Microfiltration is the usual method of sterilization of tissue culture media. The filtrates of eight commercial filters were tested for embryotoxicity using mouse embryo growth. Four of five disk filters tested showed embryotoxicity in the first 5 ml of filtrate and the three largercapacity filters showed embryotoxicity for up to the first 10 ml of filtrate. The ethylene oxide-sterilized filters appeared to give poorer results than the gamma-irradiated filters. The importance of discarding the initial filtrate or preflushing such filters before use is emphasized. 相似文献

A 2,5-disubstituted tetrazole with p-nitrophenyl and 3-pyridyl units as acceptors (1a), and three push–pull tetrazoles with p-nitrophenyl as an acceptor and phenyl (1b), 2-(dibenzo[b,d]furan-4-yl) (1c), and 4-(N,N-diphenylamino)phenyl (1d) as donor groups, were synthesized by copper-catalyzed aerobic C–N coupling of p-nitrophenyl tetrazole with appropriately substituted aryl boronic acids. The absorption and emission spectra of 1a–c showed minimal dependence on the polarity of the solvent; however, in the case of 1d a blue shift was noted in the longest absorption band (λ1) as the polarity increased. The fluorescence intensity of the title compounds was found to be solvent-dependent; however, no apparent correlation to solvent polarity could be established. The absorption and emission characteristics of 1a–d were also influenced by the nature of the substituent as 1d, bearing a strong electron donating 4-(N,N-diphenylamino)phenyl group, displayed a significant red shifted absorption (λ1) as well as emission (λem) bands compared to other compounds. Time dependent density functional calculations (CAM-B3LYP/6-311++G**) revealed that the longest wavelength band (λ1) is associated with an intramolecular charge transfer (ICT) from HOMO/HOMO-1/HOMO-2 → LUMO/LUMO+1 in these molecules. The first hyperpolarizability values, βHRS, of 1a–d were measured using the solution-based hyper-Rayleigh scattering technique using a femtosecond Ti:Sapphire laser and the highest NLO activity was measured for 1d with the greatest push–pull characteristics. A strong correlation was observed between the calculated hyperpolarizability (βtot) and experimentally measured values (βHRS).A 2,5-disubstituted tetrazole with a p-nitrophenyl unit as an acceptor and a 4-(N,N-diphenylamino) phenyl group as a donor exhibits strong push–pull characteristics and displays high NLO activity. 相似文献

Five patients are described with acute primary ischemic iris atrophy with almost complete loss of the normal radial vessels of the iris and fine microneovascularization. The atrophy of the iris was marked with a thin atrophic stroma, widely dilated pupil, and extensive loss of pigment from the posterior surface of the iris. The condition is of unknown etiology but occlusion of the greater vascular circle of the iris is possible. Iris angiography showed an almost total loss of the radial vessels of the iris with some patchy microneovascularization. In recent cases, the onset was acute and all patients had a raised intraocular pressure (IOP) except one who had already undergone a drainage operation. There was an accompanying low-grade uveitis or cyclitis in the chronic stage in all cases. The condition was unilateral and the posterior segment of the eye was normal so that good visual function was possible if the accompanying cataract and glaucoma were appropriately managed. 相似文献

Rats were injected with a fractionated 50 ml/kg dose of Fluosol-DA 20% with and without exposure to 100% oxygen. Animals were killed at 24, 43, 92, and 183 days post-treatment and samples taken for hematological, hepatic enzyme, histological, and perfluorochemical analyses. There were no significant differences in hemograms or hepatic enzyme findings between treatment and control groups. Differences in organ weights and histology were a result of perfluorochemical (PFC) accumulation in the tissues of treated animals. All changes were reversible. Low bone mineral density (BMD) is a strong risk factor for low trauma fractures in the postmenopausal population without known chronic kidney disease (CKD). In stage 1?C3 CKD, low BMD can also be used to predict fracture risk with the gradient of risk similar to patients without CKD even though patients with stage 3 CKD have an approximate doubling of risk compared with age-matched patients without CKD. This greater risk of fracture in stage 3 CKD is not calculated in the current FRAX model. In stage 4?C5 CKD, BMD by dual-energy x-ray absorptiometry (DXA) is a poor predictor of fracture risk probably related to the severe derangements in bone metabolism in severe CKD, which alter bone quality and strength not measured by DXA. Serial BMD by DXA, however, may be useful in all stages of CKD to monitor for potential loss of BMD or effect of pharmacological agents to improve BMD. Newer radiological technologies, particularly high-resolution peripheral quantitative computerized tomography (HRpQcT) of the radius and tibia show promise to define the microstructural changes in bone that explain the greater risk of fracture observed in patients with CKD versus patients without CKD. BMD by DXA may still be of value across the spectrum of CKD, but physicians should realize its limitations and understand the greater risk of fracture in patients in all stages of CKD as compared to age-matched and BMD-matched patients without CKD. 相似文献

本文以人乳头状瘤病毒(HPV)16型-PBR_(322)重组质粒,对大肠杆菌HB101进行转化实验。经筛选、扩增、酶切、电泳鉴定。获得满意的电泳图谱及kb值,HPV-16Bam HI DNA为7.2kb,其260/280O.D比值为1.9-2。纯度合乎要求,可用以制备探针。以HPV-16DNA7.2kb片段为探针,应用~(32)P(α)-dATP标记进行了点杂交及Southern印迹杂交实验。结果表明38例宫颈癌组织DNA,其点杂交阳性率为76.3%,Southern杂交阳性率为65.7%。点杂交与Southern杂交结果基本符合。上述结果说明了在人宫颈癌组织中,存在着HPV-16相关的基因组,Southern杂交表明其以整合形式存在。说明宫颈癌的发病可能与HPV-16型的感染有密切关系。从分子水平进一步探讨了宫颈癌的病毒病因。 相似文献

The human colon adenocarcinoma cell line WiDr was xenografted and the tumor characterized. When athymic mice (NCR-nu) were inoculated with 10(6) cells, tumors appeared after 7-14 days with a 93-100% take rate and grew with an initial volume-doubling time of around 6 days. For optimizing the tumor disaggregation method, a comparison was made of two dissociation procedures and of different dissociation times. An enzyme cocktail (collagenase, DNase, pronase) resulted in total viable cell yields of 1-3 X 10(7) cells/g tumor tissue. Cell yield decreased with increasing tumor weight. Disaggregation with trypsin gave lower cell yields; and so, although the plating efficiencies (PEs) were higher, the enzyme cocktail was chosen for tumor disaggregation. On the basis of morphologic identification, cell suspensions prepared from WiDr tumors, by use of the enzyme cocktail for 2 hours, contained 49% malignant cells as well as a significant fraction of nonneoplastic cells. The major nonneoplastic host cell component was macrophage (33%); lymphocytes (13%) and granulocytes (5%) also were present. Host cells could be separated from neoplastic cells by centrifugal elutriation. By mixing various proportions of host and tumor cells, it was subsequently shown that the presence of host cells did not influence the malignant cell PE unless the cell suspensions contained greater than 90% host cells. Single-cell suspensions prepared from WiDr tumors, with use of the enzyme cocktail for 2 hours, were irradiated and then plated for survival (D0 = 1.5 Gy; n = 5) (D0, the 37% dose slope). A comparison was made of the sensitivity to radiation, after the different dissociation methods. The radiation sensitivities after 1.5-hour trypsinization and 2- and 6-hour enzyme cocktail administrations were similar, but after 0.5 hour of trypsin, the cells were more sensitive to radiation. 相似文献
