Physical environment influences the development and maintenance of musculoskeletal tissues. The current study uses an animal model to explore the role of the physical environment on the postnatal development of the supraspinatus tendon enthesis. A supraspinatus intramuscular injection of botulinum toxin A was used to paralyze the left shoulders of mice at birth. The supraspinatus muscles of right shoulders were injected with saline to serve as contralateral controls. The supraspinatus enthesis was examined after 14, 21, 28, and 56 days of postnatal development. Histologic assays were used to examine fibrocartilage morphology and percentage osteoclast surface. Micro-computed tomography was used to examine muscle geometry and bone architecture. At 14 days there were no differences between groups in fibrocartilage formation, muscle geometry, bone architecture, or osteoclast surface. When comparing groups at 21, 28, and 56 days, muscle volume was decreased, fibrocartilage development was delayed, mineralized bone was decreased, and osteoclast surface was higher at each timepoint in the botulinum group compared to the contralateral saline control group. Our results indicate that the development of the tendon enthesis is sensitive to its mechanical environment. A reduction in muscle loading delayed the development of the tendon-to-bone insertion site by impeding the accumulation of mineralized bone. Physical factors did not play a significant role in enthesis maturation in the first 14 days postnatally, implying that biologic factors may drive early postnatal development.  相似文献

This study aims at reviewing the 25 years (1975-2000) since the first graduate program was offered by the University of S?o Paulo at Ribeir?o Preto College of Nursing based on a critical reflection about the creation and development of graduate programs. The methodology applied was a quantitative assessment regarding the scientific production published in the literature about the theme. Results showed that the graduate programs were consolidated in the 1990s, when three doctoral programs were created.: Psychiatric Nursing, Fundamental Nursing and Public Health Nursing. These accomplishments require an increase in international visibility.  相似文献

Xenobiotic metabolizing enzymes are involved in the detoxification of many carcinogens and may be important in modulating cancer susceptibility. CYP1A1, CYP2E1, GSTM3, and NAT2 polymorphisms were determined in peripheral blood DNA of 231 oral cancer patients and 212 hospital controls in Rio de Janeiro, Brazil, using the PCR-RFLP technique. NAT2 polymorphism distribution was different between cases and controls (P=0.035), with an overrepresentation of NAT2( *)11 mutant allele in controls. Risk analysis showed that NAT2 4/4 individuals (OR=1.95, 95% CI=1.05-3.60) and combined GSTM3 and NAT2 heterozygotes (OR=1.94, 95% CI=1.04-3.66) were at increased oral cancer risk. No statistically significant association was observed for CYP1A1 and CYP2E1 polymorphisms. Our results suggest that NAT2 polymorphism, alone or combined with GSTM3, may modulate susceptibility to oral cancer in Rio de Janeiro.  相似文献

Airway wall thickening and mucus plugging are important characteristics of cystic fibrosis (CF) lung disease in the first 5 years of life.The aim of this study is to investigate the association of lung disease in preschool children (age, 2‐6) with bronchiectasis and other clinical outcome measures in the school age (age >7). Deidentified computed tomography‐scans were annotated using Perth‐Rotterdam annotated grid morphometric analysis for CF. Preschool %disease (a composite score of %airway wall thickening, %mucus plugging, and %bronchiectasis) and %MUPAT (a composite score of %airway wall thickening and %mucus plugging) were used as predictors for %bronchiectasis and several other school‐age clinical outcomes. For statistical analysis, we used regression analysis, linear mixed‐effects models and two‐way mixed models. Sixty‐one patients were included. %Disease increased significantly with age (P < .01). Preschool %disease and %MUPAT were significantly associated with school‐age %bronchiectasis (P < .01 and P < .01, respectively). No significant association was found between preschool %disease and %MUPAT and school‐age forced expiratory volume 1 (FEV1%) predicted and quality of life (P > .05). Cross‐sectional, %disease in school‐age was associated with a low FEV1% predicted and low quality of life (P = .01 and P = .007, respectively). %Disease can be considered an early marker of diffuse airways disease and is a risk factor for school‐age bronchiectasis.  相似文献

Human tau seeding and spreading occur following intracerebral inoculation of brain homogenates obtained from tauopathies in transgenic mice expressing natural or mutant tau, and in wild‐type (WT) mice. The present study was geared to learning about the patterns of tau seeding, the cells involved and the characteristics of tau following intracerebral inoculation of homogenates from primary age‐related tauopathy (PART: neuronal 4Rtau and 3Rtau), aging‐related tau astrogliopathy (ARTAG: astrocytic 4Rtau) and globular glial tauopathy (GGT: 4Rtau with neuronal deposits and specific tau inclusions in astrocytes and oligodendrocytes). For this purpose, young and adult WT mice were inoculated unilaterally in the hippocampus or in the lateral corpus callosum with sarkosyl‐insoluble fractions from PART, ARTAG and GGT cases, and were killed at variable periods of three to seven months. Brains were processed for immunohistochemistry in paraffin sections. Tau seeding occurred in the ipsilateral hippocampus and corpus callosum and spread to the septal nuclei, periventricular hypothalamus and contralateral corpus callosum, respectively. Tau deposits were mainly found in neurons, oligodendrocytes and threads; the deposits were diffuse or granular, composed of phosphorylated tau, tau with abnormal conformation and 3Rtau and 4Rtau independently of the type of tauopathy. Truncated tau at the aspartic acid 421 and ubiquitination were absent. Tau deposits had the characteristics of pre‐tangles. A percentage of intracellular tau deposits co‐localized with active (phosphorylated) tau kinases p38 and ERK 1/2. Present study shows that seeding and spreading of human tau into the brain of WT mice involves neurons and glial cells, mainly oligodendrocytes, thereby supporting the idea of a primary role of oligodendrogliopathy, together with neuronopathy, in the progression of tauopathies. In addition, it suggests that human tau inoculation modifies murine tau metabolism with the production and deposition of 3Rtau and 4Rtau, and by activation of specific tau kinases in affected cells.  相似文献

Goethite–titania (α-FeOOH–TiO2) composites were prepared by co-precipitation and mechanical milling. The structural, morphological and optical properties of as-synthesized composites were characterized by X-ray powder diffraction, scanning electron microscopy and UV-Vis diffuse reflectance spectroscopy, respectively. α-FeOOH–TiO2 composites and TiO2-P25, as reference, were evaluated as photocatalysts for the disinfection of Escherichia coli under UV or visible light in a stirred tank reactor. α-FeOOH–TiO2 exhibited better photocatalytic activity in the visible region than TiO2-P25. The mechanical activation increased the absorption in the visible range of TiO2-P25 and the photocatalytic activity of α-FeOOH–TiO2. In the experiments with UV light and α-FeOOH–TiO2, mechanically activated, a 5.4 log-reduction of bacteria was achieved after 240 min of treatment. Using visible light the α-FeOOH–TiO2 and the TiO2-P25 showed a 3.1 and a 0.7 log-reductions at 240 min, respectively. The disinfection mechanism was studied by ROS detection and scavenger experiments, demonstrating that the main ROS produced in the disinfection process were superoxide radical anion, singlet oxygen and hydroxyl radical.

A photocatalytic mechanism for FeOOH–TiO2 composite is proposed under UV-Vis light, the FeOOH–TiO2 composite showed higher photocatalytic activity than TiO2-P25.  相似文献
