This work describes the development of a novel protein expression system based on Rachiplusia nu larvae for the production of the recombinant E2 protein to be used as a vaccine candidate against bovine viral diarrhea virus (BVDV). A recombinant baculovirus (Ac-E2pol+) bearing the E2 glycoprotein coding sequence of BVDV was obtained. Fourth-instar R. nu larvae were infected orally with recombinant polyhedra and the expression of E2 protein was confirmed by immunoblot. In order to test the recombinant product as a vaccine candidate, an immunization assay was performed and the neutralizing humoral immune response against BVDV NADL strain was evaluated. Mice vaccinated with Ac-E2pol+ extracts of per os infected larvae developed a neutralizing antibody titer of 3.16 after the administration of three doses of the immunogen. This report demonstrates the efficacy of per os infected larval extracts as a BVDV recombinant immunogen, which constitutes an easier and economic approach for producing recombinant antigens.  相似文献

ABSTRACT

This article makes a review and reflection on parenting practices and child development in the perinatal period; the theoretical foundations and recent data in the field are exposed. Spanish and international research in this emerging area indicates that pregnancy, postpartum and early parenting are opportunities and unique spaces to develop competencies to create family contexts that promote healthy development. An exhaustive positive parenting proposal of early promotion of child development in the perinatal period is presented. This includes the promotion of: prenatal bond, couple's relationship, social support networks, the physical and mental health of the mother alongside with the support to make informed decisions about parenting and the development of parental knowledge of the intergenerational transmission of parenting patterns and of early childhood development. It is expected, that the proposal could be a tool in the future design of public intervention programmes with families.  相似文献

Controlled drug delivery aims to achieve an effective drug concentration in the action site for a desired period of time, while minimizing side effects. In this contribution, biodegradable poly(3-hydroxybutyrate) films were evaluated as a reservoir platform for dexamethasone controlled release. These systems were morphological and physicochemically characterized. In vitro release assays were performed for five different percentages of drug in the films and data were fitted by a mathematical model developed and validated by our research group. When the profiles were normalized, a single curve properly fitted all the experimental data. Using this unique curve, the dissolution efficiency (DE), the time to release a given amount of drug (tX%), and the mean dissolution time were calculated. Furthermore, the dissolution rate, the initial dissolution rate (a%) and the intrinsic dissolution rate were determined. The a% mean value was 1.968 × 10?2% released/min, t80% was about 14 days, and the DE was 59.6% at 14 days and 66.5% at 20 days. After 2 days, when approximately 40% of the drug was released, the dissolution rate decreased about 60% respect to the initial value. The poly(3-hydroxybutyrate) platforms behaved as an appropriate system to release and control the dexamethasone delivery, suggesting that they could be an alternative to improve drug therapy.  相似文献

The ability of the immune system to respond to different pathogens throughout life requires the constant production and selection of T cells in the thymus. This immune organ is very sensitive to age, infectious processes and nutrition disorders (obesity and malnutrition). Several studies have shown that the incorporation of some probiotic bacteria or probiotic fermented milk in the diet has beneficial effects, not only at the intestinal level but also on distant mucosal tissues, improving the architecture of the thymus in a malnutrition model. The aim of the present study was to determine whether supplementation with the probiotic strain Lactobacillus casei CRL 431 and/or its cell wall could improve body weight, intestinal microbiota and thymus structure and function in both obese and aging mice. We evaluated probiotic administration to BALB/c mice in 2 experimental mouse models: obesity and senescence, including mice of different ages (21, 28, 45, 90 and 180 days). Changes in thymus size and histology were recorded. T-lymphocyte population and cytokine production were also determined. The consumption of probiotics improved the cortical/medullary ratio, the production and regulation of cytokines and the recovery of mature T-lymphocyte populations of the thymus in obese and old mice. Probiotic incorporation into the diet could not only modulate the immune system but also lead to thymus function recovery, thus improving quality of life.  相似文献
