The potential anticonvulsant effect of group III metabotropic glutamate receptor (mGluR) agonist (R,S)-4-phosphonophenylglycine ((R,S)-PPG) against seizures induced in immature 12-day-old rats by bilateral intracerebroventricular (icv) infusion of DL-homocysteic acid (DL-HCA, 600 nmol/side) was examined in the present study. Rat pups were sacrificed during generalized clonic-tonic seizures, approximately 45 to 50 min after infusion. Comparable time intervals were used for sacrificing the pups which had received (R,S)-PPG. Low doses of (R,S)-PPG (10 nmol, icv) provided a pronounced anticonvulsant effect which was abolished by pretreatment with a selective group III mGluR antagonist (R,S)-alpha-methylserine-O-phosphate. Generalized clonic-tonic seizures were completely suppressed and cortical energy metabolite changes which normally accompany these seizures were either normalized (glucose and glycogen decreases) or markedly ameliorated (an accumulation of lactate). Despite the absence of obvious motor phenomena, EEG recordings revealed sporadic ictal activity, mostly in the dorsal hippocampus. Spreading of this activity into the frontal cortex was rather exceptional. The latency of ictal EEG in pretreated rats was significantly prolonged. Our data suggest that the predominant effect of (R,S)-PPG might concern seizure spread. The administration of (R,S)-PPG alone did not cause any overt behavioral side effects; it did not change the EEG pattern and did not influence cortical metabolite levels, with the exception of increased concentrations of glucose. We investigated, whether the effects on paclitaxel, docetaxel or their combinations on T-cell lymphomas in Sprague-Dawley/Cub rats were mainly caused by their different efficiency or combination of different mechanism of action, or limited by metabolic inactivation by P450 enzymes or drug efflux caused by P-glycoprotein (P-gp). Docetaxel most effectively prolonged the survival of rats and the time of lymphoma appearance, inhibited their intravital size and weight after sacrifice. Paclitaxel was poorly effective and combined administration had intermediate effects. Blood levels of both drugs were similar. Repeated administration of paclitaxel, but not docetaxel, decreased its area under concentration, but the effect disappeared 6h after dosing and was not sufficient to explain lower effects of paclitaxel. The faster metabolism of docetaxel than paclitaxel in vitro did not limit its higher efficiency and repeated administration of paclitaxel did not induce its metabolism to decrease its blood levels sufficiently. Likewise, undetectable expression of P-gp protein in tumours could not explain lower effects of paclitaxel, which is a better substrate of P-gp. Docetaxel was three-fold more effective than paclitaxel against P388D1 lymphoma cell line, used as a model of the T-cell lymphoma and combined action was dominated by the effects of docetaxel. Hydroxyapatite/poly-L-lactide (HAp/PLLA) is a ceramic/polymer composite, whose application as a resorbable biomaterial for the substitution and repair of hard bone tissue is widely promising in orthopedic, oral, maxillofacial, and reconstructive surgery. Hot processing is a necessary step for obtaining HAp/PLLA composite blocks with mechanical properties similar to those of bones. In this article, the changes in structure and physicochemical properties of HAp/PLLA composite, hot pressed for different pressing times (5, 15, 30, 45, and 60 min), were studied. Because the morphology of HAp/PLLA composite biomaterial is very sensitive to this procedure, its surface microstructure was analyzed by scanning electron microscopy (SEM) coupled with an energy-dispersive X-ray (EDX) detector system. Structural changes occurring in the material, mostly changes in crystallinity of PLLA, were studied by wide-angle X-ray structural analyses (WAXS) and infrared (FTIR) spectroscopy. Using differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), and gel permeation chromatography (GPC), the influence of hot pressing on the properties, crystallization kinetics, and decomposition of HAp/PLLA composite biomaterial was analyzed. 相似文献

Interleukin-2 (IL-2) is able to generate nonspecific cytotoxic effectors from hematopoietic precursors. We evaluated the feasibility
and efficacy of chronic myeloid leukemia (CML) treatment with autologous hematopoietic stem cell transplantation (HSCT) using
grafts cultured in IL-2 followed by immunotherapy with IL-2, granulocyte-macrophage colony-stimulating factor (GM-CSF), and
interferon (IFN)-α. Eight patients with CML were enrolled: five in an accelerated phase and three in a chronic phase. They
received peripheral blood stem cells (PBSC) or bone marrow (BM) cultured in a medium containing IL-2 for 24 h. A median of
1.29 × 106 CD34+ cells/kg were infused after conditioning with busulfan (12–16 mg/kg) in PBSC recipients. BM was infused without prior
myeloablative therapy. The engraftment occurred with a median of 15 d. Engraftment failure developed in one patient. The transplantation
was followed by a 1-mo regimen of IL-2 (0.5 × 106 IU/m2 daily) and GM-CSF, and 6 mo of IFN-α. One complete and one transient minor cytogenetic remission were observed. At 24 mo
after transplantation, two patients had died of progressive disease and one of infection. Five patients had stable disease
in the chronic phase. Autologous transplantation using IL-2-activated graft is feasible and the subsequent IL-2, GM-CSF, and
IFN-α administration has acceptable toxicity. However, no benefits in comparison with conventional autologous transplantation
for CML were identified in our study.

OBJECTIVE: The purpose of this study was an objective consideration of possible benefits of phototherapy implemented with therapeutic laser or possibly polarized light in treating episiotomy, which is the most frequent obstetric intervention. MATERIALS AND METHODS: In the present study, the authors treated a total of 2,436 women. The light sources were as follows: a laser of a wave length 670 nm, power 20 mW, with continuous alternations of frequencies 10 Hz, 25 Hz, and 50 Hz, a polarized light source of a 400-2,000 nm wavelength in an interval of power 20 mW and frequency 100 Hz and a monochromatic light source of a 660 nm wavelength and power 40 mW, with simultaneous application of a magnetic field at an induction 8 mT. RESULTS: The work demonstrated high healing effects with minimum secondary complications in the treatment of episiotomies using a therapeutic laser at an energy density of 2 J/cm(2). The application of polarized light at an energy density of 5 J/cm(2) also exerted favorable therapeutic effects. 相似文献

Surfactin, a highly powerful biosurfactant produced by various strains of the genus Bacillus, exhibits antibacterial, antiviral, antitumor and hemolytic action. This anionic cyclic lipopeptide is constituted by a heptapeptide
interlinked with a β-hydroxy fatty acid. Due to its amhipathic nature surfactin incorporates into the phospholipid bilayer
and induces permeabilization and perturbation of target cells. The rising antibiotic resistance as well as a number of remarkable
surfactin activities shows that it deserves special interest and is considered as a candidate compound for combating several
