Id3-/- mice represent a model for T cell mediated primary Sjogren's syndrome (PSS). An intriguing feature of this disease model is the early appearance of impaired salivary function or exocrinopathy prior to lymphocytic infiltration of the salivary glands. This phenomenon prompted us to examine the role of cytokines produced by T cells in the systemic regulation of gland function. A comprehensive examination of serum cytokine profiles revealed elevated levels of IL-13 in Id3-/- mice. We found that the increase in serum IL-13 levels in Id3-/- mice was largely dependent on αβ T cells. Removal of αβ T cells in Id3-/- mice also eliminates disease symptoms, including lymphocytic infiltration in the gland tissues, and impaired saliva production. We further show that the number of mast cells in the salivary glands of Id3-/- mice is significantly increased, in a trend inversely related to the saliva production. This increase in the number of mast cells is also dependent on the presence of αβ T cells. Treatment of young Id3-/- mice with anti-IL-13 antibodies over a two-month period resulted in a reduction of both serum IL-13 levels and the number of mast cells in the salivary gland tissues, as well as correspondingly improved saliva production. Nucleocapsid (N) protein of porcine reproductive and respiratory syndrome virus (PRRSV) plays a central role in virus replication. In this study, serial N- and C-terminal truncations of N protein were performed in the context of type 2 PRRSV infectious cDNA clone, and our results revealed that a stretch of inter-genotypic variable N terminal residues aa 5-13 ((5)NGKQQKKK(13)K) and the last four inter-genotypic variable aa residues ((120)SPS(123)A) at the C terminus of N protein were dispensable for type 2 PRRSV infectivity. All the recovered deletion mutant viruses had spontaneous mutations in the N coding region, including substitution, deletion and insertion. We re-engineered the additional internal deletion with or without the original C-terminal deletion back into wild-type APRRS and found that the internal domain spanning the inter-genotypic variable residues 39-42 ((39)KGP(42)G) and conserved residues 48-52 ((48)KNPE(52)K), respectively, were dispensable for type 2 PRRSV viability. These results demonstrated that N protein contains non-essential regions for virus viability in cell culture. Such dispensable regions could be utilized as insertion site for foreign tag expression and the rescued viruses could be the candidates for marker vaccine. 相似文献

Th17 cells represent a novel subset of CD4(+) T cells, which is associated with chronic inflammation. The present study evaluated Th17 cell responses to Helicobacter pylori infection in mouse model and CD4(+) T cell differentiation in response to H. pylori-infected macrophages. Th17 cells were observed in the H. pylori-infected gastric tissue. Co-culture of CD4(+) T cells with H. pylori-infected macrophages elevated IL-17 and IFN-γ secretion, up-regulated retinoid-related orphan receptor gamma t (RORγt) and T box expressed in T cells (T-bet) expression and increased the numbers of Th17 and Th1 cells. The expression of CD40, CD80, and CD86 and the secretion of IL-6, TGF-β1, IL-23, and CCL20 were significantly increased in H. pylori-stimulated macrophages. NF-κB pathway participated in the production of IL-6, IL-23, and CCL20 from macrophages in response to H. pylori, and inhibition of NF-κB pathway of macrophages resulted in less Th17 cell differentiation. 目的建立小柴胡汤滴丸的质量标准。方法采用薄层色谱(TLC)法对小柴胡汤滴丸中的柴胡、黄芩进行薄层色谱定性鉴别,采用HPLC法对制剂中黄芩的黄芩苷进行含量测定,采用Hypemil C18色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.2%磷酸溶液(45∶55),检测波长为280 nm,柱温30℃。结果薄层鉴别的色谱斑点清晰,阴性对照无干扰;黄芩苷在0.1~1.2μg/mL范围内呈良好线性关系,平均回收率为99.20%,RSD为2.29%(n=6)。结论本方法可用于小柴胡汤滴丸的质量控制,具有简便、准确及专属性强的特点。 相似文献

目的: 探讨脾脏对链脲菌素(STZ)损伤胰岛β细胞是否具有保护作用。方法: 以外科手术切除小鼠全脾。60只脾切除小鼠随机分为3组,分别以80 mg/kg、160 mg/kg STZ给小鼠腹腔注射,另一组腹腔注射生理盐水。60只正常小鼠做同样分组及处理。1周后,测定各组小鼠空腹血糖、血清胰岛素水平,免疫组化分析各组胰岛β细胞总量,ELISA法分析胰岛细胞凋亡,Luminol化学发光法测定各组胰腺活性氧簇(ROS)水平。结果: 80 mg/kg STZ处理脾切除组空腹血糖浓度显著升高,血清胰岛素显著降低;同剂量STZ处理的正常小鼠血糖及胰岛素水平则正常;此外, 80 mg/kg STZ处理的脾切除小鼠β细胞总量、胰岛凋亡细胞核小体聚集值及胰腺组织ROS的产生与同剂量STZ处理的正常小鼠相比均有显著差异。结论: 脾可以阻止低剂量STZ对正常小鼠胰岛β细胞的破坏作用。完全脾切除增加了胰岛对STZ的敏感性,这一作用与胰腺组织ROS增加相关。 相似文献
