Tripartite motif protein 21 (TRIM21) is an interferon-inducible E3 ligase, containing one RING finger domain, one B-box motif, one coiled-coil domain at the N-terminal, as well as one PRY domain and one SPRY domain at the C-terminal. TRIM21 is expressed in many tissues and plays an important role in systemic autoimmunity. However, TRIM21 plays different roles in different virus infections. In this study, we evaluate the relationship between porcine TRIM21 and PCV2 infection as well as host immune responses. We found that PCV2 infection modulated the expression of porcine TRIM21. TRIM21 can enhance interferons and proinflammatory factors and decrease cellular apoptosis in PCV2-infected cells. These results indicate that porcine TRIM21 plays a critical role in enhancing PCV2 infection, which is a promising target for controlling and developing the treatment of PCV2 infection. 相似文献

BackgroundThalamocortical circuit imbalance characterized by prefronto-thalamic hypoconnectivity and sensorimotor-thalamic hyperconnectivity has been consistently documented at rest in schizophrenia (SCZ). However, this thalamocortical imbalance has not been studied during task engagement to date, limiting our understanding of its role in cognitive dysfunction in schizophrenia.MethodsBoth n-back working memory (WM) task-fMRI and resting-state fMRI data were collected from 172 patients with SCZ and 103 healthy control subjects (HC). A replication sample with 49 SCZ and 48 HC was independently obtained. Sixteen thalamic subdivisions were employed as seeds for the analysis.ResultsDuring both task-performance and rest, SCZ showed thalamic hyperconnectivity with sensorimotor cortices, but hypoconnectivity with prefrontal-cerebellar regions relative to controls. Higher sensorimotor-thalamic connectivity and lower prefronto-thalamic connectivity both relate to poorer WM performance (lower task accuracy and longer response time) and difficulties in discriminating target from nontarget (lower d′ score) in n-back task. The prefronto-thalamic hypoconnectivity and sensorimotor-thalamic hyperconnectivity were anti-correlated both in SCZ and HCs; this anti-correlation was more pronounced with less cognitive demand (rest>0-back>2-back). These findings replicated well in the second sample. Finally, the hypo- and hyper-connectivity patterns during resting-state positively correlated with the hypo- and hyper-connectivity during 2-back task-state in SCZ respectively.ConclusionsThe thalamocortical imbalance reflected by prefronto-thalamic hypoconnectivity and sensorimotor-thalamic hyperconnectivity is present both at rest and during task engagement in SCZ and relates to working memory performance. The frontal reduction, sensorimotor enhancement pattern of thalamocortical imbalance is a state-invariant feature of SCZ that affects a core cognitive function. 相似文献

This research aims to explore the high-temperature and low-temperature performances of lignin–waste engine oil-modified asphalt binder and its mixture. For this research, the lignin with two contents (4%, 6%) and waste engine oil with two contents (3%, 5%) were adopted to modify the control asphalt binder (PG 58-28). The high-temperature rheological properties of the lignin–waste engine oil-modified asphalt binder were investigated by the viscosity obtained by the Brookfield viscometer and the temperature sweep test by the dynamic shear rheometer. The low-temperature rheological property of the lignin–waste engine oil-modified asphalt binder was evaluated by the stiffness and m-value at two different temperatures (−18 °C, −12 °C) obtained by the bending beam rheometer. The high-temperature and the low-temperature performances of the lignin–waste engine oil-modified asphalt mixture were explored by the rutting test and low-temperature bending beam test. The results displayed that the rotational viscosity and rutting factor improved with the addition of lignin and decreased with the incorporation of waste engine oil. Adding the lignin into the control asphalt binder enhanced the elastic component while adding the waste engine oil lowered the elastic component of the asphalt binder. The stiffness of asphalt binder LO60 could not meet the requirement in the specification, but the waste engine oil made it reach the requirement based on the bending beam rheometer test. The waste engine oil could enhance the low-temperature performance. The dynamic stabilities of LO40- and LO60-modified asphalt mixture increased by about 9.05% and 17.41%, compared to the control mixture, respectively. The maximum tensile strain of LO45 and LO65 increased by 16.39% and 25.28% compared to that of LO40 and LO60, respectively. The high- and low-temperature performances of the lignin–waste engine oil-modified asphalt LO65 was higher than that of the control asphalt. The dynamic stability had a good linear relationship with viscosity, the rutting factor of the unaged at 58 °C, and the rutting factor of the aged at 58 °C, while the maximum tensile strain had a good linear relationship with m-value at −18 °C. ObjectiveTo evaluate the risk factors for dysphagia after anterior cervical discectomy and fusion (ACDF) with the Zero‐P Implant System by multidimensional analysis and investigated the predictive values of these risk factors for dysphagia.MethodsA retrospective analysis of 260 patients who underwent ACDF with the Zero‐P Implant System and had at least 2 year of follow‐up were performed. All patients were divided into a non‐dysphagia group and a dysphagia group. Sex, age, body mass index (BMI), intraoperative time, estimated blood loss, diabetes mellitus, hypertension, smoking, alcohol consumption, prevertebral soft‐tissue thickness, the levels of surgery, O‐C2 angle, C2–7 angle, T1 slope and segmental angle were analyzed. The Modified Japanese Orthopaedic Association (JOA) scoring system was used to determine functional status. NDI was used to evaluate neck pain and disability. The Bazaz grading system was chosen to evaluate dysphagia after surgery. Postoperative cerebrospinal fluid (CSF) leakage, infection, and dysphagia were recorded in both groups. An independent t‐test was used to compare quantitative variables, a chi‐square test was used to compare qualitative data between the two groups. To eliminate the influence of confounding factors, logistic regression was performed for multifactor regression of factors. The results were regarded as significant when the P‐values were less than 0.05 in this study.ResultsIn total, the non‐dysphagia group comprised 70 patients and the dysphagia group comprised 190 patients, with an average age of 58.33 ± 4.68 years (ranging, 42–82 years). These patients were followed up for 28.5 ± 3.5 months (range, 24–32 months). For clinical outcomes, both groups demonstrated significant improvement in the NDI and JOA scores (P < 0.001). According to the Bazaz dysphagia grading system, mild, moderate, and severe dysphagia were found in 50, 17, and 3 patients, respectively. In total, 37.1% (n = 26) had resolved by 3 month, 38.6% (n = 27) by 6 months, and 17.1% (n = 12) by 12 months. Chi‐square test results indicated that number of operated levels, operation time dT1 slope, dO‐C2 angle, dC2–7 angle, segmental angle and dPSTT were associated with a high incidence of dysphagia. Multivariate logistic regression analysis showed that number of operated levels, operation time, dC2–7 angle and dPSTT were significantly associated with postoperative dysphagia.ConclusionsMore operated levels, more operation time, more dC2–7 angle and dPSTT were the risk factors for postoperative dysphagia. In additional, sufficient preoperative preparation, evaluation combined with proficient and precise surgical treatment were suggested to reduce the incidence of postoperative dysphagia when ACDF was performed. 相似文献

目的 基于磁共振血管造影(MRA)的椎基底动脉迂曲扩张症(VBD)计算机流体力学分析。方法 选取2018年1月至2022年1月牡丹江医学院附属红旗医院诊断为VBD的40例患者,经MRI检查正常的40例患者图像。以椎基底动脉MRA图像构建椎基底动脉三维几何模型,分为VBD组与对照组,分析血流速度(V)、壁面剪切应力(WSS)及震荡剪切系数(OSI)。结果 VBD组V、WSS及OSI均高于对照组,差异有统计学意义(P <0.05)。在血流速度方面,高速血流集中于基底动脉及右侧大脑后动脉处。在WSS方面,VBD组高WSS位于右侧椎动脉、基底动脉、双侧大脑后动脉等大部分血管区域,以血管分叉处显著。在OSI方面,VBD组高OSI值以基底动脉末端至双侧大脑后动脉发出处明显,其余血管区域为波动性高OSI分布。结论 基于真实磁共振患者图像可建立三维个体化血管模型及进行血流动力学分析,获取VBD血流速度图、血管剪切应力云图及震荡剪切系数云图。建立VBD组及对照组模型,分析血流动力学发现,VBD组的基底动脉段具有高速血流、高WSS及高OSI,以末端显著,临床上应着重关注此区域。 相似文献
