ObjectivesTo investigate the function of PAQR3 in gastric cardia adenocarcinoma (GCA) and understand the possible mechanism of PAQR3 in regulating epithelial–mesenchymal transition (EMT).MethodsWe detected PAQR3 protein in 146 GCA tissues and paired normal adjacent tissues (PNTs) specimens using immunohistochemical analysis, and explored its clinical significance. The expression levels of PAQR3 protein in 20 GCA tissues, their paired PNTs, HGC27, SGC7901, and GES‐1 cells were analyzed by Western blot. Wild‐type PAQR3 was overexpressed in HGC27 cells. The effects of PAQR3 overexpression on the function of HGC27 cells and its underlying mechanisms were then analyzed through a series of cell and molecular biology experiments.ResultsPAQR3 was significantly down‐regulated in GCA tissues when compared with paired PNTs (p < 0.0001). The expression level of PAQR3 in GCA tissues was significantly negatively correlated with Helicobacter pylori infection (p = 0.000), venous invasion (p = 0.000), invasion depth (p = 0.000), lymph node metastasis (p = 0.022), tumor stage (p = 0.000), and patient survival (p = 0.009). Downregulation of PAQR3 was highly correlated with increased EMT signature and activated TGF‐β/Smad pathway in GCA tissues. Overexpression of PAQR3 in HGC27 cells negatively regulates its cellular functions, such as cell proliferation and migration, and suppresses EMT. Mechanistically, overexpression of PAQR3 significantly down‐regulates the protein expression levels of TGF‐1, p‐Smad2, and p‐Smad3 in HGC27 cells.ConclusionPAQR3 was significantly down‐regulated in GCA tissues, HGC27, and SGC7901 cells. PAQR3 significantly inhibits the proliferation, migration, and invasion of HGC27 cells. Mechanistically, PAQR3 can inhibit the EMT process in HGC27 cells by regulating TGF‐β/Smad signaling pathway. 相似文献

In this work, the antioxidant abilities of NADH coenzyme analogue BNAH, F420 reduction prototype analogue F420H, vitamin C analogue iAscH−, caffeic acid, and (+)-catechin in acetonitrile in chemical reactions were studied and discussed. Three physical parameters of the antioxidant XH, homolytic bond dissociation free energy ΔG°(XH), self-exchange HAT reaction activation free energy ΔG≠XH/X, and thermo-kinetic parameter ΔG≠°(XH), were used to evaluate the antioxidant ability of XH in thermodynamics, kinetics, and thermo-kinetics. By comparing ΔG°(XH), ΔG≠XH/X and ΔG≠°(XH) of these five bioactive antioxidants to release hydrogen atoms, it is easy to find that iAscH− is the best hydrogen atom donor both thermodynamically and kinetically among these antioxidants. Caffeic acid is the worst hydrogen atom donor thermodynamically, and F420H is the worst hydrogen atom donor kinetically. In addition, the thermodynamic hydride donating abilities of BNAH, F420H, and iAscH− were also discussed, and the order of thermodynamic hydride donating abilities was BNAH > F420H > iAscH−. Four HAT reactions BNAH/DPPH˙, (+)-catechin/DPPH˙, F420H/DPPH˙, and caffeic acid/DPPH˙ in acetonitrile at 298 K were studied by the stopped-flow method. The actual order of H-donating abilities of these four antioxidants in the HAT reactions is consistent with the order predicted by thermo-kinetic parameters. It is feasible to predict accurately the antioxidant abilities of antioxidants using thermo-kinetic parameters.In this work, the antioxidant abilities of NADH coenzyme analogue BNAH, F420 reduction prototype analogue F420H, vitamin C analogue iAscH−, caffeic acid, and (+)-catechin in acetonitrile in chemical reactions were studied and discussed. 相似文献

Bone is a common site of metastasis in lung cancer, but the regulatory mechanism remains incompletely understood. Osteoclasts are known to play crucial roles in osteolytic bone metastasis by digesting bone matrix and indirectly enhancing tumor colonization. In this study, we found that IL receptor 20 subunit β (IL-20RB) mediated a direct tumoral response to osteoclasts. Tumoral expression of IL-20RB was associated with bone metastasis of lung cancer, and functionally, IL-20RB promoted metastatic growth of lung cancer cells in bone. Mechanistically, tumor cells induced osteoclasts to secrete the IL-20RB ligand IL-19, and IL-19 stimulated IL-20RB–expressing tumor cells to activate downstream JAK1/STAT3 signaling, leading to enhanced proliferation of tumor cells in bone. Importantly, blocking IL-20RB with a neutralizing antibody significantly suppressed bone metastasis of lung cancer. Overall, our data revealed a direct protumor role of osteoclastic niche in bone metastasis and supported IL-20RB–targeting approaches for metastasis treatment. 相似文献

Ammonia (NH3) is a common air pollutant and is a biomarker for kidney disease. Therefore, the preparation of ammonia gas sensors with high sensitivity, good selectivity and low operating temperature is of great importance for health protection. Using the in situ electrostatic self-assembly approach, a chemoresistive gas sensor based on Co(OH)2/Ti3C2Tx hybrid material was created in this study. The prepared samples were characterized by XRD, XPS, TEM, BET and other testing methods for structure, surface topography and elements. These samples were fabricated into sensors, and the gas sensing properties of the materials were investigated under different test conditions. The results show that the gas response value of the C/M-2 sensor is up to about 14.7%/100 ppm, which is three times the response value of the sensor made of pure MXene to NH3. In addition, the Co(OH)2/Ti3C2Tx hybrid sensors exhibit excellent repeatability, high sensitivity under low concentration (less than 5 ppm), fast response/recovery time (29 s/49 s) and long-time stability, which indicates their promising utility in the IoT field.The prepared Co(OH)2@Ti3C2Tx composite shows good gas sensing performance for ammonia at room temperature, making it a promising candidate for a practical detector for NH3. 相似文献

目的:探究益气活血泄浊法治疗浊毒内蕴、肾气衰惫型慢性肾衰竭患者的效果。方法:选取2018年10月至2019年4月辽宁中医药大学附属医院收治的慢性肾衰竭患者100例作为研究对象,按照随机数字表法随机分为对照组和观察组,每组50例。对照组患者进行利水消肿、纠正电解质失衡、控制心率血压、低盐低脂饮食等常规西医治疗。观察组患者在对照组患者的治疗基础上给予益气泄浊汤口服。治疗周期为15 d。观察2组患者治疗前后中医证候积分、血肌酐(SCr)、转化生长因子(TGF-β)、Ⅳ-C、肝细胞生长因子(HGF)及临床有效率情况。结果:治疗后,2组的中医证候主证评分、SCr、TGF-β、HGF、Ⅳ-C含量较治疗前均有所改善,且观察组明显优于对照组,差异有统计学意义(P<0.05)。治疗后对照组临床总有效率为70%,观察组临床总有效率为88%,差异有统计学意义(P<0.05)。结论:益气泄浊汤治疗浊毒内蕴、肾气衰惫型慢性肾衰竭可有效抑制肾纤维化形成,提高肾小球滤过率,改善临床症状,提高临床疗效。 相似文献
