增殖率(RGR);选择细胞增殖率约为50%的药物浓度组,重新培养细胞24 h,更换成不含药物的培养液连续培养7 d,每天用MTT法测定吸光度值,了解细胞增殖回复情况.结果:0.5g/L蜂胶组的RGR达到93.9%,与蜂胶其他2组间的差别有统计学意义(P<0.05);奥硝唑溶液加入蜂胶后,对应浓度组比较,两合剂组的平均吸光度值均大于奥硝唑组,其中当奥硝唑浓度为0.4g/L和0.8g/L时,差别有统计学意义(P相似文献

目的比较根尖切除术中传统慢速手机与超声器械对于根尖倒预备的不同影响。方法借助扫描电镜对48颗离体前磨牙进行观察研究,使用AutoCAD2005制图软件辅助计算。结果使用传统慢速手机进行根尖倒预备后的牙根硬组织最小厚度(均值0.15mm)明显低于使用超声器械(S129D均值0.82mm,S129均值0.76mm),具有统计学显著意义(P<0.05)。结论使用超声器械进行根尖倒预备可以有效的保护牙根硬组织,保持牙根的强度。 相似文献

目的 探讨口腔正畸治疗中微型种植体支抗的应用效果.方法 选取2019年5月至2020年3月于本院接受口腔正畸治疗的54例患者作为研究对象,采用电脑分组法分为常规组与微型组,各27例.常规组采用常规口腔正畸治疗方法,微型组采取微型种植体支抗治疗方法,比较两组临床治疗效果、治疗参数变化及并发症发生率.结果 微型组治疗总有效率为92.59%,高于常规组的70.37%(P<0.05).微型组上中切牙凸距差、上中切牙角及磨牙位移改善情况均明显优于常规组(P<0.05).微型组并发症发生率为7.41%,低于常规组的29.63%,差异有统计学意义(P<0.05).结论 在进行口腔正畸治疗时,选择微型种植体支抗治疗方案,能有效矫正患者牙齿问题,提高治疗效率,降低并发症发生率,提升治疗舒适度,具有一定的临床应用价值. 相似文献

目的探讨PICC置管患者携管出院期间的维护方法。方法静脉治疗团队将进修护士、肿瘤专科护士、静疗专科护士及静疗学习班学员纳入网络维护人员,逐步建立PICC维护网点并不断扩大规模,通过建立维护花名册、QQ群、专科护士返基地活动等措施进行管理。结果 PICC维护网络已覆盖到江苏省126所二、三级医院及安徽省3个市县的3所二、三级医院,参与PICC维护网点维护的护士达195人。携管出院患者导管相关并发症(静脉炎、导管堵塞、皮肤湿疹、渗液、非计划拔管)的总发生率由2008年的17.9%下降至2013年的3.5%;借助QQ群解决了近600例置管及维护疑难问题。结论 PICC维护网络的建立,满足了携管出院患者治疗间歇期间的导管维护需求,既增加了维护点医院的经济效益又提升了社会效益。 相似文献

Stereotypical connections between olfactory sensory neuron axons and mitral cell dendrites in the olfactory bulb establish the first synaptic relay for olfactory perception. While mechanisms of olfactory sensory axon targeting are reported, molecular regulation of mitral cell dendritic growth and refinement are unclear. During embryonic development, mitral cell dendritic distribution overlaps with olfactory sensory axon terminals in the olfactory bulb. In this study, we investigate whether olfactory sensory neurons in the olfactory epithelium influence mitral cell dendritic outgrowth in vitro. We report a soluble trophic activity in the olfactory epithelium conditioned medium which promotes mitral/tufted cell neurite outgrowth. While the trophic activity is present in both embryonic and postnatal olfactory epithelia, only embryonic but not postnatal mitral/tufted cells respond to this activity. We show that BMP2, 5 and 7 promote mitral/tufted cells neurite outgrowth. However, the BMP antagonist, Noggin, fails to neutralize the olfactory epithelium derived neurite growth promoting activity. We provide evidence that olfactory epithelium derived activity is a protein factor with molecular weight between 50–100 kD. We also observed that Follistatin can effectively neutralize the olfactory epithelium derived activity, suggesting that TGF-beta family proteins are involved to promote mitral/tufted dendritic elaboration. 相似文献
