Wound healing is a complex process in which injured skin and tissues repaired by interaction of a complex cascade of cellular events that generates resurfacing, reconstitution and restoration of the tensile strength of injured skin. It follows β-catenin, extracellular signal regulated kinase (ERK) and Akt signaling pathways. Aegle marmelos L., generally known as bael is found to act as anti-inflammatory, antioxidant and anti-ulcer agent. Furthermore, studies have demonstrated that this Indian traditional medicinal plant, A. marmelos flower extract (AMF) was used for wound injury. Henceforth, the current study was investigated to ascertain the effect of its active constituents in vitro wound healing with mechanism involve in migration of cells and activation of β-catenin in keratinocytes, inhibition of PGE2 in macrophages and production of collagen in fibroblasts. We have taken full thickness wound of rats and applied AMF for 2 weeks. Cutaneous wound healing activity was performed using HaCaT keratinocytes, Hs68 dermal fibroblasts and RAW264.7 macrophages to determine cell viability, nitric oxide production, collagen expression, cell migration and β-catenin activation. Results shows that AMF treated rats demonstrated reduced wound size and epithelisation was improved, involved in keratinocytes migration by regulation of Akt signaling, beta-catenin and extracellular signal-regulated kinase (ERK) pathways. AMF and its active constituent’s increased mRNA expression, inhibited nitric oxide, PGE2 release, mRNA expression of mediators in RAW 264.7 macrophages and enhances the motility of HaCaT keratinocytes in vitro wound healing of rats.  相似文献

The BCR/ABL gene rearrangement is cytogenetically visualized in most chronic myeloid leukemia (CML) cases. About 5–10 % of CML patients lack its cytogenetic evidence, however, shows BCR/ABL fusion by molecular methods. We describe two CML patients with Philadelphia (Ph) negative (−ve) and BCR/ABL positive by fluorescence in situ hybridization (FISH). Both the cases were in chronic phase at diagnosis. Conventional cytogenetics and different FISH assays were adopted using BCR/ABL probes. Home-brew FISH assay using bacterial artificial clone (BAC) for BAC-CTA/bk 299D3 for chromosomal region 22q13.31-q13.32 was performed in case 1. Both the patients were Ph-ve. In first case, dual color dual fusion (DCDF)-FISH studies revealed 1 Red (R) 2 Green (G) 1 Fusion (F) signal pattern in 80 % of cells indicating BCR/ABL fusion signals on chromosomes 9 instead of Ph and 2G2F signal pattern in 20 % of cells indicating two BCR/ABL fusions on both chromosomes 9q34 on presentation. In second case, FISH studies revealed the 1R1G1F signal pattern indicating BCR/ABL fusion signals on chromosomes 9 instead of Ph in 100 % of cells at presentation. During follow-up, both the patients exhibited 2G2F signal pattern indicating two BCR/ABL fusions on both chromosomes 9q34, which indicated a clonal evolution in 100 % cells. Both the patients did not achieve therapeutic response. Relocation of BCR/ABL fusion sequence on sites other than 22q11 represents a rare type of variant Ph, the present study highlights the hot spots involved in CML pathogenesis and signifies their implications in Ph−ve BCR/ABL positive CML. This study demonstrated the genetic heterogeneity of this subgroup of CML and strongly emphasized the role of metaphase FISH, especially in Ph−ve CML cases, as it detects variations of the classical t(9;22).  相似文献

Biochemical and physiological alterations have been observed in the ovary and uterus of pregnant rats treated with an extract of Pueraria tuberosa DC. Its administration from day 1 to 5 post-coitally caused a significant increase in the wet weight, protein and glycogen contents. Activity of acid phosphatase and ATPase was depleted sharply on days 4 and 5, but the activity of alkaline phosphatase was elevated on days 4 and 5. Ovaries of treated rats showed an increased number of follicles of different age. Cells of the corpora lutea showed hypertrophy. Uterine histoarchitecture appeared nonreceptive on the day of implantation. No deciduoma was observed on day 5. These changes have been correlated with the antiimplantation action of the butanol extract.  相似文献

We have investigated the effect of shilajit on lipid peroxidation and glutathione content in rat liver homogenate. It inhibited lipid peroxidation induced by cumene hydroperoxide and ADP/Fe++ complex in a dose dependent manner. It also reduced the rate of oxidation of reduced glutathione content and inhibited ongoing lipid peroxidation, induced by these agents immediately after its addition to the incubation system.  相似文献

Ursolic acid isolated from the leaves and stems of Duboisia myoporoides (Solanaceae) was bioassayed by leaf disc method for feeding deterrence using Spilosoma obliqua and Spodoptera litura as test insects. This compound was proved to be a potent antifeedant under laboratory conditions. Azadirachtin was used as standard. Ursolic acid produced 90.12% and 91.96% inhibition at 5000 ppm concentration, respectively, against S. obliqua and S. litura .  相似文献

Administration of CCl4 at a dose of 0.1 mL/kg/day for 7 days to normal rats provoked significant alterations in the various biochemical variables. It also caused severe histopathological lesions. Priming with the butanol extract of P. tuberosa or conjoint administration of extract along with CCl4, provided significant protection against CCl4 induced injury.  相似文献
