Sleep is homeostatically regulated in all animal species that have been carefully studied so far. The best characterized marker of sleep homeostasis is slow wave activity (SWA), the EEG power between 0.5 and 4 Hz during nonrapid eye movement (NREM) sleep. SWA reflects the accumulation of sleep pressure as a function of duration and/or intensity of prior wake: it increases after spontaneous wake and short-term (3–24 h) sleep deprivation and decreases during sleep. However, recent evidence suggests that during chronic sleep restriction (SR) sleep may be regulated by both allostatic and homeostatic mechanisms. Here, we performed continuous, almost completely artifact-free EEG recordings from frontal, parietal, and occipital cortex in freely moving rats (n = 11) during and after 5 d of SR. During SR, rats were allowed to sleep during the first 4 h of the light period (4S+) but not during the following 20 h (20S). During the daily 20S most sleep was prevented, whereas the number of short (<20 s) sleep attempts increased. Low-frequency EEG power (1–6 Hz) in both sleep and wake also increased during 20S, most notably in the occipital cortex. In all animals NREM SWA increased above baseline levels during the 4S+ periods and in post-SR recovery. The SWA increase was more pronounced in frontal cortex, and its magnitude was determined by the efficiency of SR. Analysis of cumulative slow wave energy demonstrated that the loss of SWA during SR was compensated by the end of the second recovery day. Thus, the homeostatic regulation of sleep is preserved under conditions of chronic SR.  相似文献

Thermo-exfoliated graphite (TEG)/CuO/Cu2(OH)3NO3:(Co2+/Fe3+) composites were prepared using a wet impregnation method and subsequent thermal treatment. The physicochemical characterization of the composites was carried out by powder X-ray diffraction (PXRD), scanning electron microscopy (SEM) and Ar temperature-desorption techniques. The catalytic efficiency toward CO conversion to CO2 was examined under atmospheric pressure. Characterization of species adsorbed over the composites taken after the activity tests were performed by means of temperature programmed desorption mass-spectrometry (TPD MS). (TEG)/CuO/Cu2(OH)3NO3:(Co2+/Fe3+) composites show superior performance results if lower temperatures and extra treatment with H2SO4 or HNO3 are used at the preparation stages. The catalytic properties enhancements can be related to the Cu2(OH)3NO3 phase providing reaction centers for the CO conversion. It has been found that prevalence of low-temperature states of desorbed CO2 over high-temperature ones in the TPD MS spectra is characteristic of the most active composite catalysts.  相似文献

Motivation for change and recovery from addiction has been commonly assessed using the Transtheoretical Model's stages of change. Analogous to readiness for change, this measure of motivation may not recognize other elements of motivation relevant to successful change. The aim of this study was to examine the relationship between stages of change and reasons for change according to the Self-Determination Theory among problem gamblers motivated to quit. Motivations for change were examined for 200 adult problem gamblers with intent to quit in the next 6 months (contemplation stage) or 30 days (preparation stage). Analyses revealed that higher autonomous motivation for quitting gambling predicted greater likelihood of being in the preparation stage, whereas those with higher external motivation for change were less likely to be farther along the stage of change continuum. The findings suggest that autonomous motivations relate to readiness for quitting gambling, and may predict successful resolution from problem gambling.  相似文献

Mycobacterium tuberculosis Beijing strains represent targets of special importance for molecular surveillance of tuberculosis (TB), especially because they are associated with spread of multidrug resistance in some world regions. Standard 24-locus mycobacterial interspersed repetitive-unit–variable-number tandem-repeat (MIRU-VNTR) typing lacks resolution power for accurately discriminating closely related clones that often compose Beijing strain populations. Therefore, we evaluated a set of 7 additional, hypervariable MIRU-VNTR loci for better resolution and tracing of such strains, using a collection of 535 Beijing isolates from six world regions where these strains are known to be prevalent. The typeability and interlaboratory reproducibility of these hypervariable loci were lower than those of the 24 standard loci. Three loci (2163a, 3155, and 3336) were excluded because of their redundant variability and/or more frequent noninterpretable results compared to the 4 other markers. The use of the remaining 4-locus set (1982, 3232, 3820, and 4120) increased the number of types by 52% (from 223 to 340) and reduced the clustering rate from 58.3 to 36.6%, when combined with the use of the standard 24-locus set. Known major clonal complexes/24-locus-based clusters were all subdivided, although the degree of subdivision varied depending on the complex. Only five single-locus variations were detected among the hypervariable loci of an additional panel of 92 isolates, representing 15 years of clonal spread of a single Beijing strain in a geographically restricted setting. On this calibrated basis, we propose this 4-locus set as a consensus for subtyping Beijing clonal complexes and clusters, after standard typing.  相似文献

Mycobacterium bovis caused 3% of human tuberculosis cases in southwest Ireland during 1998–2006. Of 11 M. bovis strains genotyped, 9 belonged to common animal spoligotypes. Seven strains were from sputum and potential sources of human-centered disease transmission. Ten-locus variable-number tandem repeat typing gave unique strain profiles and would detect disease outbreaks.  相似文献

Objectives and methodsThe Xpert® MTB/RIF assay (Cepheid, Sunnyvale, CA, USA) has been in routine use in Odessa Oblast, a region with the highest tuberculosis (TB) incidence in Ukraine, since 2013. We assessed the performance of the assay in routine settings and evaluated its effect on treatment outcomes.ResultsThe sensitivity of Xpert for TB detection was 93.7% (1165/1243) and 69.5% (448/645) for smear-positive and smear-negative sputum specimens, respectively, and its sensitivity for rifampicin resistance was 93.4% (1212/1298). Median time to TB detection using the Xpert assay was 0 days. Treatment initiation within 1 week increased the proportion of successful outcomes (60.1% versus 25.9%, RR = 1.86, 95%CI = 1.46–2.42), but the introduction of Xpert MTB/RIF has not led to a significant improvement in treatment outcomes (57.2% versus 46.2%; RR = 0.93, 95%CI = 0.77–1.12).ConclusionPerformance characteristics of the Xpert assay demonstrated during its routine implementation in an area of high TB and drug-resistant TB incidence in Ukraine were in line with those demonstrated in similar settings elsewhere. Rollout of rapid molecular testing may lead to better treatment results provided that it is implemented in conjunction with other programmatic improvements.  相似文献
