1. Prostaglandin E(1) increases sodium transport as measured by short circuit current (SCC) across isolated frog skin whereas calcium, added to the external Ringer fluid, decreases sodium transport. To help establish the site of action of prostaglandin the possible interaction of these two agents on sodium transport has been examined.2. The effect of a standard dose of prostaglandin (0.5 x 10(-6)M) on the short circuit current was tested on paired skins with either zero or high calcium (22.4 mM) in the external Ringer fluid. In ten experiments the responses to prostaglandin (expressed in muA/cm(2)) were not significantly affected by external calcium.3. In another series of experiments the chelating agent, EGTA, was included in calcium-free external Ringer in order to promote greater depletion of skin calcium. The response of these skins to the standard dose of prostaglandin was of the same order of magnitude as that of control skins. The response was not sustained in contrast to that of normal skins and skins in high-calcium fluids.4. In a further series of experiments the reverse procedure was adopted whereby the response of the skin to low and high doses of calcium in the external Ringer was recorded in control conditions and when the skin had responded fully to twice the standard dose of prostaglandin. In addition, the calcium-sensitive current was calculated for each skin in both circumstances. The latter was unchanged on addition of prostaglandin, and graded doses of calcium caused the same degree of inhibition of the short circuit current.5. The results show no interaction between external calcium and prostaglandin and also no need for external calcium in prostaglandin stimulation of sodium transport.6. The findings do not support the concept of chelation by prostaglandin of calcium from critical sites on the skin as the primary mechanism of its action on sodium transport. The results closely parallel those of a similar type of study into the relationship between vasopressin and external calcium on frog skin also.7. When frog skin has responded fully to either prostaglandin E(1) or vasopressin, it shows no response to the other, although removal of calcium from the external Ringer fluid causes a further increase in short circuit current.8. Vasopressin causes a further increase in short circuit current in skins treated with prostaglandin F(1alpha). Prostaglandin F(1alpha) may be a weaker agonist on frog skin than either vasopressin or prostaglandin E(1).  相似文献

Purified peptidoglycans from various gram-negative bacteria can substitute for mycobacteria in Freund's type adjuvant. The effect of adjuvants on the production of delayed hypersensitivity in the guinea pig was studied with azobenzenearsonate? N? acetyl? L ? tyrosine as antigen. It was found that only a few μg of peptidoglycan per animal were needed. An adjuvant effect on antibody response to egg albumin was also obtained.  相似文献

Acute local gamma irradiation of porcine skin induces, as in human skin, an extensive and mutilating sclerosis characterized by continuous expansion of the fibrosis invading the adjacent muscle and by accumulation of the macromolecular components of the extracellular matrix. Collagen synthesis, content, and types were studied in the presence of heparin fragments (100 micrograms/10(6) cells) in the culture medium, by measuring the incorporation of the radiolabeled precursor [3H]proline into confluent primary cultures of porcine fibroblasts obtained from normal and irradiated fibrotic dermis. Enhancement in collagen biosynthesis and deposition and preferential increase in collagen type III synthesis were observed in fibrotic fibroblast cultures when compared to those in normal dermis fibroblasts. The total collagen synthesis and the rate of collagen hydroxylation appear unmodified by heparin fragments both in normal and in fibrotic fibroblast cultures. But heparin fragments induce a 10- and 2-fold decrease, respectively, in collagen type III and type V syntheses by fibrosis fibroblasts. As only minor effects upon collagen type III and V are observed in cultures of normal dermis fibroblasts, these results highly suggest that heparin fragments are capable of specifically modulating the collagen phenotype of fibroblasts derived from radiation-induced dermis fibrosis and thus are able to regulate the fibrotic process.  相似文献

The quantitation of CAP57, a highly hydrophobic, native cationic antigen of human polymorphonuclear leukocytes has been achieved using ELISA. An important feature determining the sensitivity and precision of the ELISA was the reduction of non-specific protein-protein binding, particularly in the inhibition assays, thus eliminating high backgrounds obtained with presently available methodology. Washing of the solid phase-bound antigen and blocking of the non-specific binding sites using a potassium phosphate buffer containing heparin largely contributed to this increased sensitivity. The inhibition assays were conducted using antigen concentrations over the range of 0.9-120 ng. The assay is highly specific and can be performed using monoclonal antibodies and polyclonal antibodies. Non-specific reactions were observed only when high concentrations of antigen (greater than 100 ng) were present in the inhibition mixture. The technique as described is extremely simple, highly reproducible and could be of value in the detection of cationic antimicrobial proteins in the clinical setting in the future.  相似文献

Synthesis and characterization of methacrylate-monofunctionalized crown ether compounds based on derivatives of 2-hydroxymethyl-1,4,7,10,13-pentaoxacyclopentadecane and 4′-hydroxymethyl-1,4,7,10,13-pentaoxabenzocyclopentadecane is described. By differential scanning calorimetry (DSC) and thermooptical analysis (TOA) it is shown that the synthesized compounds and their sodium triflate complexes exhibit columnar mesophases.  相似文献

Two experiments investigated selective processing as a result of transient and sustained visual-spatial attention. In Experiment 1, attention was cued on a trial-by-trial basis and event-related brain potentials were measured to stimuli preceded by valid, invalid, or neutral symbolic precues. Trial validity had only small effects on posterior PI and N1 components. At midline electrodes, an enhanced negativity for valid as compared with invalid trials was present, which appeared to reflect initial processing costs for invalid trials followed by an additional processing benefit for valid trials. Experiment 2 investigated whether these effects are specific for transient spatial attention by comparing transient and sustained attention conditions. No indication of early processing costs was found in the latter case.  相似文献

Leu-19 antigen, which seems to be identical with neural cell adhesion molecule (N-CAM), plays a major role in the innervation of muscle cells, and in adult muscle appears after denervation and during regeneration of muscle fibres, where it acts as part of a signalling system increasing the probability of re-innervation. This combined enzyme-histochemical and immunohistochemical study examined whether this signalling process was regulated in a uniform or differential pattern for type 1 and type 2 muscle fibres. The subscapular nerve of 18 rabbits was transsected with subsequent complete denervation of the supraspinatus muscle. Leu19 and N-CAM immunohistochemistry was performed 2 to 64 days after surgery. Whereas in normal muscle there are virtually no Leu-19/N-CAM positive muscle fibres; from day 2 after denervation an increasing proportion of fibres expressed Leu-19/N-CAM, prior to any neurogenic atrophy. In the early stage of denervation Leul9/N-CAM expression was confined to type 1 fibres. After 11 days nearly all fibres were Leul9/N-CAM positive irrespective of their fibre type. Sixty-four days after denervation type 1 fibres became Leu19/N-CAM negative, while atrophic type 2 fibres showed intensive staining. Thus, expression of Leu-19 antigenity is differently regulated in both fibre types.  相似文献
