Nuclear factor-kappaB (NF-kappaB) plays a key role in inflammation, which is involved in the development of cerebral vasospasm after subarachnoid hemorrhage (SAH). In the present study, we assessed the potential role of NF-kappaB in regulation of cerebral vasospasm. Nuclear factor-kappaB DNA-binding activity was measured in cultured vascular smooth muscle cells (VSMCs) treated with hemolysate and pyrrolidine dithiocarbamate (PDTC, 80 micromol/L), an inhibitor of NF-kappaB. Forty-two rabbits were divided into three groups: control, SAH, and PDTC groups (n=14 for each group). The caliber of the basilar artery was evaluated. Nuclear factor-kappaB DNA-binding activity and the gene expression levels of cytokines and adhesion molecules in the basilar artery were measured. Immunohistochemical study was performed to assess the expression and localization of tumor necrosis factor (TNF)-alpha, intercellular adhesion molecule (ICAM)-1, and myeloperoxidase (MPO). It was observed that NF-kappaB DNA-binding activity was significantly increased by treatment with hemolysate in cultured VSCMs, but this increase was suppressed by pretreatment with PDTC. Severe vasospasm was observed in the SAH group, which was attenuated in the PDTC group. Subarachnoid hemorrhage could induce increases of NF-kappaB DNA-binding activity and the gene expression levels of TNF-alpha, interleukin (IL)-1 beta, ICAM-1, and vascular cell adhesion molecule (VCAM)-1, which were reduced in the PDTC group. Immunohistochemical study demonstrated that the expression levels of TNF-alpha, ICAM-1, and MPO were all increased in the SAH group, but these increases were attenuated in the PDTC group. Our results suggest that NF-kappaB is activated in the arterial wall after SAH, which potentially leads to vasospasm development through induction of inflammatory response.  相似文献

Transiliac bone biopsies were obtained from 55 women treated with teriparatide or placebo for 12-24 months. We report direct evidence that modeling bone formation at quiescent surfaces was present only in teriparatide-treated patients and bone formation at remodeling sites was higher with teriparatide than placebo. INTRODUCTION: Recombinant teriparatide [human PTH(1-34)], a bone formation agent for the treatment of osteoporosis when given once daily subcutaneously, increases biochemical markers of bone turnover and activation frequency in histomorphometry studies. MATERIALS AND METHODS: We studied the mechanisms underlying this bone-forming action of teriparatide at the basic multicellular unit by the appearance of cement lines, a method used to directly classify surfaces as modeling or remodeling osteons, and by the immunolocalization of IGF-I and IGF-II. Transiliac bone biopsies were obtained from 55 postmenopausal women treated with teriparatide 20 or 40 microg or placebo for 12-24 months (median, 19.8 months) in the Fracture Prevention Trial. RESULTS: A dose-dependent relationship was observed in modeling and mixed remodeling/modeling trabecular hemiosteons. Trabecular and endosteal hemiosteon mean wall thicknesses were significantly higher in both teriparatide groups than in placebo. There was a dose-dependent relationship in IGF-II immunoreactive staining at all bone envelopes studied. The greater local IGF-II presence after treatment with teriparatide may play a key role in stimulating bone formation. CONCLUSIONS: Direct evidence is presented that 12-24 months of teriparatide treatment induced modeling bone formation at quiescent surfaces and resulted in greater bone formation at remodeling sites, relative to placebo.  相似文献

Polytetrafluoroethylene (PTFE) microvascular prostheses with a fibril length of 30 microns were pretreated with alcohol (n = 18), implanted into the abdominal aorta of rats and were evaluated at 1 day (n = 3), 1 week (n = 3), 3 weeks (n = 6) and 6 weeks (n = 6) to determine whether alcohol-pretreatment might improve their healing. Untreated PTFE microvascular prostheses (n = 18) functioned as controls (all prostheses: length 10 mm, I.D. 1.5 mm). The alcohol-pretreated PTFE microarterial prostheses were initially completely filled and covered with clot and showed fast and complete healing within 6 weeks: endothelial cells and smooth muscle cells on the luminal surface, and the interstices filled with fibrous-like tissue. In contrast, the untreated PTFE prostheses were initially not filled or covered with clot and showed only healing near the anastomotic sites and scarce tissue ingrowth into the wall. These results demonstrate that alcohol-pretreatment improves the healing characteristics of PTFE microarterial prostheses with a fibril length of 30 microns. The alcohol-pretreatment renders the PTFE material more accessible to clot and subsequently to cells.  相似文献

A slow-moving gamma chain variant was discovered in the cord blood of an infant born to parents of the Han nationality from Jiangsu. The variant, which migrated electrophoretically at alkaline pH between Hb A and Hb D, was characterized by an Asp----His substitution at gamma 73, while residues gamma 75 and gamma 136 were occupied by Ile and Ala, respectively. As the A gamma T-chain was also present, this baby has two types of abnormal gamma-chain. The variant was named Hb F-Xin-Su.  相似文献

通过对重建图像噪声特性的分析,间接证明SPECT的解具有多样性。分别用TretiakMetz、Gulberg、富谷武浩等推导的3种等价重建方法和Belini等推导的重建方法,经计算机模拟重建衰减均匀分布的图像,先预置相同的重建图像的空间分辨率,再比较它们的噪声特性。两种方法重建图像的噪声分布:前者近图像边缘时增大,而后者减小,从而间接证明在假定均匀衰减条件下,SPECT的重建有多解性。  相似文献

Thirty-one patients who received a prolonged maintenance therapy with cordarone to treat and prevent cardiac arrhythmias underwent an ophthalmologic examination. The treatment lasted 1.5 to 51 months. The signs of keratopathy were detected in 24 (77.4%) patients using a slit lamp biomicroscopy. There is a relationship between the severity of keratopathy and a daily dose of cordarone: in most cases signs of stage I keratopathy were detectable when the daily dose was up to 200 mg. Keratopathy appears to be reversible after the drug has been discontinued.  相似文献

伽玛刀治疗垂体大腺瘤临床分析(附30例报告)   总被引:2,自引:0,他引:2  
目的 研究伽玛刀治疗垂体大腺瘤的效果及影响疗效的因素。方法 30例垂体大腺瘤病人用1.0T磁共振定位,Gamma-plan计划系统规划,等剂量曲线45%~60%(平均49.7%),周边剂量12~35Gy(平均22.9Gy),中心剂量24~70Gy(平均45.4Gy).Leksell伽玛刀实施放射外科治疗。分析治疗后垂体大腺瘤病例,总结其疗效和影响疗效的因素。结果 随访12~60个月,平均28个月,肿瘤完全消失6例(20%),肿瘤缩小19例(63.3%),无变化5例(16.7%);19/20(95.5%)功能性垂体腺瘤内分泌症状改善,临床症状好转,1例过早绝经(年龄36岁);5/10(50%)无功能性垂体腺瘤体积缩小.症状改善。结论 伽玛刀对于垂体大腺瘤仍是一种有效的治疗方法,其疗神与腺瘤的举类型、体积、照射剂量、剂量体剂、MRI信号特征密切相姜。  相似文献
