建立同时测定茯苓不同部位中去氢土莫酸、猪苓酸C、3-表去氢土莫酸、去氢茯苓酸和茯苓酸含量的方法。采用紫外-波长转换检测的RP-HPLC法: 色谱柱为Kromasil C18柱 (250 mm × 4.6 mm, 5 μm); 流动相为乙腈 (A)-0.05%磷酸水溶液 (B) 系统, 梯度洗脱 (0~5 min, 60%A~64%A; 5~35 min, 64%A~65%A; 35~35.01 min, 65% A~73% A; 35.01~53 min, 73% A); 流速为1.0 mL·min−1; 检测波长为0~48 min, 241 nm (去氢土莫酸、猪苓酸C、3-表去氢土莫酸、去氢茯苓酸), 48~55 min, 210 nm (茯苓酸)。去氢土莫酸、猪苓酸C、3-表去氢土莫酸、去氢茯苓酸和茯苓酸分别在30.5~610.0 μg·mL−1 (r = 0.999 6)、12.66~253.2 μg·mL−1 (r = 0.999 5)、2.99~59.7 μg·mL−1 (r = 0.999 7)、6.13~122.5 μg·mL−1 (r = 0.999 5)、11.3~226.0 μg·mL−1 (r = 0.999 5) 内浓度与峰面积呈良好的线性关系; 方法回收率分别为98.5% (RSD = 1.9%)、99.4% (RSD = 1.7%)、97.9% (RSD = 1.2%)、96.7% (RSD = 2.5%) 和97.9% (RSD = 2.3%)。结果表明, 本方法简便、准确、重现性好, 为茯苓药材的质量控制提供了依据。 相似文献

目的:寻找金银花中的抗呼吸道病毒感染的成分,并进行定量研究。方法:采用各种柱层析和制备薄层层析等方法分离、细胞病变法进行抗病毒作用的研究,采用RP-HPLC方法进行定量方法的研究。结果:分离得到了13种咖啡酰奎宁酸类化合物和咖啡酸、咖啡酸甲酯,确认咖啡酰奎宁酸类成分具有较强的抗呼吸道病毒作用,并采用HPLC方法测定了金银花中的 6种咖啡酰奎宁酸类成分和咖啡酸的含量。结论:咖啡酰奎宁酸类成分为金银花中的有效成分,定量方法准确、重复性好。 相似文献

Deoxynivalenol (DON) is a kind of Fusarium toxin that can cause a variety of toxic effects. Oxidative stress and DNA damage play a critical role in the toxicity of DON. However, previous studies focused more on acute toxicity in vivo/vitro models and lacked subchronic toxicity study in vivo. The potentially harmful effect of DON given at doses comparable to the daily human consumption in target organs, especially the liver, which is the main detoxification organ of DON, is also still not fully understood. Otherwise, Heme Oxygenase-1 (HO-1) has also reduced cell damage under the DON condition according to our previous study. Therefore, we used a rodent model that mimicked daily human exposure to DON and further explored its mechanism of toxic effects on liver tissue and Hepa 1–6 cell line. We also used adeno-associated virus (AAV)-modified HO-1 expressing by tail vein injection and constructed lentivirus-Hepa 1–6 cell line for mimicking HO-1 protective ability under the DON condition. The main results showed that both 30 d and 90 d exposures of DON could cause low-grade inflammatory infiltration around hepatic centrilobular veins. The reactive oxygen species (ROS) and 8-hydroxy-2 deoxyguanosine (8-OHdG) increased during DON exposure, indicating oxidation stress and DNA damage. Significantly, AAV-mediated liver-specific overexpression of HO-1 reduced DON-induced liver damage and indirectly protected the abilities of antioxidant enzyme/DNA damage repair system, while AAV-mediated silence of HO-1 produced the opposite effect. In addition, we found that overexpression of HO-1 could enhance autophagy and combined it with an antioxidant enzyme/DNA damage repair system to inhibit DON-induced hepatocyte damage. Altogether, these data suggest that HO-1 reduces the oxidative stress and DNA damage caused by DON sub-chronic exposure through maintaining DNA repair, antioxidant activity, as well as autophagy. 相似文献

背景与目的: 研究双酚A(BPA)和敌匹硫磷单独和联合作用对MCF-7细胞增殖的影响。 材料与方法: 设BPA、敌匹硫磷1.0×10-12~1.0×10-5 mol/L 的不同浓度组(组间10倍浓度差),分别作用雌激素敏感型与不敏感型MCF-7细胞,观察其对细胞增殖的影响,及分别加入雌激素受体抑制剂ICI182780后,对细胞增殖影响的变化。实验并设立溶剂对照组。BPA和敌匹硫磷在1.0×10-8、5.0×10-9和2.5×10-9 mol/L 浓度时,两者联合作用MCF-7细胞,采用2×2析因设计分析对其增殖的影响。 结果: BPA和敌匹硫磷在1.0×10-8 mol/L时促增殖效应均达到最大,相对增殖率分别为2.473和2.167,与溶剂对照组比较差异均具有统计学意义(P<0.05);加入ICI182780后,二者对MCF-7的促增殖作用均减弱;二者在各浓度组对雌激素不敏感型MCF-7细胞均无明显促增殖效应;BPA和敌匹硫磷在1.0×10-8、5.0×10-9和2.5×10-9 mol/L各浓度组均表现出明显拮抗作用。 结论: BPA和敌匹硫磷具有明显的拟雌激素活性,并且通过雌激素受体途径对细胞发挥促增殖效应,二者的联合作用为拮抗作用。 相似文献

目的观察和探讨铅对小鼠脑皮质区蛋白激酶C-γ(PKC-γ)及细胞外信号调节激酶(ERK)蛋白表达的影响。方法小鼠出生1d起,染铅组母鼠开始通过饮水饲以不同浓度醋酸铅2.4,4.8,9.6mmol/L。幼鼠出生后,先通过哺乳接触铅,断乳后则自行饮用与母鼠饮用浓度相同的含铅水。分别在14,21,28,35d处死小鼠,用蛋白免疫印迹法观察各组小鼠脑皮质区PKC-γ蛋白表达状况;分别在7,14,21,28d处死小鼠,用蛋白免疫印迹法观察各组小鼠脑皮质区ERK蛋白表达状况。结果慢性铅暴露对小鼠脑皮质区PKC-γ蛋白表达总体上呈现下降趋势,各染铅组中,PKC-γ蛋白表达与相应期对照组相比差异均有统计学意义(P<0.05)。低浓度铅2.4mmol/L使ERK表达升高,此后随铅浓度升高ERK表达量逐渐降低,铅浓度升高至9.6mmol/L时,随着铅浓度升高,ERK表达量不再降低,相反有所回升。结论铅扰乱小鼠脑皮质区PKC-γ及ERK蛋白正常表达。 相似文献

目的:研究运动干预对肥胖男性青少年血管生成素样蛋白8(ANGPTL8)与血管内皮功能的影响,探讨ANGPTL8在血管内皮功能变化中的可能作用。方法:选取42名肥胖男性青少年作为实验组(EG组),年龄15.9±2.25岁,体质指数(body mass index,BMI)为35.27±3.98 kg/m2,对其进行6周运动干预。测定运动干预前后的身体形态、ANGPTL8、血脂和血管内皮功能水平,分析ANGPTL8与不同血脂状态下肥胖青少年血管内皮功能的关系。另招募10名体重正常的男性青少年作为对照组(CG组),不接受运动干预。结果:与干预前相比,肥胖男性青少年6周运动干预后身体形态指标、血脂指标和ANGPTL8水平均显著降低(其中腰臀比P<0.05,其余均P<0.01),血管内皮功能显著改善(P<0.01)。36名血脂异常的肥胖青少年运动干预前后的ANGPTL8变化与Δ体重(r=0.422,P<0.01)、ΔBMI(r=0.426,P<0.01)、Δ低密度脂蛋白胆固醇(LDL-C)(r=0.381,P<0.05)、Δ甘油三酯(TG... 相似文献

Girdin has been proven to play a vital role in the process of proliferation, apoptosis, and invasion in various cancer cells, yet the underlying molecular mechanism in primary hepatocellular carcinoma (HCC) has not yet been clarified. Thereafter, we performed immunohistochemistry to detect the expression of Girdin in 40 primary HCC tissues and 30 matched adjacent tissues using hepatic carcinoma tissue microarray. Our data showed that the positive expression rate of Girdin in hepatocellular carcinoma tissues was 67.5%, higher than that found in adjacent tissues of 16.7% (P < 0.05). It closely correlates to tumor size, T stage, TNM stage and Edmondson-Steiner stage (P < 0.05) of HCC patients. After specific small interfering RNA of Girdin was transfected into HepG2 and Huh7.5.1 cells, the proliferation and invasion ability of tumor cells were significantly inhibited. In summary, we suggest that the oncogenic role of Girdin could provide new molecular target for the treatment of HCC. 相似文献

The purpose of this paper is to examine the effects of polydatin on cognitive function in rats self-administered with chronic ethanol levels. The levels of cyclin-dependent kinase 5 (Cdk5) were also determined. In the in vivo study, adult male Sprague-Dawley rats were used to establish an ethanol-administered rat model. Cognitive function was measured using the Morris water maze and the level of Cdk5 expression was measured to evaluate the effect of polydatin treatment. Cdk5 kinase activity and cell survival rate in primary hippocampal neuron cultures treated with ethanol or ethanol and polydatin were measured in the in vitro study. Polydatin reversed the performance impairments in chronic ethanol treated rats in Morris water maze test, and decreased unregulated Cdk5 expression. Moreover, polydatin increased cell survival rate, and decreased Cdk5 activity in the ethanol-treated primary culture of hippocampal neurons. The study results suggest that polydatin exhibits neuroprotective potential for ethanol induced neurotoxicity, both in vivo and in vitro, which is most likely related to its ability to target Cdk5 in neurons. 相似文献
