脑膜淋巴管转运功能障碍加重脂多糖诱导的小鼠中枢炎症

目的：探讨脑膜淋巴管（meningeal lymphatic vessel,mLV）转运功能障碍对脂多糖（lipopolysaccharide,LPS）诱导的小鼠中枢神经系统炎症的影响。方法：①16只C57BL/6雄性小鼠随机分为4组,Control组腹腔注射生理盐水,LPS组分别腹腔注射LPS（2 mg/kg）12、24、72 h,而后观察小鼠mLV转运功能、海马区小胶质细胞（microglial,MG）活化、白介素（interleukin,IL）-6 和IL-1β水平。②将8只小鼠随机分为2组,分别为Control组和VEGFR3抑制剂MAZ51组,观察MAZ51对mLV转运功能的影响。③将24只小鼠分为4组,分别为Control组、MAZ51组、LPS组、LPS+MAZ51组,对MAZ51组和LPS+MAZ51组预先腹腔注射 MAZ51（10 mg/kg）,每周5 d,共30 d,6周后对LPS组和LPS+MAZ51组注射LPS,1 d后行为学实验评估小鼠的逃避恐惧能力;免疫组化检测MG 活化情况;ELISA 法检测IL-6、IL-1β的表达量。结果：小鼠腹腔注射LPS 24 h后脑膜LYVE-1面积和颈深淋巴结内OVA-647面积明显减少（P < 0.01）,72 h内均低于基础水平（P < 0.01）,24 h后海马MG活化、IL-6和IL-1β均明显增加 （P < 0.01）。与Control组相比,MAZ51组OVA-647荧光面积显著减少（P < 0.01）。LPS组小鼠海马区MG活化和IL-6、IL-1β的表达相对Control组均明显增加（P < 0.01）且小鼠僵直时间明显降低（P < 0.01）;与LPS组相比,LPS+MAZ51组小鼠海马区MG 活化和炎症因子的表达均增加（P < 0.01）且僵直时间明显减少（P < 0.01）。结论：mLV转运障碍通过增加炎性介质聚积,活化 MG,加重LPS诱导的小鼠中枢炎症和认知功能障碍。     

网约护士参与“互联网+护理服务”意愿及影响因素研究

目的 探讨网约护士参与“互联网+护理服务”意愿及影响因素,为提高网约护士参与“互联网+护理服务”的积极性提供参考。方法 2022年1—2月,采用便利抽样法选取广州市开展“互联网+护理服务”医院的279名网约护士为研究对象,采用一般资料调查表、“互联网+护理服务”意愿影响因素量表和组织支持量表进行调查。结果 网约护士参与“互联网+护理服务”意愿得分(12.39±1.61),控制一般资料(职务、获得从业资格时间、居家护理服务经历)的影响后,发现感知风险、感知价值、便利条件、工具支持是网约护士参与“互联网+护理服务”意愿的影响因素(R²=0.641),可解释参与意愿变异的64.1%。结论 网约护士参与“互联网+护理服务”意愿处于中等偏上水平,为患者提供“互联网+护理服务”的意愿受感知风险、感知价值、便利条件和工具支持的影响。     

血清骨膜蛋白和骨硬化蛋白对老年2型糖尿病患者骨质疏松的诊断价值

目的 探讨血清骨膜蛋白(PN)和骨硬化蛋白(SOST)与老年 2 型糖尿病(T2DM)患者骨密度的关系。方法 选取2020年8月—2022年3月海南医学院第二附属医院收治的192例老年T2DM患者作为研究对象,依据骨密度值分为骨量正常组、骨量减少组及骨质疏松组,每组64例。比较3组患者的临床资料,血清PN、SOST、25羟维生素D₃[25(OH)D₃]、骨钙素(OST)及I型胶原氨基端前肽(PINP)水平。多因素Logistic回归分析影响老年T2DM患者骨质疏松发生的危险因素。绘制受试者工作特征(ROC)曲线,以ROC曲线下面积(AUC)评价血清PN、SOST及两者联合对老年T2DM患者骨质疏松的诊断价值。结果 骨量正常组、骨量减少组及骨质疏松组HbA1c、HDL-C、PN、SOST、25(OH)D₃、OST及PINP水平比较,差异有统计学意义(P <0.05),骨质疏松组HbA1c、PN、SOST、OST及PINP水平高于骨量正常组和骨量减少组(P <0.05),骨质疏松组HDL-C、25(OH)D     

Spectrum of morphological appearance of amyloid deposits in Alzheimer's disease

Summary Immunocytochemical staining with monoclonal antibodies to the -protein on tissue sections which have been pretreated with formic acid is not only a very specific but also a highly sensitive method for the detection of amyloid deposits in the brains of Alzheimer's disease victims. We report here a spectrum of morphological appearance of the brain amyloid deposits which are one of the main histopathological correlates of this disorder. Deposits of the -protein are not only found in the well-known lesions [congophilic angiopathy and senile (neuritic) plaques] but are also seen under various morphological forms for which the word plaques does not appear an appropriate term: amyloid fibrils are found as large areas of diffuse infiltration of the neuropil, as ribbon-like infiltration in the subpial layer of the cerebral cortex, as granular deposits in the white matter, as diffuse deposits in the molecular layer of the cerebellum and the basal ganglia and as star-shaped deposits in the cerebellar Purkinje cell layer. The morphology of these deposits seems to depend on the cyto-and fibroarchitectonics of the brain region in which they are found, on the amount of amyloid deposited, and also on the type of staining technique used. It is only under specific circumstances that the deposition of amyloid in the neuropil is accompanied by the formation of paired helical filaments in nerve cell processes and their parent perikarya. In conclusion, our studies suggest that the extent of brain amyloidosis in Alzheimer's disease is much wider than so far appreciated.Supported in part by grants 5-AGO-4220-05 and 5-HD-22634-02 from the National Institutes of Heath     

海洛因对小鼠初级精母细胞影响的形态定量分析

将昆明小鼠随机分为低(0.025)、中(0.05)、高(0.1)剂量组及生理盐水对照组,以2号海洛因经腹腔注射染毒,20天后,取睾丸组织制成石蜡切片.HE染色,光镜下观察生精小管上皮细胞的形态变化,并应用图像分析系统对初级精母细胞的形态参数进行了分析;结果表明,(1)光镜观察 对照组及低剂量组小鼠睾丸组织未见明显的变化;中剂量组小鼠睾丸曲细精管上皮有不同程度的组织改变;高剂量组小鼠生精管壁细胞明显地出现上皮层次减少,精子细胞和精子减少,并发现曲细精管内有脱落的生精细胞;(2)形态定量分析显示 低剂量组及中剂量组各形态参数(中剂量组圆形度除外)较对照组无显著差异;高剂量组初级精母细胞核面积、核灰度与对照组相比无显著差异,细胞面积、细胞周长及浆面积较对照组明显增大(P<0.05).而核浆比例、圆形度较对照组有减小(P<0.05).结果提示 海洛因可明显改变小鼠生殖细胞的形态结构.     

Synaptic transmission of chaotic spike trains between primary afferent fiber and spinal dorsal horn neuron in the rat

Primary sensory neurons can generate irregular burst firings in which the existence of significant deterministic behaviors of chaotic dynamics has been proved with nonlinear time series analysis. But how well the deterministic characteristics and neural information of presynaptic chaotic spike trains were transmitted into postsynaptic spike trains is still an open question. Here we investigated the synaptic transmission of chaotic spike trains between primary Adelta afferent fiber and spinal dorsal horn neuron. Two kinds of basic stimulus unit, brief burst and single pulse, were employed by us to comprise chaotic stimulus trains. For time series analysis, we defined "events" as the longest sequences of spikes with all interspike intervals less than or equal to a certain threshold and extracted the interevent intervals (IEIs) from spike trains. Return map analysis of the IEI series showed that the main temporal structure of chaotic input trains could be detected in postsynaptic output trains, especially under brief-burst stimulation. Using correlation dimension and nonlinear prediction methods, we found that synaptic transmission could influence the nonlinear characteristics of chaotic trains, such as fractal dimension and short-term predictability, with greater influence made under single-pulse stimulation. By calculating the mutual information between input and output trains, we found the information carried by presynaptic spike trains could not be completely transmitted at primary afferent synapses, and that brief bursts could more reliably transmit the information carried by chaotic input trains across synapses. These results indicate that although unreliability exists during synaptic transmission, the main deterministic characteristics of chaotic burst trains can be transmitted across primary afferent synapses. Moreover, brief bursts that come from the periphery can more reliably transmit neural information between primary afferent fibers and spinal dorsal horn neurons.     

酶联免疫法检测PAF受体RNA的聚合酶链反应产物

目的：建立PCR—ELISA法检测附受体RNA。方法：提取20例银屑病病人和20例正常对照组血液单个核细胞标本中的总RNA，用双标记的PAF—R(Biotion与Digoxin标记)及β-actin(Biotin或Fluorescein标记)引物进行RT-PCR，用ELISA检测PCR产物，用传统的琼脂糖凝胶电泳对照。结果；ELISA法和琼脂糖凝胶电泳法对β-actin检测结果均为阳性；ELISA法检测PAF-R，20例病人标本均为阳性，20例对照组16例阳性；琼脂糖凝胶电泳法检测PAF—R，20例病人标本18例阳性，20例对照组16例阳性。结论：PCR-ELISA引物双标记法检测PAF—R操作简单，有较高的敏感性。     

Tumor Vascularity Is Not a Prognostic Factor for Malignant Melanoma of the Skin

Tumor vascularity has been proposed as a prognostic indicator for a number of solid tumors. Although a correlation between microvessel number and metastatic behavior has also been suggested for cutaneous melanoma, the small number of cases studied to date allows one to draw only preliminary conclusions. In this study, we have assessed tumor vascularity in cutaneous melanoma by comparing 60 cases of metastasizing and non-metastasizing tumors matched for tumor thickness, age, sex, and anatomic site. Ulex europaeus agglutinin I appeared to be the most suitable vascular marker for this study. Our results indicate that there was no statistically significant difference between the two groups with regard to tumor vascularity. Even after identifying 15 cases of thin (<1.0 mm thick) melanoma, there was no significant difference in the number of microvessels between metastasizing and non-metastasizing tumors. Comparison of patterns of vascular microarchitecture also failed to discriminate between the two groups. Thus, our results indicate that tumor vascularity may not be an independent prognostic factor for cutaneous melanoma.     

妊娠期间脾脏T淋巴细胞的改变与鼠胚着床

应用ＦＩＴＣ标记的Ｌ３Ｔ４及Ｌｙ２单克隆抗体，及流式细胞计细胞分选，对小鼠妊娠期间脾脏Ｔ淋巴细胞亚群进行检测。结果表明，妊娠早期小鼠脾脏ＣＤ８＋淋巴细胞比例减少，在着床时尤为明显。提示脾脏Ｔ淋巴细胞数量和功能变化，参与维持正常妊娠，尤其是参与着床。
