An alternative to the usual technique of preclotting porous textile vascular prostheses prior to surgical implantation is to render them impermeable to blood by impregnation with a cross-linked albumin filler matrix. This material subsequently becomes the foundation for cellular development. The compatibility of such impregnated fabrics with newly formed endothelial cells has been evaluated by an in vitro organotypic culture method. This technique enables the characterization and numeration of cells that develop on blood contact surfaces and enables determination of their rate of development. Woven, knitted, and velour fabrics were evaluated following coating with albumin and either storage in Tyrode solution or 40% ethanol or desiccation by critical point drying. Preclotted cardiovascular repair fabrics prepared according to conventional surgical protocol served as controls. The identification of the newly formed cells was confirmed histologically. The most extensive and rapid cellular development was observed on the woven fabric and is believed may have resulted from the smoother surface topography of this substrate. Good cellular development was noted particularly on fabrics which had been stored in Tyrode solution. Ethanol had a deleterious effect on the apparent compatibility.  相似文献

The objective of this study was to determine whether magnesium consumption is associated with inflammation (C-reactive protein [CRP]) in children. The study was an analysis of child (age 6-17 years) participants in the cross-sectional, nationally representative National Health and Nutrition Examination Survey (NHANES). Children consuming less than 75% of RDA were 1.94 times more likely (p < 0.05) to have elevated serum CRP levels than children consuming above the RDA. In adjusted analyses controlling for demographics, cardiovascular risk factors, and BMI, children with consumption of less than 75% RDA were 58% more likely to have elevated CRP (OR 1.58, 95% CI 1.07-infinity). Children with intakes below the RDA are more likely to have elevated CRP levels.  相似文献

This paper deals with methods of reducing the total time required to acquire the projection data for a set of contiguous computed tomography (CT) images. Normally during the acquisition of a set of slices, the patient is held stationary during data collection and translated to the next axial location during an interscan delay. We demonstrate using computer simulations and scans of volunteers on a modified scanner how acceptable image quality is achieved if the patient translation time is overlapped with data acquisition. If the concurrent patient translation is ignored, structured artifacts significantly degrade resulting reconstructions. We present a number of weighting schemes for use with the conventional convolution/backprojection algorithm to reduce the structured artifacts through the use of projection modulation using the data from individual and multiple slices. We compare the methods with respect to structured artifacts, noise, resolution and to patient motion. Review of preliminary results by a panel of radiologists indicates that the residual image degradation is tolerable for selected applications when it is critical to acquire more slices in a patient breathing cycle than is possible with conventional scanning.  相似文献

Laser Doppler flowmetry with optical-fibre beam transmission is a sensitive fast and convenient method of measuring tissue blood flow. However, its sensitivity can also be a problem because of movement artefacts. This study applies some basic considerations of fibre optics and Rayleigh light scattering to the field of laser Doppler blood flow meters. Practical suggestions are given by which movement arterfacts can be reduced by choice of optical fibres, attention to probe geometry, cladding the fibres to reduce their movements and in the method of application. Experiments which test the normalisation circuitry of laser Doppler instruments are described and the effects of movement artefacts on the interpretation of the pulsatile component of laser Doppler records are also discussed. Probe and fibre line movements cause high-frequency intensity fluctuations due to speckle movement. The intensity fluctuations produce an apparent Doppler shift much greater than the Doppler shift produced by the relative movements of probe and tissue. It has been found that it is important to ensure that the fields of view of the illuminating and detecting fibres do not overlap at the skin surface and that probe contact with the skin surface should be maintained.  相似文献

BACKGROUND: Patients with familial adenomatous polyposis have a nearly 100 percent risk of colorectal cancer. In this disease, the chemopreventive effects of nonsteroidal antiinflammatory drugs may be related to their inhibition of cyclooxygenase-2. METHODS: We studied the effect of celecoxib, a selective cyclooxygenase-2 inhibitor, on colorectal polyps in patients with familial adenomatous polyposis. In a double-blind, placebo-controlled study, we randomly assigned 77 patients to treatment with celecoxib (100 or 400 mg twice daily) or placebo for six months. Patients underwent endoscopy at the beginning and end of the study. We determined the number and size of polyps from photographs and videotapes; the response to treatment was expressed as the mean percent change from base line. RESULTS: At base line, the mean (+/-SD) number of polyps in focal areas where polyps were counted was 15.5+/-13.4 in the 15 patients assigned to placebo, 11.5+/-8.5 in the 32 patients assigned to 100 mg of celecoxib twice a day, and 12.3+/-8.2 in the 30 patients assigned to 400 mg of celecoxib twice a day (P=0.66 for the comparison among groups). After six months, the patients receiving 400 mg of celecoxib twice a day had a 28.0 percent reduction in the mean number of colorectal polyps (P=0.003 for the comparison with placebo) and a 30.7 percent reduction in the polyp burden (the sum of polyp diameters) (P=0.001), as compared with reductions of 4.5 and 4.9 percent, respectively, in the placebo group. The improvement in the extent of colorectal polyposis in the group receiving 400 mg twice a day was confirmed by a panel of endoscopists who reviewed the videotapes. The reductions in the group receiving 100 mg of celecoxib twice a day were 11.9 percent (P=0.33 for the comparison with placebo) and 14.6 percent (P=0.09), respectively. The incidence of adverse events was similar among the groups. CONCLUSIONS: In patients with familial adenomatous polyposis, six months of twice-daily treatment with 400 mg of celecoxib, a cyclooxygenase-2 inhibitor, leads to a significant reduction in the number of colorectal polyps.  相似文献

Summary Rates of 5-hydroxytryptamine synthesis in various brain tissues can be estimated from the linear increase in 5-hydroxytryptophan levels following inhibition of 5-hydroxytryptophan decarboxylation with RO4-4602 or NSD-1015. In addition, NSD-1015 can prevent 5-hydroxytryptamine oxidative-deamination via monoamine oxidase inhibition, leading to linear decreases in a major metabolite of this amine, 5-hydroxyindole acetic acid. In the rat pineal gland we demonstrated similar increases in 5-hydroxytryptophan levels after nocturnal or diurnal injection of RO4-4602 (100 mg · kg–1) or NSD-1015 (200 mg · kg–1). Similar decreases in 5-hydroxyindole acetic acid were also observed after nocturnal or diurnal injection of NSD-1015 or pargyline (an inhibitor of monoamine oxidase) (75 mg · kg–1). 5-Hydroxytryptamine levels increased after nocturnal pargyline injection but remained constant after diurnal pargyline administration. 5-Hydroxytryptamine levels exhibited little change following nocturnal injection of NSD-1015 but decreased linearly after diurnal injection of NSD-1015. We suggest that (1) rat pineal 5-hydroxytryptamine synthesis is increased nocturnally, (2) metabolic utilization, primarily by oxidative-deamination, of 5-hydroxytryptamine is increased diurnally and (3) basal levels of pineal 5-hydroxytryptamine may be stored within adrenergic nerve endings which innervate the pinealocytes responsible for synthesizing this amine, thus protecting or otherwise making unavailable this pool of 5-hydroxytryptamine for metabolic utilization.  相似文献

Serotonin and 5-hydroxyindoleacetic acid were measured in the pineal gland of Syrian hamsters by high performance liquid chromatography over a period of 24 h. A distinct 24 h rhythm was detected for both indoles. Turnover studies revealed a higher rate of serotonin synthesis during the day than during the night. We therefore suggest that the serotonin content in the pineal gland of the Syrian hamster is regulated by changes in its synthesis rate, rather than by changes of serotonin catabolism, via N-acetyltransferase activity.  相似文献

Differentiation of extraembryonic mesoderm in the rhesus monkey was studied from the epithelial penetration stage of implantation (stage 4) through the first week of postimplantation development (to stage 6). It was found that the first cells that appeared between the primitive endoderm (hypoblast) and trophoblast were separated from the latter by a basal lamina but appeared to be either loosely attached to the endoderm or to have been detached from it. Cells in this intermediate position differentiated cytologically into mesenchymal cells, which, by stage 5, had a distinctive intraendoplasmic reticulum marker. This differentiation occurred prior to the time at which the primitive streak could be recognized. By the time the primitive streak was readily discernible (stage 6), the extraembryonic mesoderm had already produced substantial extracellular matrix. The sequence of differentiation was repeated, with a 1- to 2-day lag, in the secondary implantation site. No evidence of a contribution from cytotrophoblast or primitive streak to the extraembryonic mesoderm was found. It is concluded that the origin of the first extraembryonic mesoderm in the rhesus monkey is probably a two-step process, with formation of a reticulum from primitive endoderm followed by differentiation in situ into mesenchymal cells. The first blood vessels formed also differentiated in situ from the extraembryonic mesenchymal cells. Primitive capillaries were identifiable as early as the 13th day of pregnancy.  相似文献
