Antiserum was raised against an acetic acid extract of partly mineralized bovine enamel. By use of immunoblotting it was shown that rabbit antibodies react with both high and low molecular weight proteins. Most but not all of the enamel proteins isolated by different extraction solutions were antigenic identical. The blotting time was found to be important when immunoblotting of enamel proteins is carried out.  相似文献

When an in vitro system is used to study the influence of ischemia on hepatic protein synthesis, an important question is whether alterations observed in vitro reflect changes in vivo. In the present study the effects of liver ischemia on protein synthesis were investigated in rats both in vitro and in vivo. Liver ischemia was induced by hepatic artery ligation. Protein synthesis in vitro was determined from leucine incorporation into proteins in liver slices incubated in a medium containing 14C-leucine (0.5 mmol/l) and in vivo from leucine incorporation into hepatic proteins after intraportal injection of a tracer dose of 14C-leucine. Leucine incorporation rate in non-ischemic liver was 0.16 pmol * g pror1 h-1 in vitro and 19.6 μmol g prot-1. h-1 in vivo. After hepatic artery ligation protein synthesis in vitro was reduced by about 60% and in vivo by about 80%. Thus, the relative changes were of the same magnitude in vitro and in vivo. This indicates that an in vitro system can be used to evaluate the effects of liver ischemia on hepatic protein synthesis.  相似文献

Summary. During the second pregnancy of 56 Swedish women resulting in a term birth, energy intake and physical activity were measured for 3 days at weeks 17 and 33. The values were related to maternal lean body mass, pregnancy weight gain, maternal fat accretion and infant birth-weight by multiple linear regression analyses. A significant regression coefficient was found for energy intake at week 17 on maternal fat accretion. Energy intake was not significantly correlated with infant birthweight, not even when physical activity and maternal lean body mass were taken into account. Thus in a well-nourished Swedish population, energy intake is positively related to maternal fat accretion but not to the birthweight of term infants.  相似文献

Early or localized forms of arrhythmogenic right ventricular dysplasia (ARVD) have been proposed as the arrhythmogenic substrate of repetitive monomorphic ventricular tachycardia (RMVT) originating in the right ventricular outflow tract in patients without any underlying cardiac abnormality on clinical examination and echocardiography. To further examine this hypothesis, magnetic resonance imaging (MRI) and signal-averaged electrocardiography (SAECG) were performed on 23 patients with RMVT and normal 12-lead standard ECG of conducted sinus beats. MRI was performed using ECG-gated turbo spin-echo images of the heart in order to detect signs of early or localized forms of ARVD, such as localized wall thickness reductions, signal intensity increase indicating adipose tissue infiltrates, and regional bulgings or aneurysms. MRI was normal in 22 (96%) of 23 study patients. In the remaining patient (4%), MRI demonstrated signal intensity increase in the intraventricular septum but not in the right ventricular outflow tract. Time-domain analysis of the SAECG was normal in 21 (91 %) of 23 patients and revealed ventricular late potentials in 2 study patients (9%). Frequency-domain analysis of the SAECG was normal in 22 (96%) of 23 patients and revealed ventricular late potentials in one study patient (4 %). We conclude that normal MRI findings of the heart and absence of ventricular late potentials in the SAECC in most patients with RMVT and otherwise normal ECG do not support the hypothesis that early or localized forms of ARVD create the arrhythmogenic substrate in the majority of these patients.  相似文献

Mode switching algorithms are commonly used to protect the ventricles against high rates induced by atrial tachycardia. In the case of atrial fibrillation (AF), the response of these algorithms depends on the quality of atrial sensing. The Chorum 7234 DDDR pacemaker uses a new mode switching algorithm, based on a statistical analysis of the atrial rhythm. It includes two criteria of diagnosis: "high" if more than 28 of 32 cycles are abnormally accelerated; and "low" if more than 36 of 64 cycles are abnormally accelerated. Methods: From a taped database of electrophysiological studies, episodes of AF lasting more than 2 minutes were selected. A tape recorder replayed the atrial signals into an external Chorum device. Each episode was replayed eight times with a programmed atrial sensitivity increasing from 0.4 –2.0 mV. For each criterion of diagnosis and each programmed sensitivity, the percentage of atrial sensing, the time to switching, and the mean ventricular rate were measured. Ten episodes of AF from 10 patients (9 men and 1 woman; ages 62 ± 16 years) were included: 1.95 ± 0.97 mV and 196 ± 64 ms. The sensitivity of the algorithm to diagnose atrial tachycardia reached 100%, for an atrial sensitivity set between 0.4 and 1.0 mV. The mean percentages of atrial sensed events were 74%± 18% and 46%± 9% for the "high" and "low" criteria, respectively. The mean diagnostic times were 28 ± 26 seconds and 68 ± 27 seconds, respectively. Sensing of < 23% of AF events resulted in failure to diagnose the arrhythmias by both algorithms. In the event of diagnostic failure, the mean ventricular pacing rate was 79 ± 9 ppm. Conclusion: Up to an atrial sensitivity of 1 mV, 100% of AF episodes were diagnosed. The Chorum mode switching algorithms are 100% reliable if > 45% of the AF waves are sensed. In the event of switching failure, the ventricle is protected by an average rate remaining below 80 ppm. Abstract. The effect of lowering total plasma and low density lipoprotein (LDL) cholesterol in heterozygous familial hypercholesterolaemia type IIa (FH) on platelet function, thromboxane (TX) formation and platelet sensitivity against iloprost, a stable prostacyclin mimetic, was studied in platelet-rich plasma ex vivo . Seven FH patients were treated with cholestyramine (12 g day-1) for 8–11 months and were compared with eight untreated FH patients and 11 healthy control subjects. In comparison with platelets from healthy controls, platelets from untreated FH patients exhibited a significantly increased aggregation response and TX formation, and a reduced reactivity against inhibition of platelet aggregation by prostacyclin. Treatment with cholestyramine for 8–11 months resulted in a 21% reduction in total serum and LDL-cholesterol. This was not accompanied by any change in platelet hyperreactivity or TX formation. However, cholestyramine treatment normalized the platelet reactivity of FH patients against iloprost, being no more different from healthy controls. It is concluded that reduction in plasma cholesterol by cholestyramine results in normalization of the reduced platelet sensitivity against prostacyclin. This might contribute to beneficial effects of cholestyramine treatment in preventing thrombembolic complications of atherosclerosis.  相似文献

This study has evaluated the possible role played by the l-arginine-nitric oxide pathway in the vasorelaxant action of the hydroalcoholic extract from Eugenia uniflora, and fractions from the extract, in rings of rat thoracic aorta. The addition of an increasing cumulative concentration of hydroalcoholic extract from E. uniflora (1–300 μg mL?1) caused a concentration-dependent relaxation response in intact endothelium-thoracic aorta rings pre-contracted with noradrenaline (30–100 nm). The IC50 value, with its respective confidence limit, and the maximum relaxation (Rmax) were 7.02 (4.77–10.00) μg mL?1 and 83.94 ± 3.04%, respectively. The removal of the endothelium completely abolished these responses. The nitric oxide synthase inhibitors Nω-nitro-l-arginine (l-NOARG, 30 μm) and Nω-nitro-l-arginine methyl ester (l-NAME, 30 μm), inhibited the relaxation (Rmax) to ?10.43 ± 7.81% and ?3.69 ± 2.62%, respectively. In addition, l-arginine (1 mm), but not d-arginine (1 mm), completely reversed inhibition by l-NOARG. Methylene blue (30 μm), a soluble guanylate cyclase inhibitor, reduced the relaxation induced by the extract to 14.60 ± 7.40%. These data indicate that in the rat thoracic aorta the hydroalcoholic extract, and its fractions, from the leaves of E. uniflora have graded and endothelium-dependent vasorelaxant effects.  相似文献

BACKGROUND: Malignant melanoma is the third most common skin cancer in the United States. It is commonly thought that sun exposure is causative in these tumors. Recently, however, the significance of the role of sun exposure in melanoma has come into question. Some have suggested that other factors, such as genetics, play a larger role, and that sun protection may even be harmful. OBJECTIVE AND METHODS: To investigate the role of sun exposure in melanoma etiology. An extensive review of basic science and clinical literature on this subject was conducted. RESULTS: Although exceptions exist, sun exposure likely plays a large role in most melanomas. The pattern of this exposure, however, is not fully known, and controversy exists, especially in the use of sunscreens. Sun exposure may interact with genetic factors to cause melanomas, and sun protective measures appear to be prudent. CONCLUSIONS: The cause of melanoma is probably variable and multifactorial. Sun exposure may play a primary or supporting role in most melanoma tumors.  相似文献

Cellular DNA in skin biopsies from eighteen patients with positive patch tests and eleven patients with benign lymphocytic skin infiltates (cutaneous lymphocytoma, lymphoplasia or Jessner's lymphotic infiltrate) was estimated by flow cytometry of propidium iodide-stained dermal cells. Specimens from three patients with patch tests and one with cutaneous lymphocytoma had DNA histograms similar to those of normal blood mononuclear cells. The remaining patients demonstrated DNA histogram abnormalities in the form of hyperdiploid peaks in forty-two of forty-five specimens. The estimated proportion of cells in the S-phase was similar in patch tests (median: 9.2%) and in benign lymphocytic skin infiltrates (median: 9.6%). The median number of cells in the G2/M-phase was 3.1% in both groups of disorders. These data indicate DNA content heterogeneity of the dermal cells in benign lymphocytic skin infiltrates. This may be attributed to DNA replication and/or chromatin dispersion during cellular activation and proliferation. The data also emphasize that hyperdiploid peaks are not necessarily indicative of the presence of separate cell clones.  相似文献

abstract — The barrier property of inflamed palatal mucosa to water has been studied in eight adult edentulous persons with a generalized denture stomatitis, by measuring the transmucosal water flow under varying osmotic gradients. Flow rates were registered gravimetrically in solute saturated filter paper discs after 10-min periods of mucosal contact, using solutions with an osmolarity of 0, 0.25, 0.30, 0.38, 0.50 and 0.75 osmol sucrose/1. The histology of the mucosal areas was evaluated from cytologic scrapings, and biopsy material from two persons. The inflow with use of pure water was 2.98 mg/cm2/10 min, being about three and a half times greater than through the intact mucosal surface. The point of isotony of the inflamed mucosa ranged between 0.30 and 0.36 with a mean value of 0.33 osmol/1, thus being of the same magnitude as in plasma and tissue fluid. The observations from the biopsy material were in accordance with earlier histological evidence from generalized denture stomatitis, indicating that the permeability properties of the inflamed mucosa belonged to epithelial cell layers located in the lower part of the spinous layer.  相似文献
