Objective To investigate the effect of SiO2 on the expression of matrix metalloproteinase-9 (MMP-9)and tissue inhibitor of metalloproteinase-1 (TIMP-1)in human silicofic alveolar macrophages (AM).Methods Human alveolar macrophages were collected from a silicotic patient by bronchoalveolar lavage and exposed to silicon dioxide for 3h,6h,12h,18h,24h and 36h.The expression of the MMP-9 in the AM were detected with zymography and immunological method and the expression of the TIMP-1 in the AM with immunological method.Results The expressions of MMP-9 in the AM increased clearly along with the time,reached peak at 24 h when detected with zymography (average optical density:3.061±0.153 vs 2.851±0.164,P<0.05);and reached peak at 18h when detected with immunological method (average optical density:0.386±0.037 vs 0.322±0.034,P<0.05).The expression of the TIMP-1 in the AM did not vary when detected with immunological method (P>0.05).Conclusion SiO2 may affect the expression of MMP-9 and MMP-9 activity in the cultured AM. 相似文献

目的探讨益气活血法治疗充血性心力衰竭(CHF)理论机制及临床疗效.方法将患者 90例随机分为益气活血组、活血化瘀组、益气组和对照组,所有患者均给予常规西医治疗,并分别予益气活血(益气活血组)、活血化瘀(活血化瘀组)、益气(益气组 )之剂和单纯西医治疗(对照组).结果益气活血组疗效及总有效率均明显优于对照组,其它各组间疗效比较无显著性差异;益气活血组、活血化瘀组和益气组 Boston积分差值均显著高于对照组,此三组组间疗效相近;各组治疗后左室射血分数 (LVEF)均较治疗前显著升高,组间比较无显著性差异.结论 CHF主要中医证型为气虚血瘀,治疗应以益气活血为大法. 相似文献

目的制备隐丹参酮固体分散体,提高隐丹参酮的溶出度。方法采用溶剂蒸发法制备隐丹参酮-PVP固体分散体,熔融法制备隐丹参酮-PEG固体分散体,利用体外溶出度、差热分析、显微观察研究固体分散体的性质及其对溶出度的影响。结果PVP及PEG固体分散体在45min的溶出度分别达到原料药的9.7倍和7.5倍,固体分散体的DTA曲线中隐丹参酮的特征熔融峰消失。结论两种固体分散体均能显著提高隐丹参酮的溶出度,而PVP固体分散体比PEG固体分散体具有更高的溶出度。 相似文献

In this study, dietary modulation of 7,12-dimethylbenz[a]anthracene (DMBA)-induced adrenal toxicity in rats was investigated. Beginning at postnatal day (PND) 21, female Sprague-Dawley rats were fed either soy-containing NIH-31 diet or soy- and alfalfa-free 5K96 diet. On the first day of diestrus when the animals were PND 50 +/- 5, rats received either an oral dose of 80 mg/kg DMBA or sesame oil, the vehicle, and were sacrificed at 24, 36, or 48 h after treatment. Apoptosis was manifested at 24 and 36 h after DMBA treatment in the zona reticularis (ZR) and the zona fasciculata (ZF) of the adrenal cortex; this was followed by severe hemorrhagic necrosis at 48 h. DMBA-induced apoptosis, evaluated by the TUNEL assay, immunohistochemical analysis of activated caspase 3, and the ratio of expression of pro-apoptotic Bax to anti-apoptotic Bcl2, was greater in rats fed NIH-31 diet relative to rats fed 5K96 diet at 24 h after treatment. Four of six DMBA-treated rats fed 5K96 diet had severe adrenal necrosis by 48 h, whereas this lesion was present in only two of six DMBA-treated rats fed NIH-31 diet. DMBA also caused a significant decrease of serum corticosterone relative to controls at 48 h in rats fed 5K96 diet. The present study indicated that diet modulates DMBA-induced adrenal toxicity in female rats, with increased apoptosis early and reduced necrosis later in rats fed a soy-containing diet. 相似文献

Human UDP-glucuronosyltransferase (UGT) 2B7 is the major isoform catalyzing the glucuronidation of a variety of endogenous compounds including bile acids. To determine the role of bile acids in the regulation of UGT2B7 expression, Caco-2 cells were incubated with the natural human farnesoid X receptor (hFXR) ligand, chenodeoxycholic acid, as well as the secondary bile acid, lithocholic acid, derived from chenodeoxycholic acid. Incubation of Caco-2 cells with lithocholic acid in the absence of exogenous hFXR resulted in a dose-dependent down-regulation of UGT2B7 mRNA levels, with an IC(50) of 13 microM. Similar down-regulation was also observed with chenodeoxycholic acid; however, much higher concentrations were required. Transient transfection of Caco-2 cells with hFXR suppressed UGT2B7 mRNA expression both in the absence and presence of ligand. UGT2B7 promoter transfection experiments and deletion/mutation analysis showed that lithocholic acid-activated hFXR decreased UGT2B7 promoter activity via a negative hFXR response element (NFRE) located between nucleotides -148 and -134. Cotransfection with hFXR and/or human retinoid X receptor further enhanced the repression. Electrophoretic mobility shift assays additionally confirmed the role of NFRE in UGT2B7 down-regulation by lithocholic acid. These findings suggest that lithocholic acid, an activator of nuclear hFXR, acts as a negative regulator of UGT2B7 expression, indicating that hFXR may play an essential role in lithocholic acid homeostasis through negative regulation of this UGT that is involved in lithocholic acid biotransformation. Therefore, it is postulated that lithocholic acid toxicity may be due to down-regulation of genes involved in its detoxification, including UGT2B7, leading to limited excretion of lithocholic acid from the body. 相似文献
