Conformations of neurotensin in solution and in membrane environments studied by 2-D NMR spectroscopy

Two-dimensional HOHAHA and ROESY nuclear magnetic resonance techniques are used to obtain complete proton resonance assignments and to perform a conformational investigation of the neuropeptide neurotensin (pGlu-Leu-Tyr-Glu-Asn-Lys-Pro-Arg-Arg-Pro-Tyr-Ile-Leu) in aqueous solution, methanol, and membrane-mimetic [deuterated sodium dodecylsulfate (SDS)] environments. Results suggest the absence of discernible elements of secondary structure in water and methanol. ROESY spectra confirm that Lys-Pro and Arg-Pro peptide bonds are all-trans, but that a significant population of cis Arg-Pro bonds arises in aqueous solution, which increases in the environment of SDS micelles. The conformational ensemble of the peptide is observed to narrow as it becomes bound through its cationic mid-region to SDS micelles, with the accompanying advent of local extended structure. The overall results indicate the inherent conformational flexibility of neurotensin, and emphasize the environmental dependence of conformation in peptides of medium length.     

应用3H-TdR掺入法对SMMC-7721细胞株药敏试验条件的探讨与应用

目的探讨3H-TdR掺入法在进行SMMC-7721系细胞肿瘤药敏试验实验时的最佳实验条件.方法确定出3H-TdR掺入法药敏实验时的最适细胞浓度、最适实验药物浓度、3H-TdR掺入最适时间;在最适条件下采用3H-TdR掺入法测定肝癌SMMC-7721细胞株对临床常用的9种抗癌药物的敏感性.结果应用3H-TdR掺入法检测肝癌SMMC-7721细胞株药敏试验的最适实验细胞浓度为1×104个/孔,最适试验药物浓度为1×PPC,3H-TdR最适掺入时间为收集细胞前8 h;肝癌SMMC-7721细胞株对DDP、ADM、5-FU、CPT高度敏感,对MTX、VP-16、MMC、NVB低度敏感,对PYM不敏感.结论 3H-TdR掺入法可用于肿瘤药物敏感性测定并确定出它的最适实验条件.     

四种口服头孢菌素的体外抗菌活性的比较研究

本文报道口服Cefixime对900株需氧菌和厌氧菌的体外抗菌作用,并与其它口服的头孢克罗、头孢羟氨苄和头孢氨苄进行了比较。Cefixime对链球菌具较强作用,但对葡萄球菌的作用较差;对肠球菌无效。对肠杆菌科细菌的抗菌活性四者之中以Cefixime最强。Cefixime对头孢氨苄耐药株仍具高度活性,其MIC均值可降低60～1200倍以上。绿脓杆菌、硝酸盐阴性杆菌、鲁氏不动杆菌和产碱杆菌对Cefixime和其它3种头孢菌素的敏感性均较差。四者对厌氧菌的作用除厌氧球菌外其抗菌活性都很弱。Cefixime对β-内酰胺酶稳定,口服吸收良好,抗菌作用强,是一个值得进行临床研究的口服头孢菌素。     

Rb基因对人视网膜母细胞瘤株SO-Rb50的影响

目的：探讨视网膜母细胞瘤（Rb）的发生与Rb基因（Rb1）缺失、失活等异常的关系。方法：用逆转录病毒载体pDOR与全长4．7kb野生型Rb1cDNA构建逆转录病毒表达载体pDOR－Rb1+。用脂质体介导法将pDOR－Rb1+转入CRIP包装细胞系。结果：实验产生了0．5X105Cfu具有一次感染能力的重组逆转病毒。利用该病毒感染SO-Rb50，经G418筛选，获得了抗G418细胞群体。运用PCR及Southern杂交技术对转染细胞进行检测，结果表明该抗G418细胞中有完整的外源Rb1存在。Northern杂交发现其Rb1mRNA表达水平有所提高。对细胞群体生长速率、软琼脂集落形成能力的测定表明，外源Rb1的表达使SO-Rb50在软琼脂中集落形成能力降低，而群体生长速率无明显影响。结论：外源Rb1对SO-Rb50恶性表型有一定影响。     

持续压力超负荷对兔左心室细胞凋亡的影响

OBJECTIVE: To study the effect of chronic pressure overload on the apoptosis of the left ventricle myocytes in rabbits. METHODS: Rabbit models of chronic pressure overload-induced heart failure were prepared in which dynamic changes of apoptotic myocytes in the left ventricle were observed by way of terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) assay. RESULTS: Only a few apoptotic cells was observed in the sham-operated group, while in the experimental group, the apoptotic left ventricle myocytes significantly increased after operation, presenting a peak level between day 3 and day 7. Seven days after the operation, the apoptotic myocytes began to decrease and till day 14, the apoptotic cell number had been smaller than that measured on day 1. When signs of heart failure set in, the apoptotic myocytes were again increased (P<0.001). CONCLUSION: During chronic pressure overload, myocyte apoptosis in the left ventricle is elevated at the early stages and undulates subsequently, with the peak occurring before hypertrophy is obvious.     

The Receptor Role of Glycophorin A during the Invasion of Erythrocytes by Plasmodium Falciparum

Under the condition of in vitro culture,the effects of glycophorin A(GPA)of human erythrocyte membrane,anti-GPA IgG,α_1-acid glycoprotein,ovomucoid,wheat germ agglutinin,etc.on the invasion of erythrocytes by P.falciparum were observed in this study.The results show that the combination ofGPA with P.falciparum merozoites has high specificity,high affinity and atendency to saturation,and produces specific biological effects.These confirm forthe first time from the characteristics of the ligand-receptor interaction that GPAis a receptor recognized and bound by P.falciparum merozoites.     

山东省实现无脊髓灰质炎目标的卫生经济学评价(2)--成本-效益分析

为评价实现无脊髓灰质炎 (脊灰 )目标的卫生经济学效益 ,对山东省 195 6～ 2 0 0 0年的资料进行分析。结果显示 ,如不考虑无形效益和因口服脊灰疫苗 (OPV)免疫造成的疫苗相关麻痹型脊灰 (VAPP)的影响 ,按脊灰发病和控制的不同时期进行分析 ,推广使用疫苗时期 ( 196 5～ 1978年 )和无特异免疫预防时期 ( 195 6～ 196 4年 )相比 ,计划免疫时期 ( 1979～ 1990年 )和推广使用疫苗时期相比 ,消灭脊灰活动时期 ( 1991～ 2 0 0 0年 )和计划免疫时期相比 ,净效益 (以人民币元计算 ,下同 )分别为 3 6 4亿元、33 6 2亿元、80 83亿元 ,其成本 效益比分别为 1∶7 74、1∶47 6 9、1∶2 5 5 7。在 36年 ( 196 5～ 2 0 0 0年 )消灭脊灰进程中 ,共获得净效益 118 0 9亿元 ,其成本 效益比为 1∶2 6 90。消灭脊灰活动的效益是长久的 ,在全球消灭脊灰之后 ,随着成本投入的停止 ,其效益仍将持续增长。
