Objective
To investigate the expression of antigen processing-1 (Tap-1) and Tapasin in oral squamous cell carcinoma (OSCC), and observe the immune response against OSCC by use of IFN-γ-antigen induced dendritic cells (DCs) in vitro and in vivo.Design
Expression of Tap-1 and Tapasin in different cell lines was analysed. CAL27 cells were treated with IFN-γ. Antigen from the treated cells was presented by DCs. Pulsed DC was then co-cultivated with CD8+ T lymphocyte to induce antigen specific cytotoxic lymphocytes (CTLs). The immune response elicited by CTLs against OSCC was observed.Results
A significant lower expression of Tap-1 and Tapasin was observed in OSCC cell lines. IFN-γ exerted time-dependent effect for increasing the expression of these genes. Antigen from the treated CAL27 cells was presented by DCs. CTLs were induced and generated a strong immune response in vitro and in vivo.Conclusions
Tap-1 and Tapasin were downregulated in OSCC. IFN-γ increased the expression of these genes. Use of IFN-γ-antigen induced DCs could induce stronger immune response in vitro and in vivo. 相似文献22.
目的:探讨玻璃渗透铝瓷基底厚度变化对In-Ceram全瓷修复体颜色影响规律,为临床选色、配色提供实验依据.方法:分别制作不同厚度的铝瓷基底,并在此基础上烧结In-Ceram全瓷试件,采用标准D65光源,使用Data Color SF300型分光光度仪,观测玻璃渗透铝瓷基底厚度变化对In-Ceram全瓷修复体的明度、色相、彩度的影响.结果:当不透明牙本质瓷和牙本质瓷厚度固定时,玻璃渗透铝瓷基底厚度的改变能引起颜色的变化.当铝瓷基底厚度在0.5~0.7 mm时,符合临床比色要求,能够较好的提高修复体的颜色复现率.结论:通过对铝瓷基底厚度的调节,可以使In-Ceram全瓷修复体获得较理想的色泽,取得较好的修复效果. 相似文献

Expression and significance of stromal cell-derived factor-1alpha and its receptor CXCR4 in human dental pulp cells 总被引:1,自引:0,他引:1
OBJECTIVE: To investigate the expression of CXCR4 in cultured human dental pulp cells (HDPC) in vitro and the corresponding ligand SDF-1alpha level of HDPC supenatants stimulated by lipopolysaccharide (LPS) and tumor necrosis factor-alpha (TNF-alpha), and to explore the role of SDF-1alpha on the proliferation and the migration of HDPC. METHODS: The expression of CXCR4 in HDPC was detected by immunocytochemistry technique and indirect immunofluorescence technique. The culture supernatants of HDPC were collected after HDPC had been simulated by LPS and TNF-alpha of different concentrations for 48h and then the SDF-1alpha level was assayed by quantitative sandwich ELISA. Meanwhile, the effects of recombinant human SDF-1alpha (rhSDF-1alpha) on the proliferation and the migration of HDPC at different concentrations were observed by MTT and Boyden Chamber Assay. RESULTS: CXCR4 was expressed in cytomembrane of HDPC and SDF-1alpha was secreted into their normal cell supernatants with a concentration of (4513.55 +/- 962.92) ng/L. The secretion of SDF-1alpha was both significantly decreased by stimulation with LPS and TNF-alpha (P < 0.05). In addition, rhSDF-1alpha stimulated the HDPC proliferation at the concentrations of 50, 100, 200 microg/L (P < 0.01) and increased the chemotactic migration of HDPC significantly after 9h's incubation with the concentrations of 50, 100 microg/L (P < 0.05). CONCLUSIONS: SDF-1alpha accelerated the proliferation and the migration of HDPC which expressed CXCR4. SDF-1-CXCR4 axis may play a role in repair of pulp injury. 相似文献

本研究对13例人的正常涎腺经三种固定双固定后的组织标本进行了11种角蛋白单克隆抗体的免疫组化染色.其中5例腮腺和3例颌下腺分别同时用常规Formalin固定,Bouin’s液固定,Carnoy‘s液固定.染色采用ABC法,结果发现:Carnoy固定者,K_8、K_18、K_19和K_8.12的染色效果最好.其导管染色比Bouin‘s和Formalin固定者染色强而明确,尤其K_8和K_8.12不仅在导管中呈现强阳性,而且腺泡也呈阳性;而K_(17)和K_(L1)没有看出明显的染色差别;K_(14)、K_(10)、K_(13)、K_(17)、K_(20)的染色几乎全为阴性,与常规中性缓冲Formalin固定者结果一致. 相似文献

Bleaching tetracycline-stained teeth is the most challenging form of tooth lightening. This article reports on 44 subjects who bleached their tetracycline-stained teeth for 6 months using trays with reservoirs overnight in a half-mouth designed study and 2 of 3 different concentrations of carbamide peroxide (10%, 15% or 20%). The subjects were followed for 5 years. The area evaluated was the middle third of the teeth. More than 55% of tooth lightening occurred within 1 month; after 5 years, more than 65% of the maximum tooth whitening remained for all 3 gel concentrations. Objectives
To investigate the effects of acidic and neutral 30% hydrogen peroxide (HP) on human tooth enamel in terms of chemical structure, mechanical property, surface morphology and tooth colour.Methods
Twenty-seven human dental blocks were obtained from premolars and randomly divided into three groups (n = 9): Group acidic HP (30% HP, pH ≈ 3.6), Group neutral HP (30% HP, pH ≈ 7.0) and Group DW (distilled water, pH ≈ 6.8). Attenuated total reflection-Fourier transform infrared spectroscopy (ATR-FTIR), Raman spectroscopy, atomic force microscopy (AFM) investigation, microhardness test and colour measurements were carried out before and after treatments. ATR-FTIR and Raman spectroscopy were analysed and then the carbonate:mineral ratio (C:M), Raman absolute intensity (RAI), Raman relative intensity (RRI), and laser-induced fluorescence intensity (FI) were obtained for evaluation.Results
The C:M, percentage microhardness and percentage RRI of group acidic HP decreased more significantly than those of group neutral HP (P = 0.02, P = 0.001, P < 0.001, respectively) and group DW (P = 0.01, P = 0.008, P < 0.001, respectively). Whilst group neutral HP and group DW had no statistical difference in above terms (P = 0.818, P = 0.528, P = 0.158, respectively). Significant morphological alterations were observed in group acidic HP. Group acidic HP and neutral HP had no significant difference in percentage FI (P = 0.652) and ΔE (P = 0.906).Conclusions
This study suggested that neutral 30% HP had the same efficiency in tooth bleaching and it caused less deleterious effects on enamel than acidic 30% HP. 相似文献27.
