黄热病毒特异性抗原片段的筛选与鉴定

目的通过系统的生物信息学分析和实验验证,筛选黄热病毒的特异抗原片段,为免疫诊断试剂的研制奠定基础。方法分别利用DNAStar及ANTHEPROT软件对黄热病毒蛋白进行分析,参考亲水性、抗原性、可塑性、表面可及性及二级结构信息,对黄热病毒可能的共有及特异性抗原表位进行系统的预测分析,分析其在不同毒株中的保守性,并对预测得分值较高的抗原区域进行RT-PCR扩增,利用pMal-c2x原核表达系统进行原核表达,Western blot验证其免疫学反应原性,检测阳性抗原片段经亲和纯化后,包被ELISA微孔板,进一步验证其免疫学反应特异性及检测敏感性。结果其中27段抗原片段获高效表达,经Western blot筛选及ELISA进一步验证,原核表达抗原片段YFV22表现良好特异抗,可检测低至1∶12 800倍稀释的黄热病毒多克隆抗体,与所试其他参考病毒多克隆抗体无交叉反应。结论经系统筛选、验证,获黄热病毒特异性抗原片段1段,为其免疫学诊断试剂的研制奠定了基础。     

乙肝疫苗初免失败婴儿再免疫方法研究

目的探索乙肝疫苗初次全程免疫失败婴儿的再免疫方法.方法对乙肝疫苗全程免疫后乙肝病毒表面抗体(抗-HBs)和乙肝病毒表面抗原(HBsAg)均阴性的144名婴儿,随机分成5组,1组为对照组,其余4组分别采用不同的再免疫方法,观察再免成功率.结果抗-HBs总阳转率为93.79%,抗-HBs几何平均滴度(GMT)为546.97mIU/ml,各组抗-HBs阳转率分别为87.88%、90.91%、100%、96.88%;各组GMT分别为475.34mIU/ml、658.65mIU/ml、625.18mIU/ml、455.90mIU/ml.结论乙肝疫苗初免失败婴儿,再免成功率达93.79%.     

德都红花-7味散原药方与优化方对肝纤维化大鼠Ⅰ,Ⅲ型胶原mRNA表达的影响

目的:探讨德都红花-7味散原药方及优化方对肝纤维化大鼠Ⅰ,Ⅲ型胶原mRNA表达的影响。方法:40只Wistar大鼠分为正常组、模型组、阳性药组、原药组、优化组。大鼠ip 30%CCl4橄榄油溶液建立肝纤维化模型。同时1次/日ig给药,阳性药组给予秋水仙碱片0.4 mg·kg-1;原药组给予德都红花-7味散0.6 g·kg-1;优化组给予德都红花-7味散优化方0.6 g·kg-1。连续40 d后处死大鼠。取肝脏天狼星红染色,观察肝组织纤维化程度和Ⅰ,Ⅲ型胶原分型。酶联免疫吸附法(ELISA)检测肝组织匀浆Ⅲ型前胶原和Ⅳ型胶原,层粘连蛋白(LN),透明质酸(HA)含量。实时荧光定量PCR法检测Ⅰ型胶原,Ⅲ型胶原mRNA表达变化。结果:天狼星红染色模型组Ⅰ,Ⅲ型胶原纤维较正常组增多,阳性药组、原药组、优化组Ⅰ,Ⅲ型胶原纤维较模型组减少。与正常组比较,酶联免疫法检测模型组Ⅳ型胶原,HA含量升高(P0.05)。与模型组比较,阳性药组、原药组、优化组Ⅲ型前胶原、Ⅳ型胶原、HA含量明显降低(P0.05)。实时荧光定量PCR法检测模型组Ⅰ型胶原mRNA和Ⅲ型胶原mRNA表达较正常组上调(P0.01)。阳性药组、原药组、优化组Ⅰ型胶原mRNA和Ⅲ型胶原mRNA表达较模型组下调(P0.01)。原药组、优化组Ⅰ型胶原mRNA表达较阳性药组下调(P0.05)。优化组Ⅰ型胶原mRNA表达较原药组下调(P0.05)。结论:德都红花-7味散原药方与优化方是通过抑制Ⅰ型胶原mRNA和Ⅲ型胶原mRNA的转录,达到阻断或延缓肝纤维化的发生发展,德都红花-7味散优化方优于原药方。     

妊娠晚期孕妇抑郁与睡眠质量的相关性研究

目的 了解妊娠晚期孕妇抑郁和睡眠质量的现况及其影响因素,探讨妊娠晚期孕妇抑郁与睡眠质量的相关性。方法 选择中卫市2所医院535名符合调查要求的孕妇进行问卷调查,采用匹兹堡睡眠质量指数量表（pittsburgh sleep quality index,PSQI）和抑郁自评量表（self-rating depression scale,SDS）评定孕妇睡眠质量及抑郁。结果 535名孕妇抑郁检出率为34.2%,睡眠障碍的检出率为20.6%。有抑郁的孕妇睡眠障碍检出率（29.0%）高于无抑郁的孕妇（16.2%）,差异有统计学意义（χ²=12.018,P=0.001）。PSQI总分和SDS得分呈正相关（r=0.680,P<0.001）。文化程度低、夫妻关系差、有孕期并发症、睡眠障碍是妊娠晚期孕妇抑郁的危险因素。有孕期并发症、担忧孩子健康、抑郁是妊娠晚期孕妇睡眠障碍的危险因素。结论 妊娠晚期孕妇睡眠质量与抑郁呈正相关,应采取综合性干预措施改善妊娠晚期孕妇的抑郁情绪和睡眠质量。     

不同剂量链脲佐菌素建立妊娠期糖尿病大鼠模型稳定性的比较

目的：探讨链脲佐菌素（STZ）建立妊娠期糖尿病（GDM）大鼠模型中不同给药剂量对模型稳定性的影响,研究GDM对母体及胎儿的影响率、转阴率和死亡率.方法：将46只SD孕鼠随机分为STZ低、中、高不同剂量组,分别一次性腹腔注射STZ35,45,60mg/kg;对照组腹腔注射等量的柠檬酸缓冲液.于妊娠第3,9,14,19d时测空腹血糖,并观察成模孕鼠在妊娠中体质量、饮水量、摄食量和尿量变化,比较孕鼠成模情况.结果：STZ45mg/kg组孕鼠在用药后出现明显的“三多一少”症状,其成模率83.3%为最高,转阴率最低.空腹血糖值STZ45mg/kg组（21.8±3.0）mmol/L与对照组（5.9±1.2）mmol/L相比较,其高血糖状态持续时间长,最稳定,且体质量也明显降低,差异具有统计学意义（P〈0.05）.结论：STZ45mg/kg腹腔注射给药是建立GDM大鼠模型的最佳剂量,具有较好的稳定性.     

Pseudoaneurysm detection with Tc-99m-labeled red blood cells

A technique for the noninvasive diagnosis of pseudoaneurysms is described. This method employs in vivo labeling of red blood cells with Tc-99m to allow better delineation of the vascular anatomy than standard radionuclide angiography. Four cases are illustrated.     

NB4 cells show bilineage potential and an aberrant pattern of neutrophil secondary granule protein gene expression

NB4 is an acute promyelocytic leukemia cell line that has been shown to be inducible to terminal neutrophil maturation with all-trans retinoic acid (ATRA). HL60 cells are differentially inducible with 12-O- tetradecanoylphorbol-13-acetate (TPA) or dimethyl sulfoxide (DMSO) to monocytes or granulocytes, respectively. HL60 cells induced with DMSO undergo defective neutrophil maturation, manifested by a coordinate failure of secondary granule protein gene expression. We observed a similar defect in granulocytic maturation in ATRA-induced NB4 cells. In addition, because normal promyelocytes are known to have bilineage potential, we have investigated differentiation along monocytoid lines induced with TPA. We observed a striking phenotypic change along monocytoid/macrophage lines with TPA induction. Flow cytometry showed a TPA-induced increase in HLA-DR expression, and Northern blot analysis showed induction of expression of CD18, c-fos, and human neutrophil gelatinase (HNG). HNG is unique among the neutrophil secondary granule protein genes in that it is expressed in both the neutrophil and monocyte lineages. This again parallels our findings in TPA-induced HL60 cells, which retain the ability to express HNG. These findings confirm bilineage potential in NB4 cells. They also support the hypothesis of coordinate neutrophil secondary granule protein gene expression and a defect in this control as part of the leukemic phenotype.     

Respiratory distress syndrome treated with human surfactant: radiographic findings

Chest radiographs of 18 newborns treated with endotracheal instillation of human surfactant for respiratory distress syndrome (RDS) were compared with those of 18 similar but untreated infants. In the treated infants, severity of RDS significantly improved after surfactant administration. Most treated infants (16/18) exhibited a left-to-right shunt, presumably through a patent ductus arteriosus; similar findings were noted in untreated infants (17/18). Complications of respiratory assistance in the treated infants included transient pulmonary interstitial emphysema (n = 1), pneumothorax (n = 1), and mild (n = 4) to moderate (n = 2) bronchopulmonary dysplasia; the incidences of these complications did not exceed those in untreated infants. In three treated infants, a transient interstitial lung disease developed 3-4 days after surfactant administration.
