Recently, a novel 12-mer B-cell epitope, MKNNPIYSEGSL, in the type-specific region of equine herpesvirus 1 (EHV-1) glycoprotein G (gG) was identified and used as an antigen for enzyme-linked immunosorbent assay (Maeda et al., J. Clin. Microbiol. 42:1095-1098, 2004). Although our prototype strain, TH20p, possesses two repeat sequences containing the B-cell epitope, the EHV-4 NS80567 strain has two repeat sequences that are not identical. One repeat sequence stretch contained the B-cell epitope, while the other contained the 11-mer, MKNNPVYSESL (underlining indicates a different amino acid). In this study, heterogeneity of the type-specific region was compared among Japanese EHV-4 isolates. The 11-mer peptide, MKNNPVYSESL, specifically reacted with sera from horses naturally infected with EHV-4 but not with sera from horses experimentally infected with EHV-4 TH20p. The 11-mer peptide may be another B-cell epitope in the type-specific region.  相似文献

BACKGROUND: Progression of hypertrophic cardiomyopathy (HCM) to left ventricular dilatation and systolic dysfunction sometimes occurs. However, the mechanism is not known. We examined whether oxidative stress was elevated in myocardia of HCM patients and whether the levels were correlated with left ventricular dilatation and systolic dysfunction. METHODS AND RESULTS: Endomyocardial biopsy samples obtained from the right ventricular side of the septum of 31 patients with HCM, and 10 control subjects were studied immunohistochemically for the expression of 4-hydroxy-2-nonenal (HNE)-modified protein, which is a major lipid peroxidation product. Expression of HNE-modified protein was found in all myocardial biopsy samples from patients with HCM. Expression was distinct in the cytosol of cardiomyocytes. The expression levels in patients with HCM were significantly increased compared with those in control subjects (P = .0005). The expression levels in patients with HCM were correlated with left ventricular end-diastolic diameter (r = 0.483, P = .0053) and end-systolic diameter (r = 0.500, P = .0037) determined by echocardiography. The expression levels were inversely correlated with left ventricular ejection fraction determined by left ventriculography (r = -0.640, P = .0001). CONCLUSION: Oxidative stress was elevated in myocardia of HCM patients and the levels were correlated with left ventricular dilatation and systolic dysfunction. Oxidative stress is involved in the pathogenesis of heart failure in patients with HCM.  相似文献

Interleukin-18 (IL-18) is a potent cofactor for T-helper (Th-1) cell development and inducer of cytotoxic T lymphocytes (CTL), and is reported to contribute to autoimmune diseases. T-cell large granular lymphocyte (T-LGL) leukemia involves the proliferation of autoreactive CTL that is often associated with autoimmune disorders. We found increased serum IL-18 concentrations in a 55-year-old woman with systemic lupus erythematosus (SLE) and T-LGL-leukemia. Her serum IL-18 concentrations correlated with the intensity of her SLE symptoms and the number of T-LGL cells in peripheral blood. This evidence suggests that IL-18 is involved in T-LGL-related autoimmune disorders.  相似文献

In the field isolate, 91-58, of feline herpesvirus type 1 (FHV-1), one of the major immunogenic proteins was found to have different molecular masses of 75 and 130 kDa from those in the other field isolates (Maeda et al., J Vet Med Sci 57, 147-150, 1995). Immunoblot analysis using monoclonal antibodies (MAbs) indicated that the protein is glycoprotein C (gC). The gC gene of 91-58 was amplified by polymerase chain reaction (PCR) and shown to have an inserted fragment of approximately 160 base pairs (bp). Restriction endonuclease analysis of the PCR product with various restriction enzymes was carried out, indicating that the insertion located within 262 bp between Eco RV and DraI sites. Nucleotide sequence analysis indicated that the inserted fragment was 156 bp encoding 52 amino acids and composed repeat sequences. Next, five recent isolates were also examined by immunoblot analysis using anti-FHV-1 cat serum or MAbs. The result showed that one isolate, 98-064, also had the gC with different molecular weights. PCR and nucleotide sequence analyses indicated that 98-064 had an inserted sequence of 78 bp at the corresponding region identified in the gC gene of 91-58, although the inserted sequence was different from that of 91-58. These results indicated that some of FHV-1 isolates had the genetic rearrangements in the gC gene and detection of such mutations would be useful for differentiation among FHV-1 field isolates.  相似文献

Cell-based therapy has been the recent focus of attention for repairing injured organs. Several cell sources, derived from peripheral blood, bone marrow, or embryonic stem cells, have been used to induce angiogenesis successfully in various experimental ischemic models, which might be related to angiogenic cytokine production and endothelial incorporation from implanted cells within the targeted ischemic tissues after implantation. Clinical trials have also reported the feasibility and found some efficacy of therapeutic angiogenesis induced by the implantation of autologous bone marrow-(or peripheral blood)-derived cells in patients with ischemic heart disease and peripheral artery disease. Although many questions regarding the effectiveness and safety, optimal cell number and route of delivery, and mechanisms remain, cell-based therapeutic angiogenesis may be a novel and promising treatment option for ischemic disease.  相似文献

We retrospectively studied the efficacy of intravenous and intravenous-drip infusion administration of lidocaine on seizures treated in Saitama Children's Medical Center during the period of 1997-2002. Thirty eight seizures of 29 children (12 boys and 17 girls, average age 41.4 months) were treated with lidocaine. The overall rate of effectiveness was 53%. The response to lidocaine appeared within 5 minutes in all episodes. Good response to lidocaine was noted, in 36% of patients with epilepsy, in 72% with CNS infection and in 80% with benign infantile convulsions; in 42% with generalized seizures and in 63% with partial seizures. Thus there was no significant difference in efficacy among the underlying disorders and seizure types. The adverse reaction of SpO2 decrease was found in one patient. We conclude that lidocaine can be used as a second-line, anti-convulsive drug in status epilepticus based on its prompt effectiveness and low risk of side effects.  相似文献

We gave intrapleural perfusion hyperthermo-chemotherapy to a 72-year-old woman in whom malignant pleural effusion developed after surgery for primary cancer. This procedure involved irrigating the pleural space for 2 hours with a water solution at 42-43 degrees containing 240 mg cisplatin using specially devised extracorporeal circuits. Thoracoscopy was used to examine the intrapleural cavity and to place the catheters for perfusion. The patient had an uneventful postoperative course and was discharged for hospital on the second postoperative day. Thereafter, she experienced good quality of life with negative pleural cytology. Unfortunately, the patient died 3 months after the therapy, but the cause of death was unknown and there was no cancer recurrence. This technique may be safe and feasible for controlling malignant effusion to preserve quality of life, although the survival benefit has not yet been clarified.  相似文献

We report a case of fatal hemoptysis resulting from the rupture of a Rasmussen's aneurysm. Postmortem computed tomography (PMCT) clearly showed the air-fluid level within a tuberculous cavity and fluid-filled trachea. In this case, PMCT was useful as a screening tool to detect the cause of death as asphyxia from hemoptysis. It became a guideline for the following autopsy, as well as evidence to explain the importance of autopsy to the family.  相似文献

Uterine papillary serous adenocarcinoma (UPSC) is an uncommon histologic subtype of endometrial cancer that characteristically behaves aggressively with a poor prognosis. We established two novel cell lines derived from UPSC designated HEC-155 and HEC-180. Both cell lines have been growing steadily in monolayer cultures for over ten years. Overexpression of p53, Ki67 and p27 was detected in both cell lines by immunohistochemistry. Using a DNA sequencing technique, a point mutation of p53 was detected in exon 8, codon 286 in HEC-155 and in exon 6, codon 195 in HEC-180. These newly established cell lines should be useful for investigating the characteristics of UPSC.  相似文献

Though reputed for attaining the highest bonding strength, dentin bonding systems that use self-etching primer pose a grave concern. This is because the low pH environment induced by self-etching primer may have negative effects on polymerization reactions. In this study, we used electron spin resonance spectroscopy (ESR), high performance liquid chromatography (HPLC), and dynamic viscoelastic (DV) to analyze the effects of self-etching primer on the polymerization of bonding agents. The results of ESR revealed a lower concentration of polymerization radicals in a sample consisting of bonding agent and self-etching primer than in a sample consisting of bonding agent alone. However, in the results of HPLC and DV, the quantity of unpolymerized monomers decreased and the polymer produced was highly viscous in the sample with self-etching primer. It was suggested that hydroxyethyl methacrylate copolymerized as a plasticizer of Bis-GMA and that it formed the copolymer in the interface between the primer and bonding agent.  相似文献
