Diabetes mellitus is one of the most common serious metabolic disorders in humans that develops due to diminished production of insulin (type I) or resistance to its effect (type II and gestational). The present study was designed to determine the neuropsychological deficits produced following streptozotocin-induced diabetes in rats. Rats were made diabetic by the intra-peritoneal administration of 60 mg/kg streptozotocin (STZ) which induces type-1 diabetes by the destruction “β-cells” of pancreas. Body weight, food and water intake was monitored daily. Open field test (OFT) model, forced swim test (FST) and Morris water maze (MWM) model were performed for the evaluation of ambulation, depression-like symptoms and memory effects, respectively. After 10 days of diabetes induction the exploratory activity of rats was monitored by OFT while depression-like symptoms and memory effects in rats were analyzed by FST and MWM. Results showed that there was no significant effect of STZ-induced diabetes on body weight but food and water intake of STZ-induced diabetic rats was significantly increased. Exploratory activity was significantly decreased and short-term and long-term memory was significantly impaired while the depression-like symptoms was significantly increased in STZ diabetic rats. Thus, it may be suggested that STZ-induced diabetes alters the brain functions and may play an important role in the pathophysiology of certain behavioral deficits like depression, impaired learning and memory functions related to diabetes. This finding may be of relevance in the pathophysiology and in the clinical picture, which could be related to an altered brain serotonin metabolism and neurotransmission and may possibly be related to neuropsychiatric disorders in diabetic patients.  相似文献

BACKGROUND: The rupture of coronary atherosclerotic plaque and subsequent thrombus formation are major events underlying acute coronary syndromes (ACS). Pregnancy associated plasma protein A (PAPP-A), a biomarker of plaque rupture, has been studied in patients with ACS. This review aimed to provide an overview of clinical utility of PAPP-A in ACS patients and analytical issues adhering to immunological PAPP-A measurement. METHODS: The literature relating to PAPP-A in ACS, the molecular structure and immunodetection of PAPP-A was reviewed. PubMed was used to search the relevant articles published from 1974 to 2006. RESULTS: Higher PAPP-A concentrations have been found in patients with ACS than in patients with stable angina and subjects without coronary artery disease. Elevated PAPP-A concentrations have also been shown to associate with adverse cardiac events in ACS patients. The prognostic value of PAPP-A appears to be independent of cardiac troponins. Noteworthy, the PAPP-A form that accounts for increase in ACS is uncomplexed with the proform of eosinophil major basic protein (proMBP). However, PAPP-A assays applied in clinical studies published thus far detect total PAPP-A. Consequently, the clinical value may be non-optimal when total PAPP-A is measured in ACS patients. In addition, the clinical value can also be affected by the analytical factors that exert an effect on the performance of PAPP-A assays. CONCLUSIONS: PAPP-A appears to be a very promising biomarker useful in the clinical management of ACS patients. However, more prospective and interventional studies with carefully established immunoassays are required to validate its clinical utility.  相似文献

BACKGROUND: The renin-angiotensin system plays an important role in the pathogenesis of diabetes-induced vascular and renal complications. Vasopeptidase inhibitors simultaneously inhibit angiotensin-converting enzyme (ACE) and neutral endopeptidase. OBJECTIVE: To compare the effectiveness of vasopeptidase inhibition and ACE inhibition in preventing hypertension, endothelial dysfunction and diabetic nephropathy in spontaneously diabetic Goto-Kakizaki (GK) rats. METHODS: Eight-week-old GK rats received omapatrilat (40 mg/kg) or enalapril (30 mg/kg) for 12 weeks, either during a normal-sodium or high-sodium diet (7% w/w). Blood pressure, arterial functions and renal morphology were determined. RESULTS: Blood pressure and albuminuria were increased in GK rats compared to non-diabetic Wistar controls. Endothelium-dependent vascular relaxation in response to acetylcholine (ACh) and endothelium-independent vascular relaxation in response to sodium nitroprusside (SNP) were impaired in GK rats. Experiments with N-nitro-L-arginine methyl ester (L-NAME), diclofenac, and L-NAME + diclofenac suggested that cyclooxygenase and endothelium-derived hyperpolarizing factor components of endothelium-dependent vascular relaxation were also impaired. A high-sodium diet aggravated hypertension and diabetes-induced vascular and renal complications. Omapatrilat and enalapril normalized blood pressure and albuminuria during the normal-sodium diet, and effectively ameliorated diabetes-induced renal complications also during the high-sodium diet. However, omapatrilat improved endothelium-dependent relaxation to ACh to a greater extent (85 +/- 5%) than enalapril (68 +/- 6%, P < 0.05). Diclofenac pre-incubation eliminated this difference between omapatrilat and enalapril in ACh-induced vascular relaxation, suggesting that it was mediated, at least in part, via the cyclooxygenase pathway. CONCLUSIONS: Despite comparable blood pressure-lowering and renoprotective properties, omapatrilat may be more effective in preventing vascular dysfunction during diabetes compared to enalapril in GK rats.  相似文献

The magnetic resonance imaging (MRI) findings in psoriatic arthritis (PsA) are the same and at the same time different from those seen in other inflammatory arthritides. Synovial hypertrophy is seen on MRI in all arthritides. However, the location and extent of bone marrow edema in PsA is different from those seen in rheumatoid arthritis (RA) and osteoarthritis. Progression studies in PsA are hard to justify. However, treatment monitoring studies have given insight into the pattern of progression of the MRI findings and information regarding the mechanism of the effect of the drugs used for treatment. Threedimensional image analysis tools provide volumetric information and information regarding the spatial and temporal relationship between different MRI findings. The three-dimensional perfusion image analysis tool, which is used to evaluate the effect of antiangiogenic drugs in cancer treatment, can provide information regarding the disease mechanism when used in disease monitoring studies.  相似文献

Biological therapy for ankylosing spondylitis (AS) has led to improved disease control beyond that of conventional treatments. International recommendations encourage clinicians prescribing biological treatments to register patients in national registers to collect information on outcome and toxicity. Patients with AS (n = 229) from the Register of Biological Treatment in Finland (ROB-FIN) with severe disease of long duration were followed-up for up to 24 months. Due to an active disease, one or more concomitant disease-modifying antirheumatic drugs (DMARDs) were used by 86% at commencement of biological therapy. This add-on strategy with infliximab led to a rapid pain relief and improvement of patient's and physician's global assessments, C-reactive protein/erythrocyte sedimentation rate, and swollen and tender joint counts within 6 weeks. Concomitant use of NSAID and oral corticosteroid was reduced. Corresponding results were documented at 3 months with etanercept, which was more recently approved for the treatment of spondyloarthropathies. Seventy-nine percent of the patients were ASAS 20 responders. A subgroup of AS patients with only axial involvement (n = 46) responded correspondingly. The first biological drug was discontinued in only 7% due to lack of efficacy and in 6% due to adverse events. Anti-TNF agents, often used in combination with DMARDs, appeared to have persistent effectiveness and limited toxicity in a real-life clinical setting in a cohort of Finnish AS patients with severe disease and long disease duration.  相似文献

A new method of detecting structured changes in trabecular bone, such as those associated with osteoporosis, was evaluated on magnetic resonance images of the wrist. The method was based on gray-scale morphological granulometries which classify image texture by iteratively filtering an image and measuring the rate of change of structural diminution in a filtered-image sequence. A classification scheme capable of distinguishing structural changes in trabecular bone starting from normal trabeculae through sclerotic, cystic, and grossly porotic bone is presented. Results of the application of this technique to the evaluation of high resolution magnetic resonance images of the wrist are presented.  相似文献
