Background  

Bone marrow transplantation (BMT) is often used in the treatment of various diseases. Before BMT, patients are submitted to a conditioning regimen (CR), which consists of the administration of high doses of chemotherapy. The action of many cytostatic drugs involves the overproduction of reactive oxygen species, which together with inadequate antioxidant protection can lead to oxidative stress and this has been implicated in the etiology of various diseases. The objectives of this study were to look for evidence of oxidative stress and also to analyze δ-Aminolevulinato dehydratase (δ-ALA-D) activity as a possible marker of oxidative stress in autologous and allogeneic BMT patients.  相似文献

Objectives

To evaluate the effect of different glass-ceramic surface treatments and aging on the fatigue failure load of a lithium disilicate glass-ceramic adhesively cemented to a dentin analogue material.

Methods

One hundred and twenty (120) disc-shaped lithium disilicate specimens (Ø = 10 mm, thickness = 1.5 mm) were produced and randomly allocated (n = 20) into 6 groups, considering 2 study factors: “surface treatment” in 3 levels (SIL—silane application only; HF5+SIL—5% hydrofluoric acid etching and silane application; ME&P—etching with an one-step ceramic primer), and “storage” in 2 levels (baseline—storage for 7 days; aging—storage for 90 days + 12,000 thermal cycles). Ceramic discs were adhesively cemented to discs of a dentin analogue material (Ø = 10 mm, thickness = 2.0 mm) following the manufacturers’ instructions. The fatigue failure load was determined by the staircase approach (250,000 cycles; 20 Hz; initial load = 1050 N [~70% of mean load-to-failure]; step size = 52.5 N [5% of initial load]). Micro-morphologic, fractographic, and atomic force microscope analysis were also performed. Fatigue failure load data were evaluated by one-way ANOVA, Bonferroni and t-tests for independent samples.

Results

HF5+SIL presented higher fatigue failure load in both conditions (baseline and aging); ME&P presented intermediary mean values, while the SIL group presented the worst performance. All groups had a statistically significant decrease in the fatigue performance after aging.

Significance

Hydrofluoric acid followed by silane application showed the best fatigue performance for an adhesively-cemented lithium disilicate ceramic. Aging negatively influenced the fatigue performance for all tested groups.  相似文献

Essential oils obtained from leaves of 29 medicinal plants commonly used in Brazil were screened against 13 different Escherichia coli serotypes. The oils were obtained by water-distillation using a Clevenger-type system and their minimal inhibitory concentration (MIC) were determined by microdilution method. Essential oil from Cymbopogon martinii exhibited a broad inhibition spectrum, presenting strong activity (MIC between 100 and 500 microg/mL) against 10 out of 13 Escherichia coli serotypes: three enterotoxigenic, two enteropathogenic, three enteroinvasive and two shiga-toxin producers. C. winterianus inhibited strongly two enterotoxigenic, one enteropathogenic, one enteroinvasive and one shiga-toxin producer serotypes. Aloysia triphylla also shows good potential to kill Escherichia coli with moderate to strong inhibition. Other essential oils showed antimicrobial properties, however with a more restricted action against the serotypes studied. Chemical analysis of Cymbopogon martinii essential oil performed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC-MS) showed the presence of compounds with known antimicrobial activity, including geraniol, geranyl acetate and trans-cariophyllene, which tested separately, indicated geraniol as antimicrobial active compound. The significant antibacterial activity of Cymbopogon martinii oil suggests that they could serve as a source for compounds with therapeutic potential.  相似文献

We investigated the vascular effects of a crude aqueous extract (AEEG) of Echinodorus grandiflorus (Alismataceae) using the in vitro experimental models of the rabbit isolated aorta and perfused kidney. Echinodorus grandiflorus, a native semi-aquatic plant widely distributed in Brazil, has been extensively used in Brazilian folk medicine for the treatment of high blood pressure and inflammatory diseases. The bolus injection of AEEG (0.1-10 mg) into the rabbit renal circulation pre-contracted with norepinephrine induced marked and dose-dependent vasodilator responses (maximum of 37+/-4%; n=6; P<0.001), which was similar to that induced by injection of 10 mmol acetylcholine (41+/-3%). Moreover, AEEG elicited a significant and concentration-dependent relaxation in the endothelium-intact, but not endothelium-denuded aortic rings, reaching the maximum of 81+/-5% (n=7, P<0.001). Inhibition of the nitric oxide-cGMP pathway with L-NAME (100 microM) or Methylene Blue (20 microM) reduced maximum relaxation induced by AEEG from 81+/-5% to 46+/-3 and 45+/-3%, respectively (n=7, P<0.001). A similar reduction was obtained with the incubation of the aortic rings with the selective PAF receptor antagonist WEB 2086 (10 microM) (from 81+/-5% to 55+/-3%; n=7; P<0.01). Conversely, blockade of muscarinic receptors with atropine (10 microM) did not affect the vasodilator effects of AEEG, while inhibition of the enzyme cyclooxigenase not only did not block, but rather potentiated vasodilation induced by AEEG (n=7, P<0.001). Finally, blockade of Ca(2+)- and ATP-activated K(+) channels using the specific blockers charydbotoxin (100 nM) and glibenclamide (3 microM), respectively, did not modify aortic relaxation induced by AEEG. We conclude that water-soluble extracts from leaves of Echinodorus grandiflorus elicit an endothelium-dependent, nitric oxide and PAF receptor-mediated vasodilation in rabbit aortic rings, which does not appear to involve the generation of vasodilating prostaglandins or the activation of K(+) channels. This potent vasodilator effect of the extracts was confirmed in the isolated rabbit renal circulation.  相似文献

This study evaluated the influence of surface treatment on the shear bond strength of a composite resin (CR), previously submitted to the application of a temporary cement (TC), to an adhesive luting cement. Eight-four CR cylinders (5 mm diameter and 3 mm high) were fabricated and embedded in acrylic resin. The sets were divided into 6 groups (G1 to G6) (n=12). Groups 2 to 6 received a coat of TC. After 24 h, TC was removed and the CR surfaces received the following treatments: G2: ethanol; G3: rotary brush and pumice; G4: air-abrasion; G5: air-abrasion and adhesive system; G6: air-abrasion, acid etching and adhesive system. G1 (control) did not receive TC or any surface treatment. The sets were adapted to a matrix and received an increment of an adhesive luting cement. The specimens were subjected to the shear bond strength test. ANOVA and Tukey's tests showed that G3 (8.53 MPa) and G4 (8.63 MPa) differed significantly (p=0.001) from G1 (13.34 MPa). The highest mean shear bond strength values were found in G5 (14.78 MPa) and G6 (15.86 MPa). Air-abrasion of CR surface associated with an adhesive system provided an effective bond of the CR to the adhesive luting cement, regardless the pre-treatment with the phosphoric acid.  相似文献
